Significance DELLA proteins are plant-specific transcriptional regulators that act as signaling hubs at the interface between environment and networks control growth. The growth-promoting hormone gibberellin destabilizes DELLAs. Here we describe an alternative pathway to destabilize these proteins. We show DELLAs substrate of COP1, E3 ubiquitin ligase increases its activity promote growth in response shade or warmth. Our results not changes levels, mediates rapid destabilization environmental cues.
Ascorbate (vitamin C) is an essential antioxidant in fresh fruits and vegetables. To gain insight into the regulation of ascorbate metabolism plants, we studied mutant tomato plants (Solanum lycopersicum) that produce ascorbate-enriched fruits. The causal mutation, identified by a mapping-by-sequencing strategy, corresponded to knock-out recessive mutation class photoreceptor named PAS/LOV protein (PLP), which acts as negative regulator biosynthesis. This trait was confirmed CRISPR/Cas9 gene...
NADP(H) is a central metabolic hub providing reducing equivalents to multiple biosynthetic, regulatory and antioxidative pathways in all living organisms. While biosensors are available determine NADP+ or NADPH levels vivo, no probe exists estimate the redox status, determinant of cell energy availability. We describe herein design characterization genetically-encoded ratiometric biosensor, termed NERNST, able interact with ENADP(H). NERNST consists redox-sensitive green fluorescent protein...
The key basic helix-loop-helix (bHLH) transcription factor in iron (Fe) uptake, FER-LIKE IRON DEFICIENCY-INDUCED TRANSCRIPTION FACTOR (FIT), is controlled by multiple signaling pathways, important to adjust Fe acquisition growth and environmental constraints. FIT protein exists active inactive pools, phosphorylation of serine Ser272 the C-terminus, a regulatory domain FIT, provides trigger for activation. Here, we use phospho-mutant activity assays study phospho-mimicking phospho-dead...
Abstract The introduction of optogenetics into cell biology has furnished systems to control gene expression at the transcriptional and protein stability level, with a high degree spatial, temporal, dynamic light‐regulation capabilities. Strategies downregulate RNA currently rely on interference CRISPR/Cas‐related methods. However, these approaches lack key characteristics advantages provided by optical control. “Lockdown” introduces levels utilizing blue light‐dependent switch induce...
SUMMARY Gibberellins (GAs) are major regulators of developmental and growth processes in plants. Using the degradation‐based signaling mechanism GAs, we have built transcriptional regulator (DELLA)‐based, genetically encoded ratiometric biosensors as proxies for hormone quantification at high temporal resolution sensitivity that allow dynamic, rapid simple analysis a plant cell system, i.e. Arabidopsis protoplasts. These incorporate DELLA protein degradation target fused to firefly...
ABSTRACT Ascorbate (vitamin C) is one of the most essential antioxidants in fresh fruits and vegetables. To get insights into regulation ascorbate metabolism plants, a mutant producing ascorbate-enriched was studied. The causal mutation, identified by mapping-by-sequencing strategy, corresponded to knock-out recessive mutation new class photoreceptor named PAS/LOV protein (PLP, Solyc05g07020), which acts as negative regulator biosynthesis tomato. This trait confirmed CRISPR/Cas9 gene...
Abstract DELLA transcriptional regulators are central components in the control of plant body form response to environment. This is considered be mediated by changes metabolism hormones gibberellins (GAs), which promote degradation DELLAs. However, here we show that warm temperature or shade reduced stability a GA-insensitive allele Arabidopsis . Furthermore, induced warmth anticipated GA levels and depended on E3 ubiquitin ligase CONSTITUTIVELY PHOTOMORPHOGENIC1 (COP1). COP1 enhanced normal...
In article number 2000307, Tim Blomeier and co-workers describe the Blue Light-Operated CRISPR/Cas13b-mediated mRNA Knockdown. “Lockdown” introduces optical control of RNA levels utilizing a blue light-dependent switch to induce expression CRISPR/Cas13b, which mediates sequence-specific knockdown exo- endogenous mRNA. Further, they combined tool with other optogenetic switches for efficient downregulation target proteins in mammalian cells.
