We have developed a genetic approach to visualize axons from olfactory sensory neurons expressing given odorant receptor, as they project the bulb. Neurons specific receptor only two topographically fixed loci among 1800 glomeruli in mouse Our data provide direct support for model which topographic map of activation encodes odor quality Receptor swap experiments suggest that plays an instructive role guidance process but cannot be sole determinant establishment this map. This may more...

Animals with mutations in the leptin receptor (ObR) exhibit an obese phenotype that is indistinguishable from of deficient ob/ob mice. ObR expressed many tissues, including brain, and relative importance leptin's effects on central versus peripheral sites has not been resolved. To address this, we generated mice neuron-specific (ObR(SynI)KO) hepatocyte-specific (ObR(Alb)KO) disruption ObR. Among ObR(SynI)KO mice, extent obesity was negatively correlated level hypothalamus those animals...

Embryonic stem (ES) cells are fully pluripotent in that they can differentiate into all cell types, including gametes. We have derived 35 ES lines via nuclear transfer (ntES lines) from adult mouse somatic of inbred, hybrid, and mutant strains. ntES contributed to an extensive variety dopaminergic serotonergic neurons vitro germ vivo. Cloning by nuclei could result normal development fertile adults. These studies demonstrate the full pluripotency cells.

Cloning allows the asexual reproduction of selected individuals such that offspring have an essentially identical nuclear genome. by transfer thus far has been reported only with freshly isolated cells and from primary cultures. We previously a method cloning mice adult somatic after microinjection. Here, we apply this to clone widely available, established embryonic stem (ES) cell lines at late passage. With ES line R1, 29% reconstructed oocytes developed in vitro morula/blastocyst stage,...

All received heparin bolus 5,000 IU followed by 1,000 IU/hr. After 50 mg rt-PA given over 2 hr the severity embolism, determined from angiograms, declined 12% drug group (p less than .005) 15% .005); mean arterial pressure fell 31 +/- 7 to 22 6 mm Hg 12 21 9 respective groups....

Odorant receptors (ORs) mediate the interaction of odorous compounds with olfactory sensory neurons (OSNs) and influence guidance OSN axons to synaptic targets in bulb (OB). OSNs expressing same OR send convergent axonal projections defined glomeruli OB are thought share odorant response properties. This expectation functional similarity has not been tested experimentally, because it possible determine reproducibly properties that express ORs. Here, we applied calcium imaging characterize...

Although T cells bearing gamma delta T-cell receptors have long been known to be present in the epithelial lining of many organs, their specificity and function remain elusive. In study, we examined intestinal epithelia T-cell-receptor mutant mice, which were deficient either or alpha beta cells, normal littermates. The absence was associated with a reduction cell turnover downregulation expression major histocompatibility complex class II molecules. No such effects observed T-cell-deficient...

Olfactory sensory neurons (OSNs) expressing a given odorant receptor (OR) gene project their axons to few specific glomeruli that reside at recognizable locations in the olfactory bulb. Connecting approximately 1000 populations of OSNs 1800 mouse bulb poses formidable wiring problem. Additional progress understanding mechanisms neuronal connectivity is dependent on knowing how these axonal pathways are organized and they form during development. Here we have applied genetic approach this We...

TCR-beta gene rearrangement or expression is necessary and sufficient for the progression of early alpha beta thymocyte differentiation from CD3-CD4-CD8- triple negative (TN)3 to CD4+CD8+ double positive stage. The onset currently thought occur gradually. Some thymocytes were reported be rearranged at earliest (CD44+CD25-) TN stage, whereas other did not initiate until latest (CD44-CD25-) Here, we have isolated subsets on basis surface CD44 CD25, with c-kit as an additional marker. We...
