Abstract Hepatosplenic T-cell lymphoma (HSTL) is a rare and lethal lymphoma; the genetic drivers of this disease are unknown. Through whole-exome sequencing 68 HSTLs, we define recurrently mutated driver genes copy-number alterations in disease. Chromatin-modifying genes, including SETD2, INO80, ARID1B, were commonly HSTL, affecting 62% cases. HSTLs manifest frequent mutations STAT5B (31%), STAT3 (9%), PIK3CD for which there currently exist potential targeted therapies. In addition, noted...

ABSTRACT Liver lipid peroxidation, nonheme iron, antioxidants, and protein oxidation were investigated in experimental alcohol–induced liver disease the rat. Wistar male rats intragastrically continuously infused for 4 weeks with a high–fat diet plus an ethanol or isocaloric amount of dextrose, maintaining high blood alcohol level (200–300 mg%). This model induced fatty liver, spotty necrosis, focal inflammation. pathology was associated enhanced peroxidation decrease major antioxidant...

Significance Mutations in isocitrate dehydrogenase ( IDH ) 1 and IDH2 contribute to malignant progression by producing the oncometabolite 2HG. In myeloid disorders, mutations at three positions these genes are commonly observed, but angioimmunoblastic T-cell lymphoma (AITL), restricted arginine (R) 172. The complex microenvironment of AITL, where T cells comprise a minority tumor, has made it difficult evaluate role this mutation. Here, we provide clinical data showing that mutant expression...

Monomorphic epitheliotropic intestinal T-cell lymphoma (MEITL) is a rare aggressive most reported in Asia. We performed comprehensive clinical, pathological and genomic study of 71 European MEITL patients (36 males, 35 females, median age 67 years). The majority presented with gastrointestinal involvement had emergency surgery, 40% stage IV disease. tumors were morphologically classified into two groups: typical (58%) atypical (i.e., non-monomorphic or necrosis, angiotropism starry-sky...

Vascular endothelial growth factor (VEGF) is a potent cell and permeability highly expressed in rodent alveolar epithelium after injury repair. To investigate VEGF synthesis human lung epithelial cells, we examined expression by cultured cells under basal conditions cytokine treatment or oxidative stress. Basal was detected transformed lines (A549 1HAEo-) primary bronchial with RT-PCR, Western blot, immunocytochemistry. Among the cytokines tested, only transforming factor-beta1 increased...

Abstract Angioimmunoblastic T-cell lymphoma (AITL) is a frequent in the elderly population that has poor prognosis when treated with cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) therapy. Lenalidomide, which been safely combined CHOP to treat B-cell lymphoma, shown efficacy as single agent AITL treatment. We performed multicentric phase 2 trial combining 25 mg lenalidomide daily for 14 days per cycle 8 cycles of CHOP21 previously untreated patients aged 60 80 years. The...

We previously reported that ethanol elicits an increased protein oxidation in the liver of rats receiving chronic by continuous intragastric infusion (Tsukamoto–French method). This accumulation oxidized proteins could result from a decrease cytosolic proteolysis, related specifically to alkaline protease and its major components, proteasomes. Because several studies suggest intracellular proteolysis depends on severity oxidative stress, we investigated proteolytic activity under two...

Abstract Anaplastic large cell lymphoma (ALCL), an aggressive CD30-positive T-cell lymphoma, comprises systemic anaplastic kinase (ALK)-positive, and ALK-negative, primary cutaneous breast implant-associated ALCL. Prognosis of some ALCL subgroups is still unsatisfactory, already in second line effective treatment options are lacking. To identify genes defining state dependencies, we here characterize super-enhancer regions by genome-wide H3K27ac ChIP-seq. In addition to known key regulators,...

Extranodal NK/T-cell lymphoma (ENKTCL) is an Epstein-Barr virus (EBV)-related neoplasm with male dominance and a poor prognosis. A better understanding of the genetic alterations their functional roles in ENKTCL could help improve patient stratification treatments. In this study, we performed comprehensive analysis 178 cases to delineate landscape mutations, copy number (CNA), structural variations, identifying 34 driver genes including six previously unappreciated ones, namely, HLA-B,...

ALK-negative anaplastic large cell lymphoma (ALCL) comprises subgroups harboring rearrangements of DUSP22 (DUSP22- R) or TP63 (TP63-R). Two studies reported 90% and 40% 5-year overall survival (OS) rates in 21 12 DUSP22-R/TP63- not rearranged (NR) patients, respectively, making the prognostic impact DUSP22-R unclear.
