Nowadays, simultaneous inhibition of multiple targets through drug combination is an important anticancer strategy owing to the complex mechanism behind tumorigenesis. Recent studies have demonstrated that histone deacetylases (HDACs) will lead compensated activation a notorious cancer-related target, signal transducer and activator transcription 3 (STAT3), in breast cancer cascade, which probably limits anti-proliferation effect HDAC inhibitors solid tumors. By incorporating pharmacophore...

Urinary tract infections (UTIs) are among the most common acquired bacterial in humans. The current gold standard method for identification of uropathogens clinical laboratories is cultivation. However, culture-based assays have substantial drawbacks, including long turnaround time and limited culturability many potential pathogens. Nanopore sequencing technology can overcome these limitations detect pathogens while also providing reliable predictions drug susceptibility samples. Here, we...

The early identification of pathogens and their antibiotic resistance are essential for the management treatment patients affected by ventilator-associated pneumonia (VAP). However, microbiological culture may be time-consuming has a limited culturability many potential pathogens. In this study, we developed rapid nanopore-based metagenomic next-generation sequencing (mNGS) diagnostic assay detection VAP antimicrobial genes (ARGs).Endotracheal aspirate (ETA) samples from 63 with suspected...

Abstract Background Early availability of pathogen identification in urinary tract infections (UTIs) has critical importance disease management. Metagenomic next-generation sequencing (mNGS) the potential to transform how acute and serious are diagnosed by offering unbiased culture-free detection. However, clinical experience with application mNGS test is relatively limited. Methods We therefore established a MinION-based pathogens diagnostic platform evaluated its for implementation UTIs...

Colletotrichum dematium is an endophytic fungus recovered from a Pteromischum sp. growing in tropical forest Costa Rica. This makes novel peptide antimycotic, colutellin A, with MIC of 3.6 μg ml−1 (48 h) against Botrytis cinerea and Sclerotinia sclerotiorum. Collutellin A has mass 1127.7 Da contains residues Ile, Val, Ser, N-methyl-Val β-aminoisobutryic acid nominal molar ratios 3 : 2 1 1, respectively. Independent lines evidence suggest that the cyclic sequences Val-Ile-Ser-Ile Ile-Pro-Val...

Abstract Deep generative models have advanced drug discovery but often generate compounds with limited structural novelty, providing constrained inspiration for medicinal chemists. To address this, we develop TransPharmer, a model that integrates ligand-based interpretable pharmacophore fingerprints pre-training transformer (GPT)-based framework de novo molecule generation. TransPharmer excels in unconditioned distribution learning, generation, and scaffold elaboration under pharmacophoric...
