Defects in the assembly of dolichol-linked oligosaccharide or its transfer to proteins result severe, multi-system human diseases called Type I congenital disorders glycosylation. We have identified a novel CDG type, CDG-Ij, resulting from deficiency UDP-GlcNAc: dolichol phosphate N-acetyl-glucosamine-1 transferase (GPT) activity encoded by DPAGT1. The patient presents with severe hypotonia, medically intractable seizures, mental retardation, microcephaly, and exotropia. Metabolic labeling...

Escherichia coli DNA polymerase IV encoded by the dinB gene is involved in untargeted mutagenesis. Its human homologue κ (Polκ) DINB1 gene. Our recent studies have indicated that Polκ capable of both error-free and error-prone translesion synthesis vitro. However, it not known whether also plays a role To examine this possibility, we measured fidelity during from undamaged templates. Using kinetic measurements nucleotide incorporations assay with gapped M13mp2 DNA, show synthesizes...

Drug-resistant bacterial infections and lack of available antibacterial agents in clinical practice are becoming serious risks to public health. We synthesized a new class haloemodins by modifying traditional Chinese medicine component, emodin. The novel haloemodin exerts strong inhibitory activity on topoisomerase I DNA gyrase, not the topoisomerases human origin. In principle, it shows remarkable activities against laboratory clinically isolated Gram-positive bacteria, including...

Accumulating evidence suggested that circular RNAs (circRNAs) play critical roles in the initiation and progression of malignant cancers. However, circRNAs gastric cancer (GC) remain largely unknown. In present study, we investigated expression 5 GC tissues with metastasis without by microarray analysis. We focused on hsa_circ_0003506, which was spliced from CYFIP2 gene located at chr5:156786012–156788606 finally formed a sense-overlapping transcript 366 nt, thus named it circCYFIP2....

Deltonin is an active component purified from Dioscorea zingiberensis WRIGHT (DZW), and has shown anticancer effects. However, its mechanism of action remains elusive. In the present study, we investigated effect on a panel cancer cell lines analyzed in C26 cells, murine colon carcinoma cell. Our results showed that markedly inhibited growth all examined lines. induced dose- time-dependent apoptosis cells. The event was accompanied by release cytochrome c, depolarization mitochondrial...

Deltonin, a steroidal saponin isolated from Dioscorea zingiberensis Wright, exhibits high cytotoxic activity in cancer cells. In the present study, effects of deltonin on cell proliferation and apoptosis were evaluated MDA‑MB‑231 human breast carcinoma line. Following treatment with deltonin, viability cells was analyzed using MTT assay apoptosis, mitochondrial membrane potential (∆Ψm) alternation intracellular reactive oxygen species (ROS) generation determined by flow cytometry. addition,...

Gastric cancer (GC) is a very common type of cancer. Although current treatment modalities include surgical resection and chemotherapy, many patients are either not eligible for radical or have poor response to chemotherapy. Due the complex features disease, there need complementary therapy. In present study, effects oridonin on cell proliferation, invasion apoptosis were assessed in HGC‑27 line using Cell Counting Kit‑8 assay, real‑time analysis, an Annexin V‑FITC/propidium iodide (PI)...

Leaf extracts of the Malaysian plant Aglaia laxiflora provided two cytotoxic compounds, a new rocaglaol rhamnoside (1), known (2), (but inactive) flavonol−cinnamaminopyrrolidine adducts (3−6), and their probable biosynthetic precursors (7 trimethoxyflavonol). All structures were elucidated primarily by 2D NMR spectroscopy. The structure stereochemistry aglaxiflorin A (3) confirmed single-crystal X-ray crystallography.
