CAR-NK cells may represent a valuable tool, complementary to CAR-T cells, in adoptive immunotherapy of leukemia and solid tumors. However, gene transfer human NK is challenging task, particularly with non-virus-based techniques. Here, we describe new procedure allowing efficient electroporation-based transfection plasmid DNA, including CAR CCR7 genes, resting or cytokine-expanded cell populations NK-92 line. This offer suitable platform for safe effective use cancer.
Malignant gliomas are the most common primary brain tumors in adults and challenging cancers for diagnosis treatment. They remain a disease which non-invasive, diagnostic and/or prognostic novel biomarkers highly desirable. Altered microRNA (miRNA) profiles have been observed tumor tissues biological fluids. To date only small set of circulating/serum miRNA is found to be differentially expressed compared normal controls. Here restricted signature including miR-15b*,-23a, -99a, -125b, -133a,...
Toll-like receptors (TLRs) are pattern-recognition sensors mainly expressed in innate immune cells that directly recognize conserved pathogen structures (pathogen-associated molecular patterns-PAMPs). Natural killer (NK) have been described to express different endosomal TLRs triggered by RNA and DNA sequences derived from both viruses bacteria. This study was addressed establish which TLR could mediate NK activation function after proper stimuli. It also important the most suitable agonist...
Natural Killer (NK) cells are important components of the immune system in defense against tumor growth and metastasis. They release exosomes containing proteins nucleic acids, including microRNAs (miRNAs). NK-derived play a role anti-tumor NK cell function since they able to recognize kill cancer cells. However, involvement exosomal miRNAs is poorly understood. In this study, we explored miRNA content by microarray as compared their cellular counterparts. The expression selected lytic...
In early infected or severe coronavirus disease 2019 (COVID-19) patients, circulating NK cells are consistently reduced, despite being highly activated exhausted. The aim of this paper was to establish whether acute respiratory syndrome 2 (SARS-CoV-2) spike glycoprotein (SP) may directly trigger and through which receptor(s).
Precision in redox signaling is attained through posttranslational protein modifications such as oxidation of thiols. The peroxidase peroxiredoxin 1 (PRDX1) regulates signal transduction changes thiol its cysteines. We demonstrate here that PRDX1 a binding partner for the tumor suppressive transcription factor FOXO3 directly stress response. Heightened oxidative evokes formation disulfide-bound heterotrimers linking dimeric to monomeric FOXO3. Absence enhances nuclear localization and are...
Behçet disease (BD) is a chronic inflammatory multisystem characterized by oral and genital ulcers, uveitis, skin lesions. Disease etiopathogenesis still unclear. We aim to elucidate some aspects of BD pathogenesis identify specific gene signatures in peripheral blood cells (PBCs) patients with active using novel expression network analysis. 179 genes were modulated 10 PBCs when compared healthy donors. Among differentially expressed the top enriched function was immune response,...
We previously identified novel thiazole derivatives able to reduce histone acetylation and acetyltransferase (HAT) activity in yeast. Among these compounds, 3-methylcyclopentylidene-[4-(4'-chlorophenyl)thiazol-2-yl]hydrazone (CPTH6) has been selected used throughout this study. The effect of CPTH6 on acetylation, cell viability differentiation, cell-cycle distribution, apoptosis a panel acute myeloid leukemia solid tumor lines evaluated. Here, we showed that leads an inhibition Gcn5 pCAF HAT...
Systemic sclerosis (SSc) is a rare connective tissue disease characterized by three pathogenetic hallmarks: vasculopathy, dysregulation of the immune system, and fibrosis. A particular feature SSc increased frequency some types malignancies, namely breast, lung, hematological malignancies. Moreover, may also be paraneoplastic disease, again indicating strong link between cancer scleroderma. The reason this association still unknown; therefore, we aimed at investigating whether genetic or...
Background . Behçet’s disease (BD) is a chronic inflammatory multisystem characterized by oral and genital ulcers, uveitis, skin lesions. MicroRNAs (miRNAs) are key regulators of immune responses. Differential expression miRNAs has been reported in several autoimmune diseases; however, their role BD not fully elucidated. We aimed to identify miRNA signatures associated with investigate potential implication the pathogenesis. Methods microarray analysis was performed blood cells patients...
Background . Psoriatic arthritis (PsA) is an inflammatory arthritis, characterized by bone erosions and new formation. MicroRNAs (miRNAs) are key regulators of the immune responses. Differential expression miRNAs has been reported in several autoimmune diseases; however, their role PsA not fully elucidated. We aimed to identify miRNA signatures associated with investigate potential implication disease pathogenesis. Methods microarray was performed blood cells patients healthy controls....
Melanoma, the most lethal form of skin cancer, is frequently associated with alterations in several genes, among which Bcl‐2 oncogene plays an important role progression, chemosensitivity and angiogenesis. Also microRNA (miRNA) are emerging as modulators melanoma development them, miR‐211, located within melastatin‐1/TRPM1 (transient receptor potential cation channel, subfamily M, member 1 protein) gene, prevalently expressed melanocyte lineage acts oncosuppressor. Using human cell lines...
Psoriatic arthritis (PsA) is an inflammatory arthritis, characterised by inflammation of entheses and synovium, leading to joint erosions new bone formation. It affects 10% 30% patients with psoriasis, has estimated prevalence approximately 1%. PsA considered be primarily autoimmune disease, driven autoreactive T cells directed against autoantigens present in the skin joints. However autoinflammatory origin recently been proposed. Long noncoding RNAs (lncRNAs) are more than 200 nucleotides...
Neuroblastoma tumor-associated mesenchymal stromal cells (NB-TA-MSC) have been extensively characterized for their pro-tumorigenic properties, while immunosuppressive potential, especially against NK cells, has not thoroughly investigated. Herein, we study the immune-regulatory potential of six primary young and senescent NB-TA-MSC on cell function. Young display a phenotype (CD105+/CD90+/CD73+/CD29+/CD146+) typical MSC and, in addition, express high levels immunomodulatory molecules (MHC-I,...
Innate lymphoid cells are a heterogeneous subset of lymphocytes deeply implicated in the innate immune responses to different pathogens, organogenesis and maintenance tissue homeostasis. Group 3 (ILC3) have been detected human decidua, where they play role early inflammatory phase favoring implantation remodeling as well subsequent regulatory preventing fetal rejection supporting neoangiogenesis. A balance between inflammation tolerance is required maintain pregnancy, thus maternal system...
Neuroblastoma (NB) is the most common, extracranial childhood solid tumor arising from neural crest progenitor cells and a primary cause of death in pediatric patients. In tumors, stromal elements recruited or generated by cancer favor development an immune-suppressive microenvironment.
