Abstract The major cause of death after allogeneic Hematopoietic Stem Cell Transplantation (HSCT) for acute myeloid leukemia (AML) is disease relapse. We investigated the expression Inhibitory Receptors (IR; PD-1/CTLA-4/TIM-3/LAG-3/2B4/KLRG1/GITR) on T cells infiltrating bone marrow (BM) 32 AML patients relapsing (median 251 days) or maintaining complete remission (CR; median 1 year) HSCT. A higher proportion early-differentiated Memory (T SCM ) and Central BM-T express multiple IR in than...
Chimeric antigen receptor (CAR) T cell expansion and persistence represent key factors to achieve complete responses prevent relapses. These features are typical of early memory cells, which can be highly enriched through optimized manufacturing protocols. Here, we investigated the efficacy safety profiles CAR products generated from preselected naive/stem cells (TN/SCM), as compared with unselected (TBULK). Notwithstanding their reduced effector signature in vitro, limiting TN/SCM doses...
Immunotherapy with chimeric antigen receptor (CAR)–engineered T cells showed exceptional successes in patients refractory B cell malignancies. However, first-in-human studies solid tumors revealed unique hurdles contributing to poor demonstration of efficacy. Understanding the determinants tumor recognition by CAR should translate into design strategies that can overcome resistance. Here, we show multiple carcinomas express extracellular N-glycans, whose abundance negatively correlates...
T cell receptor (TCR)–based therapy has the potential to induce durable clinical responses in patients with cancer by targeting intracellular tumor antigens high sensitivity and promoting survival. However, need for TCRs specific shared oncogenic manufacturing protocols able redirect specificity while preserving fitness remain limiting factors. By longitudinal monitoring of functionality dynamics 15 healthy donors, we isolated 19 Wilms’ antigen 1 (WT1), which is overexpressed several types....
Colorectal tumours are often densely infiltrated by immune cells that have a role in surveillance and modulation of tumour progression but burdened immunosuppressive signals, which might vary from primary to metastatic stages. Here, we deployed multidimensional approach unravel the T-cell functional landscape colorectal cancers (CRC) liver metastases, genome editing tools develop CRC-specific engineered T cells. We paired high-dimensional flow cytometry, RNA sequencing immunohistochemistry...
Discrimination between hematopoietic stem cells and leukemic remains a major challenge for acute myeloid leukemia immunotherapy. CAR T specific the CD117 antigen can deplete malignant healthy before consolidation with allogeneic cell transplantation in absence of cytotoxic conditioning. Here we exploit non-viral technology to achieve early termination activity prevent incoming graft rejection. Transient expression an anti-CD117 by mRNA conferred ability eliminate CD117+ targets vitro vivo....
SARS-CoV-2 pandemic still poses a significant burden on global health and economy, especially for symptoms persisting beyond the acute disease. COVID-19 manifests with various degrees of severity identification early biomarkers capable stratifying patient based risk progression could allow tailored treatments.
Abstract Background In the scientific literature, many studies describe application of lean methodology in hospital setting. Most articles focus on results rather than approach adopted to introduce methodology. absence a clear view context and introduction strategy, first steps implementation process can take an empirical, trial error profile. Such is time-consuming resource-intensive affects adoption model at organizational level. This research aims outline role contextual factors strategy...
CAR T-cell products targeting lineage-specific cell-of-origin antigens, thereby eliminating both tumor and healthy counterpart cells, are currently clinically approved therapeutics in B- plasma-cell malignancies. While they represent a major clinical improvement, still limited terms of efficacy by e.g. single, sometimes low-expressed antigen targeting, safety e.g., lack on-off activity. Successful non-discriminative heterogeneous hematopoietic stem progenitor cell malignancies, such as acute...
Purpose The aim of this paper is to understand how resilience builds achieve a management model for sustainable resilience, as advocated by development goals (SDGs), in distressed communities. topic addressed with the case Macerata, an Italian city located at epicentre devastating earthquake 1997 and later, short time interval between August 2016 January 2017. Necessary knowledge on modes places engagement collaboration delivered attempt demonstrate that social cultural factors have stronger...
We adapted a grid into microswitch for the hand movements of girl with profound multiple disabilities who lay on her back. The grid, suspended above girl's face, was equipped two mercury devices, i.e., small sealed ampoules containing drop and ending conductive leads. A lateral or forward movement would make at least one device slide to leads activate device. During intervention, activations activations, produced occurrence favorite stimuli. Analysis showed that increased frequency during...
In adoptive T cell therapy, the long term therapeutic benefits in patients treated with engineered tumor specific cells are limited by lack of persistence infused cellular products and immunosuppressive mechanisms active microenvironment. Exhausted infiltrating characterized loss effector functions triggered multiple inhibitory receptors (IRs). patients, IR blockade reverts exhaustion but has low selectivity, potentially unleashing autoreactive clones resulting clinical autoimmune side...
The phenotype of infused cells is a major determinant Adoptive T-cell therapy (ACT) efficacy. Yet, the difficulty in deciphering multiparametric cytometry data limited fine characterization cellular products. To allow analysis dynamic and complex flow samples, we developed cytoChain, novel dataset mining tool new analytical workflow. CytoChain was challenged to compare state-of-the-art innovative culture conditions generate stem-like memory (TSCM ) suitable for ACT. Noticeably, combination...
This study assessed whether a microswitch cluster with favorite stimulation could help 9.1-yr.-old child multiple disabilities increase arm-lifting responses without dystonic, sideways head tilting. The involved two microswitches, one at the child's arm to monitor lifting response, and other it was tilted during lifting. Analysis showed that percentage of occurring tilting increased most visibly when focused on those (appropriate) responses.
Abstract The new concept of social responsibility is strongly linked to the idea enterprise as a living system, that is, system capable regenerating itself by producing “knowledge” and “trust” resources. Nevertheless, these resources can only spring from tight relationship with reference environment in which operates. This paper aims at making some first considerations on between environment, upon how may develop symbiosis “living company.”
Abstract Social innovation represents a social practice that aims to meet needs by involving communities in dialogues through participatory and collaborative approaches. One new particular solution of is represented Urban Civic Networks. However, considering the novelty this topic, neither benefits nor role public entities implementing such innovation, strengths implementation process are known, yet. This study fill gap analysis single‐case study, based upon theories change, referring City...
Background: Toe walking is associated with autism spectrum disorders (ASD). Correction of this “behavior” a health challenge. The toe walker affected by the contact refusal outside world: touching ground as little possible, trying to avoid any contact. A structured equines foot possible consequence. Method: We present “Cast and Go” protocol, used in 22 idiopathic children ASD treated from 2015 2020. treatment was performed single senior experienced doctor botulinum injection, ankle casts...
We describe a multi-step high-dimensional (HD) flow cytometry workflow for the deep phenotypic characterization of T cells infiltrating metastatic tumor lesions in liver, particularly derived from colorectal cancer (CRC-LM). First, we applied novel cytometer setting approach based on single positive rather than fluorescent beads, resulting optimal sensitivity when compared with previously published protocols. Second, set up 26-color antibody panel designed to assess functional state both...
