Abstract The major cause of death after allogeneic Hematopoietic Stem Cell Transplantation (HSCT) for acute myeloid leukemia (AML) is disease relapse. We investigated the expression Inhibitory Receptors (IR; PD-1/CTLA-4/TIM-3/LAG-3/2B4/KLRG1/GITR) on T cells infiltrating bone marrow (BM) 32 AML patients relapsing (median 251 days) or maintaining complete remission (CR; median 1 year) HSCT. A higher proportion early-differentiated Memory (T SCM ) and Central BM-T express multiple IR in than...

Chimeric antigen receptor (CAR) T cell expansion and persistence represent key factors to achieve complete responses prevent relapses. These features are typical of early memory cells, which can be highly enriched through optimized manufacturing protocols. Here, we investigated the efficacy safety profiles CAR products generated from preselected naive/stem cells (TN/SCM), as compared with unselected (TBULK). Notwithstanding their reduced effector signature in vitro, limiting TN/SCM doses...

Immunotherapy with chimeric antigen receptor (CAR)–engineered T cells showed exceptional successes in patients refractory B cell malignancies. However, first-in-human studies solid tumors revealed unique hurdles contributing to poor demonstration of efficacy. Understanding the determinants tumor recognition by CAR should translate into design strategies that can overcome resistance. Here, we show multiple carcinomas express extracellular N-glycans, whose abundance negatively correlates...

T cell receptor (TCR)–based therapy has the potential to induce durable clinical responses in patients with cancer by targeting intracellular tumor antigens high sensitivity and promoting survival. However, need for TCRs specific shared oncogenic manufacturing protocols able redirect specificity while preserving fitness remain limiting factors. By longitudinal monitoring of functionality dynamics 15 healthy donors, we isolated 19 Wilms’ antigen 1 (WT1), which is overexpressed several types....

Colorectal tumours are often densely infiltrated by immune cells that have a role in surveillance and modulation of tumour progression but burdened immunosuppressive signals, which might vary from primary to metastatic stages. Here, we deployed multidimensional approach unravel the T-cell functional landscape colorectal cancers (CRC) liver metastases, genome editing tools develop CRC-specific engineered T cells. We paired high-dimensional flow cytometry, RNA sequencing immunohistochemistry...

Discrimination between hematopoietic stem cells and leukemic remains a major challenge for acute myeloid leukemia immunotherapy. CAR T specific the CD117 antigen can deplete malignant healthy before consolidation with allogeneic cell transplantation in absence of cytotoxic conditioning. Here we exploit non-viral technology to achieve early termination activity prevent incoming graft rejection. Transient expression an anti-CD117 by mRNA conferred ability eliminate CD117+ targets vitro vivo....

CAR T-cell products targeting lineage-specific cell-of-origin antigens, thereby eliminating both tumor and healthy counterpart cells, are currently clinically approved therapeutics in B- plasma-cell malignancies. While they represent a major clinical improvement, still limited terms of efficacy by e.g. single, sometimes low-expressed antigen targeting, safety e.g., lack on-off activity. Successful non-discriminative heterogeneous hematopoietic stem progenitor cell malignancies, such as acute...

This study assessed whether a microswitch cluster with favorite stimulation could help 9.1-yr.-old child multiple disabilities increase arm-lifting responses without dystonic, sideways head tilting. The involved two microswitches, one at the child's arm to monitor lifting response, and other it was tilted during lifting. Analysis showed that percentage of occurring tilting increased most visibly when focused on those (appropriate) responses.

Nevertheless, these resources can only spring from tight relationship with reference environment in which operates.
