Abstract Precise coordination of cell growth, proliferation and differentiation is essential for the development multicellular organisms. Here, we report that although mechanistic target rapamycin complex 1 (mTORC1) activity required chondrocyte growth proliferation, its inactivation differentiation. Hyperactivation mTORC1 via TSC1 gene deletion in chondrocytes causes uncoupling normal programme within plate, resulting uncontrolled blockage chondrodysplasia mice. Rapamycin promotes restores...

This study aims to identify circulating proteins that are causally associated with osteoarthritis (OA)-related traits through Mendelian randomisation (MR)-based analytical framework.Large-scale two-sample MR was employed estimate the effects of thousands plasma on 12 OA-related traits. Additional analyses including Bayesian colocalisation, Steiger filtering analysis, assessment protein-altering variants and mapping expression quantitative trait loci protein were performed investigate...

Autophagy maintains cellular homeostasis by stimulating the lysosomal degradation of cytoplasmic structures, including damaged organelles and dysfunctional proteins. The role autophagy in renewal regeneration injured peripheral nerves remains poorly understood. current study investigated nerve motor function recovery following sciatic crush injury rats or suppressing detecting presence autophagosomes LC3-II expression electron microscopy Western blotting, respectively. Abstract Monascus pilosus has been used to produce lipid-lowering drugs rich in monacolin K (MK) for a long period. Genome mining reveals there are still many potential genes worth be explored this fungus. Thereby, efficient genetic manipulation tools will greatly accelerate progress. In study, we firstly developed the protocol prepare protoplasts recipient of CRISPR/Cas9 system. Subsequently, vector and donor DNA were co-transformed into recipients (10 6 protoplasts/mL) 60–80 transformants...

Accurately predicting knee osteoarthritis (KOA) is essential for early detection and personalized treatment. We aimed to develop test a magnetic resonance imaging (MRI)-based joint space (JS) radiomic model (RM) predict radiographic KOA incidence through neural networks by integrating meniscus femorotibial cartilage features.

Fungal contamination poses a threat to agriculture, food production and human health, safe effective method limit fungal is still needed. In this study, the inhibitory potency of heptanal, natural volatile chemical, against Aspergillus flavus growth was evaluated, its mechanism action explored. The minimum concentrations (MICs) heptanal vapor fumigation liquid contact culture were 0.085 0.8 μL/mL, respectively. A total 79 significant differentially abundant metabolites, including 37...

Abstract: Osteoporosis is a systemic skeletal disease that characterized by low bone density and microarchitectural deterioration of tissue. The increasing prevalence osteoporosis has attracted much attention. In this study, MC3T3-E1 pre-osteoblasts were treated with the natural compound, baicalein (0.1 µmol/L, 1 10 µmol/L), to stimulate differentiation over 14-day period. addition, canonical ovariectomized (OVX) mouse model was used investigate effect 3-month treatment (10 mg/kg per day) in...

The genus Neobythites is the most diverse group in family Ophidiidae. In this study, we assembled and reported complete mitochondrial genome of N. sivicola for first time. 17,316 bp length contains 13 PCGs, 2 rRNAs, 22 tRNAs. Phylogenetic analysis revealed that was sister species to unimaculatus, closer Sirembo. This study significantly contributes genomic resources available Ophidiidae Neobythites, providing a valuable molecular resource further research on taxonomy phylogenetics family.

Although the reduced muscle quality is common in elderly people, its role development of knee osteoarthritis (OA) remains unclear. Magnetic Resonance Imaging (MRI) considered gold standard imaging modality to assess quality, which might holistically capture underlying pathology. This study aimed apply MRI-based radiomics thigh muscles comprehensively, and examine prediction performance for incident OA. The participants at risk OA during 48-month visit were selected from a multicenter cohort...
