Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused the unprecedented disease 2019 (COVID-19) pandemic. Critical cases of COVID-19 are characterized by production excessive amounts cytokines and extensive lung damage, which is partially fusion SARS-CoV-2-infected pneumocytes. Here, we found that cell SARS-CoV-2 spike (S) protein induced a type I interferon (IFN) response. This function S required its cleavage proteases at S1/S2 S2' sites. We further showed damaged nuclei...
Mpox is caused by a zoonotic virus belonging to the Orthopoxvirus genus and Poxviridae family. In this study, we develop recombinase polymerase amplification (RPA)-coupled CRISPR-Cas12a detection assay for mpox virus. We design test series of CRISPR-derived RNAs(crRNAs) targeting conserved D6R E9L genes orthopoxvirus unique N3R N4R viruses. crRNA-1 exhibits most robust activity in detecting orthopoxviruses, crRNA-2 able distinguish from other orthopoxviruses. The Cas12a/crRNA alone presents limit 10
SARS-CoV-2 has become a global threat to public health. Infected individuals can be asymptomatic or develop mild severe symptoms, including pneumonia, respiratory distress, and death. This wide spectrum of clinical presentations infection is believed in part due the polymorphisms key genetic factors population. In this study, we report that interferon-induced antiviral factor IFITM3 inhibits by preventing spike-protein-mediated virus entry cell-to-cell fusion. Analysis Chinese COVID-19...
Since May 2022, the multi-country outbreak of monkeypox (mpox) has raised a great concern worldwide. Early detection mpox virus infection is recognized as an efficient way to prevent transmission. Mpox specific methods reported up now are based on SNPs among and other orthopoxviruses. We have therefore developed real-time PCR method targeting sequences (N3R B18Rplus). also optimized orthopoxvirus system which targets highly conserved E9L D6R genes. The assays high sensitivity (1...
Abstract The mpox outbreak has subdued with fewer reported cases at the present in high‐income countries. It is known that virus (MPXV) infection been epidemic for more than 50 years African ancestral MPXV strain changed into multiple clades, indicating ongoing evolution of MPXV, which reflects historical neglect Africa, especially after smallpox eradication, and bestows danger severe epidemics future. thus imperative to continue development diagnostics treatments so we can be prepared event...
Purpose: This study aimed to investigate the prognostic significance of platelet-to-platelet distribution width ratio (P/PDW), systemic inflammation response index (SIRI), and immune (SII) in patients with locally advanced nasopharyngeal carcinoma (LA-NPC). Methods: A total 549 LA-NPC were included this retrospective analysis. Clinicopathological characteristics blood test data obtained from patient records. Receiver operating characteristic (ROC) curve analysis was employed determine...
Abstract Cancer cells harbor cell-intrinsic programs capable of sculpting immunogenic landscapes, yet molecular brakes that enforce immune evasion remain poorly defined. Here, we identify C9ORF50, a previously uncharacterized intrinsically disordered protein, as liquid-liquid phase separation-driven regulator RNA processing and actionable immunotherapy target. Genetic ablation C9ORF50 induces selective intron retention in spliceosome component transcripts, leading to cytoplasmic accumulation...
SERINC5 is a multi-span transmembrane protein that incorporated into HIV-1 particles in producing cells and inhibits entry. Multiple retroviruses like HIV-1, equine infectious anemia virus murine leukemia are subject to inhibition, while pseudotyped with envelope glycoproteins of vesicular stomatitis Ebola resistant SERINC5. The antiviral spectrum the underlying mechanisms restriction not completely understood. Here we show influenza A infection by targeting virus-cell membrane fusion at an...
We report on pulse profile decomposition analysis of a bright transient X-ray pulsar 1A 0535+262 using the broadband Insight-HXMT observations during giant outburst source in 2020. show that observed shape can be described terms combination two symmetric single-pole contributions for wide range energies and luminosities fixed geometry defining basic pulsar. This corresponds to slightly distorted dipole magnetic field, i.e., one pole has offset by $\sim 12^{\circ}$ from antipodal position...
Abstract There is now increasing recognition of the important role androgen receptor (AR) in modulating immune function. To gain a comprehensive understanding effects AR activity on cancer immunity, we employed computational approach to profile 33 human tumor types using RNA-Seq datasets from The Cancer Genome Atlas. Our pan-cancer analysis revealed that genes most negatively correlated with across cancers are involved active system processes. Importantly, observed significant negative...
Long interspersed element type 1 (LINE-1, also L1 for short) is the only autonomously transposable in human genome. Its insertion into a new genomic site may disrupt function of genes, potentially causing genetic diseases. Cells have thus evolved battery mechanisms to tightly control LINE-1 activity. Here, we report that cellular antiviral protein, myxovirus resistance protein B (MxB), restricts mobilization LINE-1. This MxB requires nuclear localization signal located at its N-terminus,...
As a vital tumor suppressor, PTEN (Phosphatase and tension homolog deleted on chromosome 10) is involved in inherited syndromes, among the most frequently inactivated suppressor gene sporadic cancers. loss-of-function widely occurs human cancers via variety of mechanisms, including genetic alterations posttranslational modification. These suggest has role functional importance In present study, we constructed prokaryotic expression vector that efficiently expresses GST-PTEN (the target...
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccines provide essential tools for the control of COVID-19 pandemic. A number technologies have been employed to develop SARS-CoV-2 vaccines, including inactivated particles, mRNA express viral spike protein, recombinant proteins, and vectors. Here, we report use vaccinia virus Tiantan strain as a vector protein. When it was used inoculate mice, robust protein-specific antibody response T-cell were detected. Sera from vaccinated...
Immune-based therapies induce durable remissions in subsets of patients across multiple malignancies. However, there is limited efficacy immunotherapy metastatic castrate-resistant prostate cancer (mCRPC), manifested by an enrichment immunosuppressive (M2) tumor- associated macrophages (TAM) the tumor immune microenvironment (TME). Therefore, therapeutic strategies to overcome TAM-mediated immunosuppression are critically needed mCRPC. Here we discovered that NLR family pyrin domain...
