Hepatocellular carcinoma (HCC) is an immunotherapy-resistant malignancy characterized by high cellular heterogeneity. The diversity of cell types and the interplay between tumor non-tumor cells remain to be clarified. Single RNA sequencing human mouse HCC tumors revealed heterogeneity cancer-associated fibroblast (CAF). Cross-species analysis determined prominent CD36+ CAFs exhibited high-level lipid metabolism expression macrophage migration inhibitory factor (MIF). Lineage-tracing assays...
Background and Aims: Monocarboxylate transporter (MCT) 4 is a high‐affinity lactate that primarily involved in the maintenance of intracellular pH homeostasis highly expressed different tumors. However, role MCT4 modulating immune responses against HCC remains unknown. Approach Results: In this study, we demonstrated was overexpressed HCC, which associated with poor prognosis patients. Genetic or pharmacological inhibition using VB124 (a potent inhibitor) suppressed tumor growth...
Abstract Serine arginine-rich splicing factor 1 (SRSF1) is a key oncogenic in various cancers, promoting abnormal gene expression through post-translational regulation. Although the protumoral function of SRSF1 well-established, effects inhibiting tumor-intrinsic on tumor microenvironment and its impact CD8 + T cell-mediated antitumor immunity remain unclear. Our findings indicate that depleting cells improve immune function, glycolytic metabolism, efficacy adoptive cell therapy. The...
Abstract Background Combination conversion therapies afforded curative surgery chance for initially unresectable hepatocellular carcinoma (uHCC). This study aimed to evaluate the rate and clinical outcomes of a first-line regimen lenvatinib combined with transarterial chemoembolization (TACE) plus immunotherapy initial uHCC by interpreting real-world data. Methods Conversion therapy data patients from November 2018 January 2021 were analysed. The regimens included triple combination (t-CT:...
Tumor recurrence after liver transplantation (LT) impedes the curative chance for hepatocellular carcinoma (HCC) patients. This study aimed to develop a deep pathomics score (DPS) predicting tumor using learning. Two datasets of 380 HCC patients who underwent LT were enrolled. Residual convolutional neural networks used identify six histological structures HCC. The individual risk each structure and DPS derived by modified DeepSurv network. Cox regression analysis Concordance index evaluate...
The activation of nanoparticles (NPs) in the tumor microenvironment exerts synergistic therapeutic effects with chemotherapy against multiple cancers. In this study, an NP system prepared using biocompatible MIL-100 NPs was studied as effective vehicle to deliver oxaliplatin for hepatocellular carcinoma treatment. were coated polydopamine (PDA) and NH2-PEGTK-COOH then loaded create multi-functional Oxa@MIL-PDA-PEGTK. Oxa@MIL-PDA-PEGTK is activated microenvironment, causing generation...
Background Hepatocellular carcinoma (HCC) is characterized by inflammation and immunopathogenesis. Accumulating evidence has shown that the cystathionine β-synthase/hydrogen sulfide (CBS/H 2 S) axis involved in regulation of inflammation. However, roles CBS HCC development immune evasion have not been systematically investigated, their underlying mechanisms remain elusive. Here, we investigated tumor cells microenvironment HCC. Methods 236 samples were collected to detect expression CBS,...
Abstract Primary liver cancer (PLC) poses a leading threat to human health, and its treatment options are limited. Meanwhile, the investigation of homogeneity heterogeneity among PLCs remains challenging. Here, using single-cell RNA sequencing, spatial transcriptomic bulk multi-omics, we elaborated molecular architecture 3 PLC types, namely hepatocellular carcinoma (HCC), intrahepatic cholangiocarcinoma (ICC) combined hepatocellular-cholangiocarcinoma (CHC). Taking high-resolution...
We studied the value of circulating tumor DNA (ctDNA) in predicting early postoperative recurrence and monitoring burden patients with hepatocellular carcinoma (HCC). Plasma-free DNA, germline tissue were isolated from 41 HCC. Serial ctDNAs analyzed by next-generation sequencing before after operation. Whole-exome was used to detect HCC adjacent tissues. In total, 47 gene mutations identified ctDNA surgery. detected 63.4% 46% patient plasma pre- postoperation, respectively. The preoperative...
T cell exhaustion is a critical obstacle for durable treatment response in hepatocellular carcinoma (HCC). Developing drugs that control tumor growth and simultaneously bolster immune function of great significance. Although high-mobility group box 2 (HMGB2) has been reported to be crucial HCC prognosis, its role the microenvironment remains unclear. Here, we found HMGB2 + CD8 cells as being associated with resistance anti–PD-1 through single-cell RNA sequencing. Mechanistically, impaired...
Abstract Background Intrahepatic cholangiocarcinoma (iCCA) is a highly heterogeneous and lethal hepatobiliary tumor with few therapeutic strategies. The metabolic reprogramming of cells plays an essential role in the development tumors, while molecular classification iCCA largely unknown. Here, we performed integrated multiomics analysis to depict differences characteristics patients, hoping provide novel perspective understand treat iCCA. Methods We 116 samples, including whole‐exome...
Abstract Hepatocellular adenoma (HCA) represents a rare benign hepatic neoplasm with potential for malignant transformation into hepatocellular carcinoma (HCC), yet the underlying mechanism remains elusive. In this study, we investigated genomic landscape of process to identify therapeutic strategies blocking transformation. Using micro‐detection techniques, obtained specimens adenoma, cancerous and adjacent normal liver from three patients undergoing resection surgery. Whole‐exome...
Lung adenocarcinoma (LUAD) is a malignant tumor of the respiratory system with poor prognosis. Recent studies have revealed that N7-methylguanosine (m7G) methylation widespread modification occurring in RNA. But expression m7G methylation-related genes LUAD and their correlations prognosis are still unclear. In this study, we found 12 regulators differential between normal lung tissues. According to differentially expressed (DEGs), all cases were separated into two subtypes. The prognostic...
Determining the physical locations of CDN Points Presence (PoPs) is fundamental to understanding and diagnosing services. Yet, popular deployment IP Anycast in CDNs has rendered existing geolocation tools unreliable. To fill this gap, we present an HTTP-based solution that leverages subtle geographic hints HTTP responses locate PoPs at city-level granularity. The evaluation shows our technique achieves over 90% accuracy with average error distance less than 40 km.
GPX8, which is found in the endoplasmic reticulum lumen, a member of Glutathione Peroxidases (GPXs) family. Its role hepatocellular carcinoma (HCC) unknown.
Stack Overflow is widely recognized by software practitioners as the go-to resource for addressing technical issues and sharing practical solutions. While not typically seen a scholarly forum, users on commonly refer to academic sources in their discussions. Yet, little known about these referenced works how they intersect needs interests of community. To bridge this gap, we conducted an exploratory large-scale study landscape references Overflow. Our findings reveal that communities with...
Hepatocellular carcinoma (HCC) is an immunotherapy-resistant malignancy characterized by high cellular heterogeneity. The diversity of cell types and the interplay between tumor non-tumor cells remain to be clarified. Single RNA sequencing ~75000 from 7 HBV-related HCC tissues revealed 5 distinct cancer-associated fibroblast (CAF) subsets. prominent CD36+ CAFs exhibited high-level lipid metabolism expression macrophage migration inhibitory factor (MIF). Furthermore, CD36 mediated oxidated...
