BACKGROUND In cervical lesions, the overexpression of p16 is reported to be closely associated with high-risk human papillomavirus (HPV) infection. The objective current study was confirm usefulness liquid-based specimens for staining as well tissue sections. METHODS A total 98 patients lesions were entered into study. After cytologic examination using smears, same slides immunostained and compared simultaneously obtained, immunohistologically stained Moreover, status HPV infection examined...
HPV L1 capsid protein is expressed together with the production of infectious viral particles, but its expression and relation to p16 expression, which has been a surrogate marker for human papilloma virus (HPV) infection in cervix, are little studied cytology samples. The authors aimed elucidate between expressions liquid-based samples from uterine cervical lesions.Immunochemical analyses using antibodies against were carried out on cytological materials obtained lesions low-grade squamous...
Abstract Alterations of the Akt/mTOR pathway have been observed in numerous types cancer, thus this represents an exciting new target for molecular therapeutics. We investigated expression activated Akt (p‐Akt) and mTOR (p‐mTOR) patients with adenocarcinoma cervix involvement p‐Akt/p‐mTOR response to combination inhibitor agents, rapamycin LY294002, conventional therapy, cisplatin, vitro. Immunohistochemistry analysis p‐Akt p‐mTOR was conducted 26 cervix. Western blot performed determine...
To assess the exact response of malignant ascites to treatment, objective measurement volume is essential. We have developed a simple method measure by using standard abdomino-pelvic computed tomography (CT). The aim study validate accuracy measurements comparing them with calculation 3D-CT.Twelve consecutive patients cancer who had measurable underwent 15 helical CT examinations. On conventional images, thickness in centimeters was measured three planes such as bilateral subphrenic space (A...
Objective: This study evaluated the usefulness of p16INK4a/Ki-67 as a new biomarker in diagnosis human papillomavirus (HPV)-related cervical lesions. Study Design: From 69 women with previous positive cytology, clinician-collected (CC) samples were obtained using Cervex-Brush®. One month later, self-collected (SC) material was acquired Rovers® Viba-Brush. Liquid-based cytology specimens prepared from both samples, and then grades...
Human papillomavirus (HPV) 16 DNA can be integrated into the of cells, thereby disrupting E2 gene expression, which leads to increased expression E6 and E7 viral oncogenes progression cancer. However, relationships among HPV load, cytologic diagnosis, integration status remain unclear. The aim this study was evaluate type distribution, status, then investigate their with precancerous cancerous lesions Japanese women different age groups. Liquid-based cytology (LBC) samples were examined by...
Clear cell adenocarcinoma of the ovary has a poor prognosis due to chemoresistance and early metastasis lymph nodes. It also can result in endometriosis is second most frequent type ovarian cancer Japan. Serous another common epithelial tissue subtype Japan, it highly sensitive chemotherapy. In current study, we examined differential expression genes these types tried analyze their functions, especially as they relate chemoresistance. We used display compare clear carcinoma serous ovary....
A phase I study was performed to determine the recommended dose of carbon ion radiotherapy and 3D image-guided brachytherapy for histologically confirmed stage II (≥4 cm), III, or IVA cervical cancer. Dose-limiting toxicities (treatment-related occurring within three months from start radiotherapy) included Grade 3 non-hematological toxicity, 4 hematological interruption treatment more than two weeks due treatment-related toxicities. Carbon consisted whole-pelvic irradiation with 36.0 Gy...
We assessed the relative expression of oestrogen receptor (ER)α and (ER)β mRNAs in 36 human endometrial cancers using a multiplex polymerase chain reaction (PCR). To determine whether or not ER subtypes is associated with clinicopathological parameters, we examined correlations between age, tumour grade depth myometrial invasion. Using multiple regression analysis, invasion showed significant correlation ER-β: ER-α ratio (r = 0.54, P 0.0007). The ER-β:ER-α was high advanced invasive...
The Smad3 gene is a member of the Smad family, vertebrate homologues Drosophila Mad, and its product cytoplasmic element in transforming growth factor-beta (TGF-beta) signaling pathway. Mutations TGF-beta receptors their elements transduction signals commonly accompany various cancers. Using PCR-SSCP analysis we searched for presence mutations 36 human ovarian cancers, found that 15 cases (41. 7%) had polymorphism at codon 103. Because this mutation was not accompanied by amino acid...
In Brief BACKGROUND: Compartment syndrome is a potentially devastating complication with possible permanent neuromuscular and kidney damage. CASE: A woman who had undergone radical hysterectomy pelvic paraaortic lymphadenectomy was diagnosed compartment of the lower left limb. Thrombosis common iliac artery also found after emergency fasciotomy. CONCLUSION: Arterial thrombosis less than deep vein during gynecologic operations, but lithotomy position may cause insufficient arterial...
Objective: This study compared the usefulness of self-collected (SC) and clinician-collected (CC) materials for cervical cytology human papillomavirus (HPV) genotyping. Study Design: Fifty women with previous positive who were undergoing regular checkups included in study. CC samples collected using a Cervex-Brush Combi liquid-based cytology. One month later, SC material was acquired Rovers Viba-brush vaginal sampler, fixed at home. Thin-layer specimens...
The p16INK4A gene, which encodes the cell-cycle regulatory protein cyclin-dependent kinase 4 inhibitor, is a putative tumor-suppressor gene. We examined p16 gene alterations in 30 primary ovarian cancers and 11 cancer cell lines. Five of (16.7%) had lost both genes. In addition, four (13.3%) contained five kinds missense mutations one-base deletion. Three lines homozygous deletions genes, one line multiple intragenic mutations. There was also suppressed transcription line. Some point...
Abstract Background Recent phase III randomized trials have suggested that neoadjuvant chemotherapy followed by interval debulking surgery (NACT-IDS) is a treatment option for patients with advanced epithelial ovarian cancer. This study aimed to use CA-125 and computed tomography (CT) scanning generate simple clinically applicable model of predicting complete cytoreduction (IDS) the overall survival in who receive taxane/platinum-based as (NACT). Methods Patients stage IIIc or IV cancer...
The Smad6 and Smad7 genes are members of the Smad family, involved in transforming growth factor-beta (TGF-beta) signaling pathway. Mutations TGF-beta receptors their cytoplasmic elements transduction signals commonly accompany various cancers. Using PCR-SSCP analysis we searched for presence gene mutations 30 human ovarian cancers 4 cancer cell lines, found that 12 cases (35.3%) had a polymorphism intron 2 8 (23.5%) at codon 208 gene. Because these polymorphisms were not accompanied by...
8-oxo-deoxyguanosine triphosphate (8-oxo-dGTP) is a major oxidation product in the nucleotide pool of cell and potent mutagen, because it can be incorporated into DNA with equal frequency opposite either template C or A. The human MTH1 gene (hMTH1) homologue E. coli mutator mutT, which encodes 8-oxo-dGTPase. hMTH1 protein reduces spontaneous mutations by removing 8-oxo-dGTP from pool. To determine whether this associated carcinogenesis ovarian cancer, present study examined, for first time,...
Abstract Background In Japan, 8000 women were newly diagnosed with cervical cancer in 2018. The healthcare insurance policy Japan allows physicians to utilize vaginal volt cytology tests and serum biomarker measurement at every visit imaging analysis an adequate interval screening for recurrence after initial treatment. However, the major surveillance guidelines published United States European countries recommend focusing on pelvic examinations symptom reviews avoid unnecessary tests. This...
