The present work describes the isolation and purification of two Leishmania chagasi (= syn. infantum) recombinant proteins, rLci2B rLci1A, their use in development an immunoassay for diagnostic canine leishmaniasis. After protein expression cell disruption, was purified by immobilized metal affinity chromatography followed size exclusion chromatography, whereas expressed as inclusion body, treated with urea anion-exchange chromatography. Homogeneities were ascertained denaturing gel...
In Brazil, polysaccharide–protein conjugate vaccine against Neisseria meningitidis group C (MenCPS-TT) using hydrazine-activated-tetanus toxoid (TT) as a carrier protein has been developed. Because of the toxicity hydrazine in humans, it is necessary to monitor this substance's process control step during production. The electroanalytical methodology was developed and validated for determination MenCPS-TT production by differential pulse polarography. reduction potential −0.95 V acetone...
Introduction: Infections caused by Enterococcus spp.are a serious public health problem. They are difficult to treat and have high morbidity.Therapeutic monoclonal antibodies (mAbs) been used diseases in general. In the absence of new antibiotics for treatment multidrug-resistant bacteria, immunotherapy using is promising alternative. Objectives: Characterize binding mAb against PBP2a MRSA PBP5 spp.Methodology: An E. coli BL21 strain, previously transformed with vector containing gene sequence...
Introduction:The role of nucleocapsid (N) protein from SARS-Cov-2 is not only to compose viral particle, but also contributes many critical activities after virus invasion.Functional diversity intimately associated dynamic structure and its ability bind change RNA structure.Knowledge properties on native fold allows several applications drug design the optimization activity. Objectives: This study investigated recombinant SARS-CoV-2 N thermal nucleic acid interaction.Methodology: Benzonase...
Introduction: Leishmaniasis stands out globally among parasitic diseases and poses a significant challenge to public health. The remarkable adaptive potential of these protozoa is evident in their protein abilities, particularly proteases, which emerge as central figures the parasites virulence factors. Objectives: To assess recombinant Oligopeptidase B (OPB) Cathepsin (CPB) from Leishmania (L.) amazonensis proteolytic process biochemical structural characteristics.Methodology: Plasmids were...
Introduction: The diagnosis of canine visceral leishmaniasis (CVL) is great importance in the control leishmaniasis, a disease that neglected worldwide and represents significant public health problem. Enzyme-linked immunosorbent assay (ELISA) test widely used because possibility large-scale low cost. Objective: objective this study was to optimize stoichiometry conjugate EIE/CVL kit; for purpose, some conjugation monitoring parameters were modified through molecular exclusion chromatography...
Two monoclonal antibodies (mAbs) were produced against a serogroup B Neisseria meningitidis strain. These mAbs recognized two epitopes in the class 5 outer membrane proteins (OMP), designated P5.7 and P5.Bm, able to kill homologous strain through complement activation. Both surface exposed 68% of group meningococcal clinical isolates had one or both present their OMP. Antibodies demonstrated 17 patients with meningitis using an ELISA inhibition assay. Of paired sera, 41% 29% acute-phase sera...
Purpose The purpose of this paper is to understand workers’ resistance toward change from a semi-continuous (SCW) continuous shift work system (CW), especially focusing the perspective and its impact on personal lives. Design/methodology/approach A case study was conducted at Portuguese steel plant. questionnaire administered 98 workers two systems, three focus groups (FG) each with ten CW workers, interviews their managers. Findings results FG indicate loss days off during weekends as main...
