Abstract TLR sense microbial infections, and control activation of immune responses. Dendritic cells, macrophages, B lymphocytes express the TLR-signaling adaptor protein MyD88. The impact TLR-activated cells on T cell-mediated inflammation is unknown. In this study, we have used mice carrying cell-restricted deficiencies in MyD88 or distinct to examine a autoimmune disease, experimental encephalomyelitis (EAE). We demonstrate that suppresses inflammatory cell responses (both Th1 Th17),...
The yellow fever vaccine is very effective with a single injection conferring protection for at least 10 years. Recent evidence suggests that the innate immune cells activated through Toll-like receptors (TLRs), are critical determinants of robustness adaptive response. Therefore, we investigated NK cell status in eight healthy volunteers after vaccination YF 17DD virus. Shortly vaccination, observed increased expression TLR-3 and TLR-9 markers such as CD69, HLA-DP-DQ-DR, CD38 CD16....
Here we describe a novel vaccine vector for expressing human immunodeficiency virus (HIV) antigens. We show that recombinant attenuated yellow fever 17D simian SIVmac239 Gag sequences can be used as to generate SIV-specific CD8(+) T-cell responses in the rhesus macaque. Priming with BCG SIV antigens increased frequency of these after YF17D boosting. These YF17D-induced T cells secreted several cytokines, were largely effector memory cells, and suppressed viral replication CD4(+) cells.
Yellow fever (YF) is a viral disease transmitted by mosquitoes and endemic mostly in South America Africa with 20-50% fatality. All current licensed YF vaccines, including YF-Vax® (Sanofi-Pasteur, Lyon, France) 17DD-YFV (Bio-Manguinhos, Rio de Janeiro, Brazil), are based on live attenuated virus produced hens' eggs have been widely used. The vaccines considered safe highly effective. However, recent increase demand for reports of rare cases vaccine-associated fatal adverse events provoked...
Abstract Activated B cells can regulate immunity and have been envisaged as a potential cell‐based therapy for treating autoimmune diseases. However, activated human also propagate immune responses, the effects resulting from their infusion into patients cannot be predicted. This led us to consider resting cells, which in contrast are poorly immunogenic, an alternative cellular platform suppression of unwanted immunity. Here, we report that directly engineered with lentiviral vectors express...
Chagas' disease is a major public health problem affecting nearly 10 million in Latin America. Despite several experimental vaccines have shown to be immunogenic and protective mouse models, there not current vaccine being licensed for humans or clinical trial against T. cruzi infection. Towards this goal, we used the backbone of Yellow Fever (YF) 17D virus, one most effective well-established human vaccines, express an fragment derived from Amastigote Surface Protein 2 (ASP-2). The cDNA...
Yellow Fever vaccine is one of the most efficacious human vaccines ever made. The (YF 17D) virus induces polyvalent immune responses, with a mixed TH1/TH2 CD4+ cell profile, which results in robust T CD8+ responses and high titers neutralizing antibody. In recent years, it has been suggested that early events after yellow fever vaccination are crucial to development adequate acquired immunity. We have previously shown primary immunization humans monkeys YF 17D resulted synthesis IFN-γ....
Abstract Background/Aims: Although eosinophils are considered to play an important role in the pathogenesis of various parasitic, allergic and autoimmune digestive diseases, their fulminant hepatic failure (FHF) is unknown. Our contribution was identify quantify cytokine levels [interleukin (IL)‐6, IL‐5 macrophage inflammatory protein (MIP)‐1α] liver parenchyma peripheral blood from FHF patients at pre‐ post‐transplantation steps. Methods: Histochemical methods were used identify/quantify...
Abstract Since its emergence in late 2019, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been posing a serious threat to public health worldwide as the causative agent of disease 2019 (COVID-19). Now distributed pandemic pattern, this still lacks an effective drug treatment with low toxicity, leading pharmaceutical companies and research labs work against time find candidate molecule efficiently treat affected patients. Due well-known broad-spectrum antimicrobial activity...
Successful SARS-CoV-2 inactivation allows its safe use in Biosafety Level 2 facilities, and the of whole viral particle helps development analytical methods a more reliable immune response, contributing to improvement vitro vivo assays. In order obtain functional product, we evaluated several protocols observed that 0.03% beta-propiolactone for 24 h was best condition tested, as it promoted above 99.99% no cytopathic effect visualized after five serial passages. Moreover, RT-qPCR...
Fungal infections cause 1.7 million deaths annually, which can be attributed not only to fungus-specific factors, such as antifungal resistance and biofilm formation, but also drug-related challenges. In this study, the potential of Amphotericin (AmB) loaded polymeric nanoparticles (AmB-NPs) combined with murine monoclonal antibodies (mAbs) (i.e., CC5 DD11) was investigated a strategy overcome these To achieve goal, AmB-NPs were prepared by nanoprecipitation using different polymers...
Host factors that influence Congenital Zika Syndrome (CZS) outcome remain elusive. Interferons have been reported as the main antiviral factor in and other flavivirus infections. Here, we accessed samples from 153 pregnant women (77 without 76 with CZS) 143 newborns 66 exposed to ZIKV conducted a case-control study verify whether interferon alfa receptor 1 (IFNAR1) lambda 2 4 (IFNL2/4) single nucleotide polymorphisms (SNPs) contribute CZS outcome, characterized placenta gene expression...
Abstract Human Mpox is a highly contagious viral disease caused by virus, currently causing outbreaks in various regions of the world and considered global public health problem. Understanding how virus adapts or transfers to new host species crucial for effective prevention treatment infections. A comprehensive analysis 1,318 accessible genomes was conducted assess potential phenotypic effects, incorporating examination pseudogenes, gene duplication deletion, as well sequences obtained from...
Zika virus (ZIKV) infections during pregnancy can result in Congenital Syndrome (CZS), a range of severe neurological outcomes fetuses that primarily occur early gestational stages possibly due to placental damage. Although some placentas maintain ZIKV persistence for weeks or months after the initial infection and diagnosis, impact this viral is still unknown. Here, we aimed investigate immunological repercussion term placentas. As such, from 64 pregnant women diagnosed with different...
The yellow fever vaccine (YF17DD) is highly effective with a single injection conferring protection for at least 10 years. YF17DD induces polyvalent responses, TH1/TH2 CD4+ profile, robust T CD8+ and synthesis of interferon-gamma (IFN-γ), culminating in high titers neutralizing antibodies. Furthermore, C-type lectin domain containing 5A (CLEC5A) has been implicated innate outcomes other flaviviral infections. Here, we conducted follow-up study volunteers immunized YF17DD, investigating the...
Background Orthohantavirus infection is a neglected global health problem affecting approximately 200,000 people/year, spread by rodent hosts and associated to fatal human diseases, such as hemorrhagic fever with renal syndrome (HFRS) orthohantavirus cardiopulmonary (HCPS). Circulation of HFRS-associated orthohantaviruses, Seoul, Gou, Amur, Dobrava Hantaan, are supposed be restricted Eurasian countries even though their can worldwide distribution. Few confirmed HFRS infections in humans have...
Abstract Background Alouatta spp. are highly susceptible to yellow fever (YF) infection and develop an often fatal disease. The threat posed by outbreak started in 2016 leads us investigate vaccination as a potential tool preventing YF non‐human primates (NHP). Methods Susceptible howler monkeys were immunized with three different concentrations of the human Brazilian commercial YF17DD vaccine. Post‐vaccination viremia/RNAemia, immunogenicity, safety characterized. Results vaccine did not...
