Spiro-oxazolidinones are highly valuable compounds in the fields of medicinal and organic chemistry; however, methods for synthesizing these have not been well established. Herein, we present a biocatalytic methodology construction spiro-oxazolidinones through halohydrin dehalogenase-catalyzed ring expansion spiro-epoxides. By performing screening protein engineering dehalogenases, both chiral racemic were synthesized 24–47% yields (90–98% ee) 69–98% yields, respectively. The method was also...
Although biocatalysis offers complementary or alternative approaches to traditional synthetic methods, the limited range of available enzymatic reactions currently poses challenges in synthesizing a diverse array desired compounds. Consequently, there is significant demand for developing novel biocatalytic processes enable that were previously unattainable. Herein, we report discovery and subsequent protein engineering unique halohydrin dehalogenase develop platform enantioselective...
MacB is a noncanonic ABC-type transporter within Gram-negative bacteria, which responsible both for the efflux of macrolide antibiotics and secretion heat-stable enterotoxin II. In Escherichia coli, requires membrane fusion protein MacA multifunctional outer channel TolC to pump substrates external medium. Sequence analysis suggested that has relatively large periplasmic region. To gain insight into how assembles with TolC, we determined crystal structure region Actinobacillus...
With the continuous development of drug resistance in bacteria to traditional antibiotics, demand for novel antibacterial agents is urgent. Antimicrobial peptides (AMPs) are promising candidates because their unique mechanism action and low tendency induce resistance. Previously, we cloned temporin-GHb (hereafter referred simply as "GHb") from Hylarana guentheri. In this study, a series derived were designed, namely, GHbR, GHbK, GHb3K, GHb11K, GHbK4R. The five had stronger activities against...
The development of catalytic enantioselective cyanation methods for preparing valuable chiral nitriles is great interest in the areas pharmaceutical synthesis and organic chemistry. In this study, we presented an enzymatic strategy β-hydroxy using cyanohydrins as cyano sources. By combining enzyme screening protein engineering halohydrin dehalogenases, biocatalytic various aryl, alkyl, spiro epoxides was achieved to afford corresponding good yields (up 47%) excellent optical purities >99%...
<title>Abstract</title> The remarkable selectivity, sustainability, and efficiency afforded by biocatalytic strategies position them as complements or alternatives to traditional synthetic methods. Nevertheless, the currently narrow spectrum of enzymatic reactions available imposes limitations on synthesizing diverse desired compounds. Consequently, there continues be a high demand for developing novel processes access that were previously unattainable. Herein, we report discovery subsequent...
Abstract An unprecedented catalytic system consisting of diselenides, Selectfluor and visible light has been developed for the one‐pot synthesis oxazole acetals from N ‐propargylamides alcohols. This shows processes cyclization, aromatization acetalization, resulting in construction multiple carbon‐oxygen bonds. These transformations demonstrate excellent compatibility visible‐light promotion with organocatalytic oxidative carbon–oxygen bond formations using air as terminal oxidant.
Temporin-GHa (GHa) was cloned from , showing a weak antimicrobial activity. In order to improve its bactericidal efficacy, GHaR6R, GHaR7R, GHaR8R and GHaR9W were designed synthesized. Compared the parent peptide, GHa-derived peptides show potent activities against methicillin-resistant (MRSA), which is main pathogen with high morbidity mortality that causes various infections in humans. These exert actions on MRSA by permeabilizing cytoplasmic membranes damaging membrane integrity. All of...
Introduction Temporin-GHa obtained from the frog Hylarana guentheri showed bactericidal efficacy against Streptococcus mutans . To enhance its antibacterial activity, derived peptides GHaR and GHa11R were designed, their performance, antibiofilm potential in inhibition of dental caries evaluated. Methods Bacterial survival assay, fluorescent staining assay transmission electron microscopy observation applied to explore how inhibited killed S. The was assayed by examining exopolysaccharide...
Taking six varieties of Michelia as test materials, their responses under cold situation in the field were investigated and semilethal low temperatures calculated by fitting Logistic equation. The nine structure indexes leaf tissue observed paraffin section, a comprehensive evaluation on resistance different was given according to subordinate function value analysis. results showed that relative electrical conductivity significantly positively correlated with temperature (LT50) 3 h 0-25 ℃...
Panji is rich in coal reserves. The rapid development of social economy and increasing mining scales cause the subsided land, bulk deposition solid waste, soil, water air pollution, crop yield reduction or failure ecological deterioration, which endanger living people area. Based on current situation subsidence area evaluation land reclamation potentiality, this paper puts forward establishment economic pattern application to project improve condition, cultivate resource potentiality restore...
The ecosystem structure and functions of the manganese ore lands have been severely degraded. In order to attain purpose ecological recovery landscape restoration, this paper first summarizes characteristics current status on then lists species indigenous plants that apply restoration by starting with vegetation from perspective landscape, finally elaborates application in different arrangement modes.
