Abstract Proteolysis targeting chimeras (PROTACs) are an emerging strategy for promoting targeted protein degradation by inducing the proximity between proteins and E3 ubiquitin ligases. Although successful of numerous PROTACs has been demonstrated, elements that determine degradability PROTAC-targeted have not yet explored. In this study, we developed von Hippel-Lindau-Cereblon (VHL-CRBN) heterodimerizing induce CRBN, but VHL. A quantitative proteomic analysis further revealed VHL-CRBN...

Molecular glue degraders, such as lenalidomide and pomalidomide, bind to cereblon (CRBN) E3 ligase subsequently recruit neosubstrate proteins, Ikaros (IKZF1) Aiolos (IKZF3), for the ubiquitination-proteasomal degradation process. In this study, we explored structure-activity relationship analysis novel GSPT1 degraders utilizing a benzotriazinone scaffold previously discovered CRBN binder. particular, focused on position of ureido group scaffold, substituent effect phenylureido group, methyl...

Indoleamine 2,3-dioxygenase 1 (IDO1) is an immunosuppressive enzyme that highly overexpressed in various cancer cells and antigen-presenting cells. It has emerged as attractive therapeutic target for immunotherapy, which prompted high interest the development of small-molecule inhibitors. To discover novel IDO1 inhibitors, we designed synthesized a series N'-hydroxyindazolecarboximidamides. Among compounds synthesized, compound 8a inhibited both tryptophan depletion kynurenine production...

The adaptive wavelet method is studied for the enhancement of computational efficiency three-dimensional flows. Three-dimensional decomposition process introduced based on previous two-dimensional method. order numerical accuracy an original solver preserved by applying a proper thresholding technique. proposed algorithm applied to computation flow field around ONERA-M6 wing in transonic regime. Through application, it confirmed that can reduce time with conserving solver.
