To evaluate the diversity of extended-spectrum β-lactamases (ESBL) genes among food-producing animals, 48 isolates ESBL-producing Escherichia coli were obtained from rectal samples broilers, layers, beef cattle and pigs, at slaughterhouse level. ESBL-carrying E. isolated 60.0% individual broiler samples, 5.9% 12.5% 3% pigs. One Klebsiella pneumoniae was a broiler. The ESBL-positive broilers harbored various ESBL genes: bla (SHV-12), bla(CTX-M-2), bla(CTX-M-14), bla(CTX-M-15) bla(CTX-M-44)....
ABSTRACT Clostridium perfringens is a causative agent of food-borne gastroenteritis for which C. enterotoxin (CPE) has been considered an essential factor. Recently, we experienced two outbreaks in non-CPE producers were strongly suspected to be the cause. Here, report novel produced by isolates, BEC (binary ). Culture supernatants strains showed fluid-accumulating activity rabbit ileal loop and suckling mouse assays. Purification enterotoxic substance high-throughput sequencing genomic DNA...
ABSTRACT In Escherichia coli , more than 180 O groups and 53 H types have been recognized. The O:H serotyping of E. strains is an effective method for identifying with pathogenic potential classifying them into clonal groups. particular, the Shiga toxin-producing (STEC) provides valuable information to evaluate routes, sources, prevalence agents in outbreak investigations surveillance. Here, we present a complete practical PCR-based H-typing system, H-genotyping PCR, consisting 10 multiplex...
We compared the efficiency of universal pre-enrichment broth (UPB), modified Escherichia coli containing novobiocin (mEC + n), Tryptic Soy Broth (mTSB) and mTSB with (mTSB n) for enrichment non-O157 Shiga-toxin-producing E. (STEC).Freeze-injured control STEC (O91, O103, O111, O119, O121, O145 O165) strains were used to artificially contaminate beef radish sprout samples, which then cultivated in each four media. After incubation, detected by loop-mediated isothermal amplification (LAMP)...
Antimicrobial resistance to quinolones, which constitutes a threat public health, has been increasing worldwide. In this study, we investigated the prevalence of quinolone-resistant determinants in Escherichia coli not susceptible quinolones and isolated from food-producing animals food derived them, Philippines. A total 791 E. strains were 56.4% 601 beef, chicken, pork, egg, milk samples, as well environmental, cloacal, rectal swab-collected samples supermarkets, open markets, abattoirs,...
Abstract The spread of vancomycin-resistant enterococci (VRE) is a major threat in nosocomial settings. A large-scale multiclonal VRE outbreak has rarely been reported Japan due to low prevalence. We evaluated the transmission vancomycin resistance outbreak, conducted biological and genomic analyses isolates, assessed implemented infection control measures. In total, 149 patients harboring VanA-type were identified from April 2017 October 2019, with 153 Enterococcus faecium isolated being...
The multilocus variable-number tandem repeat analysis (MLVA) method to target eight loci, based on agarose gel electrophoresis separation of multiplexed PCR products, and the PFGE were applied clinical isolates Escherichia coli O157 : H7 with aim comparing their performance as methods typing this bacterium. Using MLVA, a total 57 from patients in Shizuoka prefecture, Japan, divided into 20 types classified 23 types. Twenty-four four sporadic infections, household contact infections one...
Kudoa septempunctata is a newly identified myxosporean parasite of olive flounder (Paralichthys olivaceus) and suspected causative agent several food-borne gastroenteritis outbreaks in Japan. Here, we report the detection K. 18S ribosomal DNA fecal samples outbreak patients using an efficient method based on real-time PCR. We first performed spiking experiment to assess whether our previously developed PCR assay was applicable detect feces. Simultaneously, compared relative extraction...
Two Gram-stain-positive strains, VE80T and VE116, which were resistant to vancomycin, isolated from retail chicken meat liver in Ho Chi Minh, Vietnam, respectively. These strains characterized by sequence analyses of 16S rRNA, RNA polymerase α-subunit (rpoA), ATP synthase (atpA), phenylalanyl-tRNA (pheS) genes, determination DNA G+C content, cellular fatty acid methyl ester analysis, DNA-DNA hybridization, conventional morphological biochemical tests. Strains VE116 had 99.6 % rRNA gene...
Human infection with Streptococcus canis is extremely rare. We describe herein a case of septicemia cellulitis caused by S. in 75-y-old woman, which developed 2 weeks after dog bite. Macrorestriction analysis pulsed-field gel electrophoresis demonstrated that the organism had been transmitted means bite to her hand.
We describe our laboratory investigation of a massive foodborne outbreak gastrointestinal illness caused by enterotoxigenic Escherichia coli (ETEC) serotype O169:H41 that occurred during 2-day traditional festival held in September 2012 Osaka Prefecture, Japan. Of 126 customers who patronized particular Japanese restaurant the event, 102 developed symptoms disease. isolated strains ETEC from 1 food sample and fecal samples collected 19 34 patients 2 4 handlers. Pulsed-field gel...
The serotype, Shiga toxin (Stx) type, and antimicrobial resistance patterns of 138 Stx-producing Escherichia coli (STEC) strains isolated from humans between 2003 2007 in Shizuoka Prefecture, Japan were characterized. predominant O serogroups the STEC isolates O157, O26, O111. Antimicrobial susceptibility testing showed that 31 (22.5%) resistant to antibiotics. Compared results reported previous studies, a higher rate O157 susceptible all agents used this study. However, data study have...
ABSTRACT In September 2016, 140 patients with primary symptoms of sore throat and fever were identified in a school dormitory Osaka, Japan. Epidemiological laboratory investigations determined that these symptomatic conditions from foodborne outbreak group G streptococcus (GGS), GGS being isolated samples patients, cooks, foods. The strain was as Streptococcus dysgalactiae subsp. equisimilis two emm types ( stG652.0 stC36.0 ). causative food, broccoli salad, contaminated the S. , totaling...
Escherichia albertii is an emerging diarrheagenic pathogen causing food- and water-borne infection in humans. However, no selective enrichment broths for E. have ever been reported. In this study, we tested several basal media, supplements culture conditions which enabled of albertii. We developed a broth, novobiocin-cefixime-tellurite supplemented modified tryptic soy broth (NCT-mTSB). NCT-mTSB supported the growth 22 strains, while inhibited other Enterobacteriaceae at 37°C, except coli...
Shiga toxin (Stx)-producing Escherichia coli (STEC) are foodborne pathogens causing serious diseases, such as haemorrhagic colitis and haemolytic uraemic syndrome. Although O157:H7 STEC strains have been the most prevalent, incidences of infections by several other serotypes recently increased. O121:H19 is one these major non-O157 STECs, but systematic whole genome sequence (WGS) analyses not yet conducted on this STEC. Here, we performed a global WGS analysis 638 strains, including 143...
