A low dose of 1μg rhBMP-2 was immobilised by four different functionalising techniques on recently developed poly(l-lactide)-co-(ε-caprolactone) [(poly(LLA-co-CL)] scaffolds. It either (i) physisorbed unmodified scaffolds [PHY], (ii) onto modified with nanodiamond particles [nDP-PHY], (iii) covalently linked nDPs that were used to modify the [nDP-COV] or (iv) encapsulated in microspheres distributed [MICS]. Release kinetics BMP-2 from quantified using targeted mass spectrometry for up...

Abstract Introduction Rapid establishment of functional blood vessels is a prerequisite for successful tissue engineering. During vascular development, endothelial cells (ECs) and perivascular assemble into complex regulating proliferation ECs, vessel diameter production extracellular matrix proteins. The aim this study was to evaluate the ability mesenchymal stem (MSCs) establish an endothelial-perivascular in tissue-engineered constructs comprising ECs MSCs. Methods Primary human MSCs were...

Significant evidence has indicated that poly(L-lactide)-co-(ɛ-caprolactone) [(poly(LLA-co-CL)] scaffolds could be one of the suitable candidates for bone tissue engineering. Oxygen-terminated nanodiamond particles (n-DP) were combined with poly(LLA-co-CL) and revealed to positive cell growth. In this study, we evaluated influence modified by n-DP on attachment, proliferation, differentiation marrow stromal cells (BMSCs) in vitro, formation using a sheep calvarial defect model. Constructs intended for bone tissue engineering (TE) are influenced by the initial cell seeding density. Therefore, objective of this study was to determine effect marrow stromal stem cells (BMSCs) density loaded onto copolymer scaffolds on regeneration. BMSCs were harvested from rat's and cultured in media with or without osteogenic supplements. Cells seeded poly(l-lactide-co-ε-caprolactone) [poly(LLA-co-CL)] at two different densities: low (1 × 10(6) cells/scaffold) high (2 using spinner...

Abstract Introduction A major determinant of the potential size cell/scaffold constructs in tissue engineering is vascularization. The aims this study were twofold: first to determine vitro angiogenic and osteogenic gene-expression profiles endothelial cells (ECs) mesenchymal stem (MSCs) cocultured a dynamic 3D environment; second, assess differentiation for osteogenesis after vivo implantation. Methods MSCs ECs grown culture poly(L-lactide-co-1,5-dioxepan-2-one) (poly(LLA-co-DXO)) copolymer...

The aim is to evaluate the effect of modifying poly[( l ‐lactide)‐co‐(ε‐caprolactone)] scaffolds (PLCL) with nanodiamonds (nDP) or nDP+physisorbed BMP‐2 (nDP+BMP‐2) on in vivo host tissue response and degradation. are implanted subcutaneously Balb/c mice retrieved after 1, 8, 27 weeks. Molecular weight analysis shows that modified degrade faster than unmodified. Resolvin D1 (RvD1) is a pro-resolving lipid mediator of inflammation, endogenously synthesized from omega-3 docosahexaenoic acid. The purpose this study was to investigate the effect RvD1 on bone regeneration using rat calvarial defect model. Collagen 3D nanopore scaffold (COL) and Pluronic F127 hydrogel (F127) incorporated with (RvD1-COL-F127 group) or COL (COL-F127 were implanted in symmetrical defects.
