Bullous pemphigoid (BP) is a type 2 inflammation- and immunity-driven skin disease, yet comprehensive understanding of the immune landscape, particularly immune-stromal crosstalk in BP, remains elusive. Herein, using single-cell RNA sequencing (scRNA-seq) vitro functional analyzes, we pinpoint Th2 cells, dendritic cells (DCs), fibroblasts as crucial cell populations. The IL13-IL13RA1 ligand-receptor pair identified most significant mediator BP. Notably, DCs expressing IL13RA1 respond to...
Background and Aims: RAD51 recombinase (RAD51) is a highly conserved DNA repair protein indispensable for embryonic viability. As result, the role of in liver development function unknown. Our aim was to characterize postnatal development. Approach Results: expressed during regeneration following hepatectomy hepatic injury, also elevated chronic diseases. We generated hepatocyte-specific Rad51 deletion mouse model using Alb -Cre ( -conditional knockout (CKO)) Adeno-associated virus...
Lepromatous leprosy (L-LEP), caused by the massive proliferation of Mycobacterium leprae primarily in macrophages, is an ideal disease model for investigating molecular mechanism intracellular bacteria evading or modulating host immune response. Here, we performed single-cell RNA sequencing both skin biopsies and peripheral blood mononuclear cells (PBMCs) L-LEP patients healthy controls. In lesions, revealed remarkable upregulation APOE expression that showed a negative correlation with...
Pathogen-induced epigenetic modifications can reshape anti-infection immune processes and control the magnitude of host responses. DNA methylation profiling has identified crucial aberrant changes associated with diseases, thus providing biological insights into roles factors in mycobacterial infection. In this study, we performed a genome-wide analysis skin biopsies from patients leprosy healthy controls. T helper 17 differentiation pathway was found to be significantly through functional...
Convective heat transfer on the exterior surface of building envelope is an important component for energy consumption. The calculation consumption depends convective coefficient (CHTC) envelope. existing research does not fully consider effects airflow field around CHTC different surfaces. In this paper, relationships between and influence factors were investigated isolated building. Response methodology (RSM) support vector machine (SVM) algorithms integrated with single simulation to...
Dapsone hypersensitivity syndrome (DHS) is restricted to HLA-B∗13:01. However, the positive predictive value for HLA-B∗13:01 only 7.8%. To explore potential coexisting factors involved in occurrence of DHS, we carried out a GWAS and genome-wide DNA methylation profile analysis comparing patients with DHS dapsone-tolerant control subjects (all carrying HLA-B∗13:01). No non-HLA SNPs associated were identified at level. pathway antigen processing presentation was enriched gene TAP2 identified....
Leprosy, a chronic infectious disease, and psoriasis, an inflammatory disorder, are distinct entities. Epidemiology data show that these two diseases almost mutually exclusive, with only few reported cases of their coexistence. Here, we present the case patient manifesting intermingled psoriatic leprosy lesions diagnosed as borderline lepromatous plaque psoriasis. Of note, Mycobacterium leprae bacilli were detected not in types but also normal-appearing skin blood.
Abstract Aseptic pustulosis involves inflammatory skin conditions with nonbacterial pustules on erythema, accompanied by neutrophil and eosinophil infiltration in the epidermis. Dysregulation of interleukin (IL)-36 pathway leads to aggregation pustule formation. Variants IL36RN, CARD14, AP1S3, MPO, SERPINA3 BTN3A3 have been identified generalized pustular psoriasis (GPP) past. Some patients acrodermatitis continua Hallopeau (ACH), palmoplantar acute exanthematous (AGEP) also exhibit...
The neurotropism of Mycobacterium leprae is a unique characteristic that distinguishes it from other mycobacteria. However, the mechanism peripheral nerve injury in leprosy still unclear. In this study, immunohistochemistry and qPCR found skin lesions people with expressed IL-22 at significantly higher rate than mycobacterium-infected lesions, was concentrated around cutaneous nerves. Further investigation involved by regulating neuroinflammation promoting apoptosis via JAK-STAT pathway.
Severe cutaneous adverse reactions (SCARs) mediated by cytotoxic T lymphocytes are a series of life-threatening conditions with mortality 4%-20%. The clinical application tumor necrosis factor-alpha (TNF-α) antagonist improves the outcome some SCARs patients; however, this is complicated elusive and varied immunopathogenesis.
M. leprae preferentially infects Schwann cells (SCs) in the peripheral nerves leading to nerve damage and irreversible disability. Knowledge of how interacts with host SCs is essential for understanding mechanisms revealing potential new therapeutic strategies. We performed a time-course single-cell sequencing analysis infected at different time points, further analyzed heterogeneity SCs, subpopulations associated infection, developmental trajectory validated by Western blot or flow...
Cutaneous Mycobacterium marinum infection is an increasingly infectious disease presenting unique diagnostic and therapeutic challenges. The aim of this study was to evaluate the differences in time treatment among patients with different types skin lesions who were treated single or multidrug therapies. In addition, clinical characteristics M. explored mechanism host immune responses investigated. electronic medical records 35 reviewed. characteristics, histopathological laboratory data,...
The thymus is required for T cell development and the formation of adaptive immunity. Stromal cells, which include thymic epithelial cells (TECs) mesenchymal stromal (MSCs), are essential function. However, immunomodulatory function thymus-derived MSCs (T-MSCs) has not been fully explored.MSCs were isolated from mouse their general characteristics including surface markers multi-differentiation potential characterized. T-MSCs stimulated by IFN-γ TNF-α was evaluated in vitro vivo....
This study examined age differences in daily life events related to different types of social goals based on the socioemotional selectivity theory (SST), and determined whether positivity effect existed context older adults' lives. Over a course 14 days, 49 adults 36 younger wrote about up three rated valence each event. The findings indicated that (1) although both recorded emotional knowledge-acquisition goals, odds ratio for reporting higher number compared was 2.12 times than observed...
Sézary syndrome (SS) is a rare and aggressive type of cutaneous T cell lymphoma (CTCL) with poor prognosis. Intra-tumoral heterogeneity caused by different disease compartments (e.g., skin, blood) understanding the pathogenesis has created obstacles to precise diagnosis targeted treatment disease. Here we performed comprehensive analysis integrating single-cell transcriptomic data 40,333 peripheral blood mononuclear cells (PBMCs) 41,580 skin cells, as well chromatin accessibility 11,058...
Keratinocytes are the predominant cell type in skin epidermis, and they not only protect from influence of external physical factors but also function as an immune barrier against microbial invasion. However, little is known regarding defence mechanisms keratinocytes mycobacteria. Here, we performed single-cell RNA sequencing (scRNA-seq) on biopsy samples patients with Mycobacterium marinum infection bulk (bRNA-seq) M. marinum-infected vitro. The combined analysis scRNA-seq bRNA-seq data...
The scale of cities is increasing with continuous urban development. Effective methods, such as the source term estimation (STE) method, must be established for identifying sources air pollution in to prevent economic losses and casualties caused by pollutant leakage. Herein, methods optimizing sensor configuration are discussed, an STE method based on regularized minimum residual proposed. Urban wind environments were simulated using a computational fluid dynamics (CFD) model, results...
