Abstract Oral squamous cell carcinoma (OSCC) is one of the most common malignancies and has a poor prognosis. Circular RNA (circRNA) been increasingly recognized as crucial contributor to carcinogenesis. circRNA_0000140 aberrantly expressed in OSCC, but its role tumor growth metastasis remains largely unclear. Sanger sequencing, actinomycin D, RNase R treatments were used confirm head-to-tail junction sequences stability circ_0000140. In vitro activities, including proliferation, migration,...

Eukaryotic chromosomes are folded into higher-order conformations to coordinate genome functions. In addition long-range chromatin loops, recent chromosome conformation capture (3C)-based studies have indicated higher levels of structures including compartments and topologically associating domains (TADs), which may serve as units organization However, the molecular machinery underlying these hierarchically three-dimensional (3D) architectures remains poorly understood. Via high-throughput...

Cervical cancer is one of the most common gynecological malignancies with poor prognosis due to constant chemoresistance and repeated relapse. Ciclopirox olamine (CPX), a synthetic antifungal agent, has recently been identified be promising anti-cancer candidate. However, detailed mechanisms related its effects remain unclear need further elucidated. In this study, we found that CPX could induce proliferation inhibition in cervical cells by targeting PARK7. Further results demonstrated...

The tumor microenvironment is comprised of cancer cells and various stromal their respective cellular components. Cancer-associated fibroblasts (CAFs), a major part the cells, are key determinant in progression, while glucose-regulated protein (GRP)78 overexpressed many human cancers involved invasion metastasis. This study developed microfluidic-based three dimension (3D) co-culture device to mimic an vitro order investigate cell real-time. bionic chip provided significant information...

Coactivator-associated arginine methyltransferase 1 (CARM1) belongs to the protein family. CARM1 has been reported be associated with high grade tumors in breast cancer. It still remains unknown expression pattern of cancer and its relationships clinicopathological characteristics molecular subtypes. Two hundred forty-seven invasive cases were collected prepared for tissue array. There thirty-seven benign glandular epithelium adjacent among these cases. Molecular subtype investigated using...

Abstract In the interphase nucleus, chromatin is organized into three-dimensional conformation to coordinate genome functions. The lamina-chromatin association important facilitate higher-order in mammalian cells, but its biological significances and molecular mechanisms remain poorly understood. One obstacle that list of lamina-associated proteins remains limited, presumably due inherent insolubility lamina proteins. this report, we identified 182 associated with lamin B1 (a constitutive...

The architecture and function of chromatin are largely regulated by local interacting molecules, such as transcription factors noncoding RNAs.However, our understanding these regulatory molecules at a given locus is limited because technical difficulties. Here, we describe the use Clustered Regularly Interspaced Short Palindromic Repeats an engineered ascorbate peroxidase 2 (APEX2) system to investigate interactions (CAPLOCUS).We showed that with specific small-guide RNA targets, CAPLOCUS...

Hepatocellular carcinoma is one of the most common fatal malignancies worldwide. Thus far, hepatocellular prognosis has been bleak due to deficiencies in identification and diagnosis early carcinoma. Ciclopirox olamine (CPX) a synthetic antifungal agent considered as an anti-cancer candidate drug recently, though detailed mechanisms related its effect have not yet revealed. Here, we found that CPX could inhibit proliferation HCC cells but intrahepatic cholangiocarcinoma by arresting cell...

The nuclear matrix-associated protein Heterogeneous Nuclear Ribonucleoprotein U (HNRNPU), also known as SAF-A, is to maintain active chromatin structure in mouse hepatocytes. However, the functional roles and molecular mechanisms of HNRNPU development hepatocellular carcinoma (HCC) remain largely unknown. Herein, we found that was upregulated HCC, proliferation HCC cells inhibited vitro vivo upon knockdown. Moreover, upregulation correlated with poor prognosis HCC. Mechanistically, bound...
