Abstract UPF1, a novel posttranscriptional regulator, regulates the abundance of transcripts, including long noncoding RNAs (lncRNAs), and thus plays an important role in cell homeostasis. In this study, we revealed that UPF1 hepatocellular carcinoma upregulated EZH2-associated lncRNA (lncRNA-HEIH ) by binding CG-rich motif, thereby regulating (HCC) tumorigenesis. UPF1-bound lncRNA-HEIH was susceptible to degradation mediated phosphorylation via SMG1 SMG5. According analysis RNA-seq public...

Abstract The stability and quality of metazoan mRNAs are under microRNA (miRNA)-mediated nonsense-mediated control. Although UPF1, a core mediator mRNA decay (NMD), mediates the target in 3′UTR-length-dependent manner, detailed mechanism remains unclear. Here, we suggest that is not mediated by nonsense but rather miRNAs downregulate via Ago-associated UPF1/SMG7. Global analyses response to UPF1 RNA interference miRNA-deficient cells reveal requires canonical miRNA targeting. destabilization...

Proteoglycans function not only as structural components of the extracellular compartment but also regulators various cellular events, including cell migration, inflammation, and infection. Many microbial pathogens utilize proteoglycans to facilitate adhesion invasion into host cells. Here we report a secreted form novel heparan sulfate proteoglycan that functions against virus The expression SPOCK2/testican-2 was significantly induced in virus-infected lungs or interferon (IFN)-treated...

Abstract UPF1 and LIN28A are RNA-binding proteins involved in post-transcriptional regulation stem cell differentiation. Most studies on have focused the molecular mechanisms of differentiated cells differentiation, respectively. We reveal that directly interacts with before UPF1-UPF2 complexing, thereby reducing phosphorylation inhibiting nonsense-mediated mRNA decay (NMD). identify interacting domains LIN28A; moreover, we develop a peptide impairs UPF1-LIN28A interaction augments NMD...

Abstract PAF complex is an evolutionarily conserved transcriptional that associates with RNA polymerase II in the coding region of actively transcribing genes. Although its activity closely related to diverse cellular processes, such as cell-cycle progression or development mammals, role immune responses has not been addressed yet. In this study, we show CTR9, a component complex, functions repressor Th17 differentiation. Both mRNA and protein levels CTR9 were significantly decreased during...

SUMMARY Cyclic GMP-AMP synthase (cGAS) is a primary sensor of aberrant DNA that governs an innate immune signaling cascade, leading to the induction type-I interferon response. We have previously identified polyglutamine binding protein 1, PQBP1, as adaptor molecule required for cGAS-mediated response lentiviruses, including human immunodeficiency virus 1 (HIV-1), but dispensable recognition viruses. HIV-1- encoded synthesized single copy from its RNA genome, and subsequently integrated into...
