The Tityus serrulatus scorpion is considered the most dangerous in Brazil and responsible for several cases of human envenomation annually. In this study, we performed transcriptome profiling T. venom gland. In addition to transcripts with housekeeping functions, such as those related protein synthesis, energy supply structural processes, from thirty-five families peptides or proteins were identified. These included three new complete sequences toxins more than a dozen putative gland...

Scorpionism is a public health problem in Brazil, and Tityus serrulatus (Ts) primarily responsible for severe accidents. The main toxic components of Ts venom are low-molecular-weight neurotoxins; however, the also contains poorly characterized high-molecular-weight enzymes. Hyaluronidase one such enzyme that has been characterized. We examined clones from cDNA library gland described two novel isoforms hyaluronidase, TsHyal-1 TsHyal-2. 83% identical, alignment their predicted amino acid...

Wissadula periplocifolia (L.) C. Presl (Malvaceae) is commonly used in Brazil to treat bee stings and as an antiseptic. The antioxidant properties of its extracts have been previously demonstrated, thus justifying a phytochemical investigation for bioactive phenolic constituents. This has yielded five new sulphated flavonoids: 8-O-sulphate isoscutellarein (yannin) (1a); 4′-O-methyl-7-O-sulphate (beltraonin) (1b); 7-O-sulphate acacetin (wissadulin) (2a); 4′-O-methyl-8-O-sulphate (caicoine)...

Tityus serrulatus is a Brazilian scorpion species with great medical significance. While the effects of neurotoxins have been extensively studied, little known about proteases expressed in venom gland this arthropod. In study, clones from T. (Ts) cDNA library were selected according to homology proteases. The sequences aligned database and classified by homology. Similarity identity analyses carried out, phylogenetic tree was constructed other These correspond ten different metalloproteases,...

The hyaluronidase enzyme is generally known as a spreading factor in animal venoms. Although its activity has been demonstrated several organisms, deeper knowledge about and the venom process from bite/sting site until elimination victim's body still need. Herein, we further pursued goal of demonstrating effects inhibition T. serrulatus (TsV) on biodistribution. We used technetium-99m radiolabeled Tityus (99mTc-TsV) to evaluate distribution kinetics mice. To understand hyaluronidase's role...

Envenoming resulting from Loxosceles spider bites (loxoscelism) is a recognized public health problem in Brazil. However, the pathophysiology of loxoscelism caused by L. similis bites, which widespread Brazil, remains poorly understood. In present work, RNA sequencing (RNA-Seq - Next Generation NGS) venom gland was performed to identify and analyze sequences key component phospholipase D. The were aligned based on their classical domains, phylogenetic tree constructed. bioinformatics...

Epsilon toxin produced by Clostridium perfringens types B and D causes enterotoxemia in sheep, goats calves.Enterotoxemia can cause acute or superacute disease, with sudden death of the affected animal. It provokes huge economic losses when large numbers livestock are affected.Therapeutic intervention is challenging, because disease progresses very rapidly. However, it be prevented immunization specific immunogenic vaccines. We cloned etx gene, encoding epsilon toxin, into vector pET-11a;...

Detection of microbial enzymes in natural environments is important to understand biochemical activities and verify the biotechnological potential microorganisms. In present report, 346 isolates from soil, water, plants were screened for enzyme production (caseinase, gelatinase, amylase, carboxymethyl cellulase, esterase). Our results showed that 89.6% produced at least one tested enzyme. A predominance amylase soil samples, cellulase plants, as well esterase gelatinase water was observed....

Scorpion sting envenoming impacts millions of people worldwide, with cardiac effects being one the main causes death on victims. Here we describe first Ca2+ channel toxin present in Tityus serrulatus (Ts) venom, a cell penetrating peptide (CPP) named CPP-Ts. We show that CPP-Ts increases intracellular release through activation nuclear InsP3R cardiomyocytes, thereby causing an increase contraction frequency these cells. Besides proposing novel subfamily active toxins, investigated its...
