Abstract The mouse Igh locus is organized into a developmentally regulated topologically associated domain (TAD) that divided subTADs. Here we identify series of distal V H enhancers (E VH s) collaborate to configure the locus. E s engage in network long-range interactions interconnect subTADs and recombination center at D J gene cluster. Deletion 1 reduces rearrangement its vicinity alters discrete chromatin loops higher order conformation. Reduction 11 used anti-PtC responses likely cause...

Polysensitized patients require allergen immunotherapy (AIT) targeting multiple allergens. However, combining extracts can lead to instability and reduced efficacy particularly due the high proteolytic activity of house dust mite (HDM) While is known that glutaraldehyde cross-linking may reduce enzymatic activity, its ability stabilize multi-allergen formulations protect key allergens from degradation remains unexplored. To evaluate impact polymerization on stability immunogenicity HDM grass...

Saliva and muscle-derived mosquito allergens have been purified characterized. However, the complete set of remains to be elucidated. In this study, we identified characterized IgE-binding proteins from species Aedes aegypti.Serum was obtained 15 allergic individuals with asthma and/or rhinitis sensitized mosquito. IgE binding determined by ELISA. Total freeze-dried bodies A. aegypti were extracted IgE-reactive 2D gel electrophoresis, followed Western blot pooled or individual sera. spots...

The mosquito Aedes aegypti is a potential source of important clinically relevant allergens. However, the allergenicity and cross-reactivity most these has not been fully described.Natural wild-type tropomyosin was purified by size exclusion anionic-exchange chromatography from an A. extract. Further characterization accomplished MALDI-TOF/TOF. Two recombinant variants were obtained expression in Escherichia coli. Specific IgE measurement ELISA skin tests for extract performed 12 patients...

Immunoglobulin (Ig) diversification occurs via somatic hypermutation (SHM) and class switch recombination (CSR), is initiated by activation‐induced deaminase (AID), which converts cytosine to uracil. Variable (V) region genes undergo SHM create amino acid substitutions that produce antibodies with higher affinity for antigen. The conversion of uracil in DNA promotes mutagenesis. Two distinct repair mechanisms regulate processing Ig genes. first involves base removal the glycosylase (UNG),...

In this study was evaluated median lethal concentration extracts obtained and hemolytic activity. The partial methanolic extract exhibited few toxicity (CL50 >1000 µg/mL) smaller activity (CE50> 1000 µg/mL), comparative other extracts. That suggests according to these results can be pharmacologic utilities.
