BackgroundChronic exposure to ultraviolet (UV) radiation (mainly UVA) induces a sustained increase of matrix metalloproteinases (MMPs), especially MMP1, MMP2, MMP3 and MMP9 in human skin fibroblasts. MMPs can lead the degradation fibrous connective tissue, which is main cause photoageing. The molecular mechanisms through fibroblasts sense UVA trigger cell signalling pathways involved upregulation have not been well elucidated.
We previously reported the presence of microtubule-associated protein, tau in nuclei primate cells culture. The present study confirms existence nuclear two human neuroblastoma lines by indirect immunofluorescence and Western blot using mAbs to tau. Northern analysis poly A+ mRNA detects a novel 2-kb transcript coexpressed with 6-kb message cultured frontal cortex. PCR cDNA sequencing demonstrate that contains entire coding region. Furthermore, actinomycin D transcription inhibition...
Many rhodopsin-like G-protein-coupled receptors (Rh-GPCRs) are known to either promote or inhibit angiogenesis. Among these, Opsin 4 and 5 specifically involved in vascular development within the eye. 3 (OPN3), another member of Rh-GPCRs, performs a variety light-dependent light-independent functions extraocular tissue. However, its role endothelial cells angiogenesis remains unclear. Here, we found that OPN3 knockdown knockout zebrafish impairs embryonic development. Similarly, silencing...
Urticaria is a common skin disease characterized by episodes of wheals, and it has negative effect on patients' quality life. Large-scale population-based epidemiological studies urticaria are scarce in China. The aim this survey was to determine the prevalence, clinical forms, risk factors Chinese population. This conducted 35 cities from 31 provinces, autonomous regions, municipalities Two three communities each city were selected investigation. Participants completed questionnaires...
Abstract G protein‐coupled receptors (GPCRs) are core switches connecting excellular survival or death signals with cellular signaling pathways in a context‐dependent manner. Opsin 3 (OPN3) belongs to the GPCR superfamily. However, whether OPN3 can control of human melanocytes is not known. Here, we try investigate inherent function on melanocytes. Our results demonstrate that knockdown by RNAi‐OPN3 epidermal leads cell apoptosis. The downregulation markedly reduces intracellular calcium...
Background: The most common type of glomerulonephritis in China is mesangial proliferative (MPGN) featured with cell overproliferation and inflammation, as well fibrosis. Albiflorin (AF) an effective composition extracted from Paeonia Alba Radix has been administrated for various diseases. Nevertheless, there no research reporting the effect AF on MPGN.Purpose: Our work aims to probe into role possible mechanism MPGN.Research Design: We investigated effects overproliferation, fibrosis vitro...
Solar ultraviolet (UV) radiation-induced DNA damage is a major risk factor for skin cancer development. UV-induced redistribution of melanin near keratinocyte nuclei leads to the formation supranuclear cap, which acts as natural sunscreen and protects by absorbing scattering UV radiation. However, mechanism underlying intracellular movement in nuclear capping poorly understood. In this study, we found that OPN3 an important photoreceptor human epidermal keratinocytes critical UVA-mediated...
Retinal G protein-coupled receptor (RGR), a photosensitive protein, functions as retinal photoisomerase under light conditions in humans. Cutaneous squamous cell carcinoma (cSCC) is linked to chronic ultraviolet exposure, which suggests that the photoreceptor RGR may be associated with tumorigenesis and progression of (SCC). However, expression function remain uncharacterized SCC. This study analysed normal skin lesions actinic keratosis, Bowen's disease invasive SCC respect initiation...
Recessive mutations in RLBP1 cause a form of retinitis pigmentosa which the retina, before its degeneration leads to blindness, abnormally slowly recovers sensitivity after exposure light. To develop potential gene therapy for this condition, we tested multiple recombinant adeno-associated vectors (rAAVs) composed different promoters, capsid serotypes, and genome conformations. We generated rAAVs sequences from promoters human RLBP1, RPE65, or BEST1 genes drove expression reporter (green...
Background: This study aims to investigate the optimal surgical techniques in reduction malarplasty by comparing difference between L-shaped osteotomy with vertical and oblique bone resection. Methods: One hundred twenty patients who visited our department for either (Group Ⅰ) or resection Ⅱ) from 2015 2021 were retrospectively reviewed. The preoperative postoperative spiral computed tomography (CT) data analyzed. CT was also used simulate virtual model surgery. Results: results showed that...
Alzheimer's disease (AD) is a common neurological with recognition ability loss symptoms and major contributor to dementia cases worldwide.
The role of radiation therapy in addition to chemotherapy has not been well established non-oligometastatic Stage IV non-small cell lung cancer (NSCLC). We aimed investigate overall survival (OS) NSCLC treated with concurrent the primary tumor. Eligible patients were screened from two prospective studies. Oligometastatic and defined as having < 5 ≥5 metastatic lesions, respectively. Prognostic factors for OS identified by using univariate multivariate analysis. Landmark analysis...
Seborrhoeic keratosis (SK) is an age-related skin disease. Amyloid precursor protein (APP) plays important role in the pathogenesis of Alzheimer's The aim this study was to elucidate expression characteristics APP SK tissues (n = 50), and explore whether production related onset ageing, including ultraviolet (UV)-induced as observed normal 79). results immunohistochemistry, Western blotting quantitative real-time PCR showed that its downstream products (i.e. amyloid-β42) were more highly...
Background: Photoreceptive proteins play critical physiological roles in human skin cells. The retinal G protein-coupled receptor (RGR) is a photoisomerase the retina, but its expression and cellular functions cells have not been reported. Objectives: We aimed to detect RGR various evaluate regulation of keratinocytes. Methods: expression, distribution, subcellular location normal epidermal keratinocytes with pathological conditions including psoriasis, seborrheic keratosis, squamous cell...
The occurrence of type 2 diabetes mellitus (T2DM), a worldwide chronic disease, is mainly caused by insufficient insulin production and places huge burden on the health system. Gastrodia elata Blume (GE), food medicine–food homology, has been reported to have ability inhibit glycosidase activity, indicating its potential in treatment diabetes. However, main pharmacological components GE for T2DM not fully clarified. Therefore, this study aims clarify changes with different drying methods...
Abstract Cyclin-dependent kinase 4/6 (CDK4/6) acts as a crucial point of regulation in the G1-to-S transition cell division cycle, its aberrant activation was found various human cancers, leading to abnormal proliferation. Recent clinical trials have reported that combined with other small-molecule targeted therapies, CDK4/6 inhibitors increase overall survival and objective response rates breast cancer (BC), non-small lung (NSCLC), head neck squamous carcinoma (HNSCC). Notably, targeting...
