ConspectusCancer vaccines provide an efficient strategy to enhance tumor-specific immune responses by redeploying systems. Despite the approval of first cancer vaccine (Sipuleucel-T) U.S. Food and Drug Administration in 2010, most therapeutic fail clinical trials. Basically, rely on not only T-cell but also B-cell immunity, which indicates that should leverage both arms adaptive system. For example, CD8+ T cells activated antigen-presenting (APCs) recognize directly kill tumor via...

Constructing an effective therapeutic cancer vaccine is very attractive and promising for immunotherapy. However, the poor immunogenicity of tumor antigens suppression immune system in microenvironment are two major obstacles developing vaccines. Invariant NKT cells (iNKT cells), which essential bridges between innate adaptive systems, can be rapidly activated by their agonists and, consequently, evoke whole systems. Herein, we conjugated a potent agonist iNKT cell, α-galactosylceramide...

The protocol of aerobic oxidative dehydroxycyclization installed in the synthesis rarely studied 1-hydroxyphenothiazines from catechols and o-mercaptoanilines is presented. Utilizing a natural renewable low-toxicity gallic acid as an organocatalyst, this established transformation proceeded smoothly aqueous ethanol solution under mild conditions with good functional group compatibility up to 94% isolated yield. This also characterized by its operational simple workup involving only...

Cyclic dinucleotides (CDNs), agonists of stimulator interferon genes (STING), are promising agents for immunotherapy. However, the application CDNs has been limited by their instability and low transmembrane efficiency. Here, we introduced a conjugated adjuvant STING TLR1/2, Pam3CSK4-CDGSF. Conjugating CDGSF with Pam3CSK4 increased stability intracellular delivery. In addition, synergistically activating TLR pathways, Pam3CSK4-CDGSF was able to enhance immune activation. Both humoral...

Glycosylation of amyloidogenic proteins enhances their solubility and reduces propensity for aggregation. We therefore, prepared tryptophan-glucosamine conjugates to modulate aggregation tau-derived PHF6-peptide. Combined in vitro silico approaches indicated that these inhibited oligomerization fibril formation PHF6 disrupted its preformed fibrils at very low concentration. These effects mainly arise from the glucopyranoside moiety.
