Abstract The molecular connections between homeostatic systems that maintain both genome integrity and proteostasis are poorly understood. Here we identify the selective activation of unfolded protein response transducer IRE1α under genotoxic stress to modulate repair programs sustain cell survival. DNA damage engages signaling in absence an endoplasmic reticulum (ER) signature, leading exclusive regulated IRE1α-dependent decay (RIDD) without activating its canonical output mediated by...

Four alkaloid constituents: harmine, vasicine, vasicinone and harmaline were isolated characterized from Peganum harmala roots. Harmine showed the best cytotoxicity with IC50 of 32, 106, 45, 33 61 μM against breast (HBL-100), lung (A549), colon (HT-29 & HCT-116) cervical (HELA) cells respectively. The harmine analogs, synthesized via click chemistry, revealed that few derivatives possess potent cytotoxic effects. m-Fluoro phenyl-1, 2,3-triazolyl was found to be selectively human cancer cell...

HCC is the most frequent primary liver cancer with an extremely poor prognosis and often develops on preset of chronic diseases. Major risk factors for include metabolic dysfunction–associated steatohepatitis, a complex multifactorial condition associated abnormal endoplasmic reticulum (ER) proteostasis. To cope ER stress, unfolded protein response engages adaptive reactions to restore secretory capacity cell. Recent advances revealed that stress signaling plays critical role in progression....

Abstract Collagen is one the most abundant proteins and main cargo of secretory pathway, contributing to hepatic fibrosis cirrhosis due excessive deposition extracellular matrix. Here we investigated possible contribution unfolded protein response, adaptive pathway that monitors adjusts production capacity at endoplasmic reticulum, collagen biogenesis liver disease. Genetic ablation ER stress sensor IRE1 reduced damage diminished in models triggered by carbon tetrachloride (CCl 4 )...

Abstract Alpha1-antitrypsin (α1AT) is an abundant serine-protease inhibitor in circulation. It has important role neutralizing the neutrophil elastase activity. Different pathogenic point mutations like Z (E342K) -α1AT have been implicated development of liver cirrhosis and Chronic Obstructive Pulmonary Disease (COPD), latter being a cluster progressive lung diseases including chronic bronchitis emphysema. M3-α1AT (376Glu > Asp) another variant α1AT which so far largely considered as...
