After isoniazid and rifampin (rifampicin), the next pivotal drug class in Mycobacterium tuberculosis treatment is fluoroquinolone class. Mutations resistance-determining regions (RDR) of rpoB, katG, gyrA genes occur with frequencies 97%, 50%, 85% among M. isolates resistant to rifampin, isoniazid, fluoroquinolones, respectively. Sequences are highly conserved, certain mutations correlate well phenotypic resistance. We developed a pyrosequencing assay determine genotypic resistance...

Setting The 3 rd national tuberculosis (TB) survey in the Philippines 2007 reported a significant decline prevalence of TB. Since then, more investments for TB control have been made, yet burden estimates from routine surveillance data remain relatively stable. Objective To estimate bacteriologically confirmed pulmonary amongst individuals aged ≥15 years 2016. Design In March–December 2016, we conducted population-based with stratified, multi-stage cluster sampling residents 106 clusters...

The mycobacterial antigen MPB-64 was formulated for delivery in a transdermal patch and used as diagnostic skin test reagent to detect active tuberculosis (TB) patients attending clinic Manila, Philippines. Transdermal Patch applied 62 patients, 49 with sputum-positive disease 13 who had completed TB chemotherapy, 28 non-TB but tuberculin-positive controls. results were read at 72 h. sensitivity of the 87.8%, an efficacy 92.9% specificity 100%. 6 months chemotherapy showed different...

Background:Pulmonary tuberculosis (PTB) and paragonimiasis remain as health problems in certain areas the Philippines. Both share similar clinical manifestations, which include chronic productive cough, hemoptysis, dyspnea, fever, weight loss, night sweats. This study aimed to determine prevalence of PTB, paragonimiasis, co-infections Zamboanga del Norte, Philippines. Methods:This was conducted selected villages two municipalities Norte. Patients with cough were interviewed, examined,...

Abstract The Philippines is a high-incidence country for tuberculosis, with the increasing prevalence of multi- (MDR-TB) and extensively-drug (XDR-TB) resistant Mycobacterium tuberculosis strains posing difficulties to disease control. Understanding genetic diversity circulating can provide insights into underlying drug resistance mutations transmission dynamics, thereby assisting design diagnostic tools, including those using next generation sequencing (NGS) platforms. By analysing genome...

An MPT64 antigen detection assay, the SD Bioline TB Ag Rapid kit, was evaluated for identification of Mycobacterium tuberculosis grown in both solid and liquid media. The rapid test showed positive results 132 133 (99.2%) M. cultures on Ogawa medium 136/143 (95.1%) culture medium, resulting an overall sensitivity 97.1%. All 18 non-tuberculous mycobacteria were found to be negative by test, indicating a specificity 100%. seems useful initial confirmation acidfast bacilli cultures.

Background: Tuberculosis remains a major public health threat. Studies have shown that medium chain fatty acids (MCFA), such as, those found in Virgin Coconut Oil (VCO), possess activity against wide range of microorganisms including Mycobacterium tuberculosis. The goal the study was to determine in-vitro susceptibility M. tuberculosis (TB) isolates two commercial brands VCOs each containing different amounts lauric acid and produced by extraction processes. Two VCO were tested for their...

Thousands of cases multidrug-resistant tuberculosis (TB) have been observed in the Philippines, but studies on Mycobacterium (MTB) genotypes that underlie drug resistance profiles are lacking. This study aimed to analyse whole genomes clinical MTB isolates representing various identify single nucleotide polymorphisms (SNPs) resistance-associated genes. The ten cultured from banked sputum sources were sequenced. Bioinformatics analysis consisted assembly, annotation and SNP identification...

Tuberculosis, caused by Mycobacterium tuberculosis, remains a high burden disease and leading cause of mortality in the Philippines. Understanding genetic diversity M. tuberculosis strains population, including those that are multi-drug resistant (MDR), will aid formulating strategies for effective TB control prevention. By whole genome sequencing isolates (n = 100) from patients Philippine 2016 National Tuberculosis Prevalence Survey, we sought to provide baseline assessment genotypic...

Tuberculosis (TB) remains an important public health problem in developing countries like the Philippines. The success of National TB Control Program depends on a clear understanding dynamics transmission and spread high-risk populations community. ABSTRACT In the Philippines, acid fast bacilli positive sputum samples commonly treated as TB due to Mycobacterium tuberculosis (MTB) complex. However, abscessus (MAB) complex is often found in MTB cultures, or patients confirmed negative for through microscopy and culture. Hence, with MAB infections are mistakenly prescribed six-month anti-TB treatments. this study, isolates from MDRTB were identified further sub-speciated using mass 3210 gene. Antimicrobial susceptibility was tested broth...

BACKGROUND: TB is the leading cause of death from a single infectious disease, particularly among people living with HIV (PLHIV). Molecular epidemiology provides information on prevalent genotypes Mycobacterium tuberculosis and disease transmission dynamics, which aid in control. Identification mutations that confer drug resistance essential for rapid diagnosis drug-resistant TB, especially high burden settings, like Philippines. METHODS: This study aimed to determine M....

Background: Despite the high tuberculosis (TB) burden in Philippines, data on genetic diversity and transmission dynamics of M. remains sparse. This study aims to determine prevalent genotype isolates obtained from a rural community factors associated with clustering. Methods: 125 culture-positive symptomatic Filipino patients aged 15 above San Juan, Batangas were genotyped using combined spoligotyping 15-loci MIRU-VNTR. Clustering rate was used as an index for recent transmission. Results:...
