Molecular glue degraders induce "undruggable" protein degradation by a proximity-induced effect. Inspired the clinical success of immunomodulatory drugs, we aimed to design novel molecular targeting GSPT1. Here, report series GSPT1 degraders. LYG-409, 2H-chromene derivative, was identified as potent, selective, and orally bioavailable degrader with excellent antitumor activity in vivo (anti-Acute Myeloid Leukemia MV4–11 xenograft model: TGI = 94.34% at 30 mg/kg; prostate cancer 22Rv1 104.49%...
The thioredoxin system plays an important role in cancer cells. Inhibiting reductase (TrxR) has emerged as effective strategy to selectively target Withangulatin A (WA), a natural product extracted from the whole herb of Physalis angulata L. (Solanaceae), exhibits potent anticancer activity and other diverse pharmacological activities. To improve targeting, we designed prepared 41 semisynthetic analogues WA. Biological evaluation indicated that most promising compound 13a displayed...
Background . Oxidative stress has a vital role in the early stages of vitiligo. Autoantigens released from apoptotic melanocytes (MC) under oxidative are involved presentation and recognition antigens. However, transport autoantigens to cell surface their release extracellular environment still unclear. Apoptotic bodies (ABs) have always been considered as key source immunomodulators autoantigens. Yet, ABs immune mechanism vitiligo is unknown. Purpose To explore whether MC’s translocate into...
Six compounds were synthesized from the lanostane-type tetracyclic triterpenoid of trametenolic acid B with aim obtaining more active compounds, which characterized by NMR, IR, ESI-MS and elemental analysis. Their cytotoxic activities evaluated against breast cancer cell line MD-AMB-231. The compound 3b exhibited best anti-proliferative effect than other derivatives IC50 17.78 µM.
The trametenolic acid B was a bioactive lanostane-type triterpenoid in Trametes lactinea (Berk.) Pat with. Five novel derivatives were synthesized, characterized and evaluated the cytotoxicity against MDA-MB-231 cells with aim of obtaining better anti-tumtor agents. compound IV exhibited significantly activities IC 50 value 22.39 μM.
Discovery of bioactive ingredients from plants and fungi is always the hot spots in medicinal chemistry. The trametenolic acid B was a lanostane-type triterpenoid Trametes lactinea (Berk.) Pat. In this work, four semi-synthetic derivatives were synthesized, characterized evaluated anti-gastric cancer activities against HGC-27 cells with aim obtaining better anti-tumor agents. compounds 2a 3b possessed good anti-proliferative effects under normal physiological conditions, their anti-cancer...
