The sustainable production of chemically recyclable polymers presents a significant opportunity to polymer scientists tackle the growing environmental and energy problems current petroleum-based plastics. Despite recent advances, however, there are still pressing needs for an expanded horizon polymers. Herein, we introduce new paradigm biosourced polythioesters (PTEs) with high polymerizability complete recyclability under mild economical conditions. thiolactone monomers ring strain can be...
The rapid and controlled synthesis of high-molecular-weight (HMW) polysarcosine (pSar), a potential polyethylene glycol (PEG) alternative, via the ring-opening polymerization (ROP) N-carboxyanhydride (NCA) is rare challenging. Here, we report well-controlled ROP sarcosine NCA (Sar-NCA) that catalyzed by various carboxylic acids, which accelerate rate up to 50 times, enables robust pSar with an unprecedented ultrahigh molecular weight (UHMW) 586 kDa (DP ∼ 8200) exceptionally narrow dispersity...
Ring-opening polymerizations of α-amino acid N-carboxyanhydrides (NCAs) initiated with amines typically form polypeptides uncontrolled molecular weights and broad weight distributions. However, we found that hexamethyldisilazane (HMDS)-mediated controlled NCA gave predictable narrow Using MS, NMR, FT-IR, demonstrated the initiation step involved cleavage N−Si bond HMDS formation a trimethylsilyl carbamate (TMS-CBM) terminal group. Polypeptide chains were propagated through migration TMS...
At every turn: Poly(γ-(4-vinylbenzyl)-L-glutamate) (PVBLG) is a bioactive and reactive template for library of cationic α-helical polypeptides (PVBLGn-X) gene delivery. The top-performing PVBLGn-X outperformed polyethyleneimine (PEI) by 12-fold. Preliminary data indicate that the helicity essential their performance, enhanced membrane disruption likely source transfection efficiency. RLU=relative light unit.
We report a new strategy that uses N-trimethylsilyl (N-TMS) amine to mediate controlled ring-opening polymerization of amino acid N-carboxyanhydrides (NCAs). This proceeds via unique, trimethylsilyl carbamate (TMS-CBM) propagating group results from the cleavage Si−N bond N-TMS followed by NCA ring opening. Propagation polypeptide chains through transfer TMS terminal TMS-CBM incoming monomer formation group. Formation was confirmed MS and NMR. Polypeptides formed amine-mediated have expected...
Gene delivery vehicles: Helical, cationic polypeptides with light-responsive domains showed potent membrane activity and promoted efficient cellular internalization of DNA (see picture). After transfection, a light trigger induces protecting-group removal to expose anionic carboxylate groups. The reduced charge helix distortion the result in enhanced unpacking material toxicity because eliminated activity.
The performance of many therapeutic proteins, including human interferon-α2b (IFN), is often impeded by their intrinsic instability to protease, poor pharmacokinetics, and strong immunity. Although PEGylation has been an effective approach improve the pharmacokinetics a few noticeable limitations have aroused vast research efforts in seeking alternatives PEG for bioconjugation. Herein, we report our investigation on use polysarcosine (PSar), nonionic hydrophilic polypeptoid, IFN...
Grafting-from (GF) is an important yet underdeveloped strategy toward protein–polymer conjugates. Here, we report a simple cryopolymerization method that enables highly efficient GF synthesis of cell-penetrating protein–polydisulfide Rapid and controlled ring-opening polymerization 1,2-dithiolanes under cryo-conditions can be initiated by proteins bearing reactive cysteine, owing to both favored thermodynamics augmented kinetics arising from frozen-induced high local concentration...
Various polymers have been tested for protein conjugation with a goal of bridging the complementary advantages both components. However, many these polymers, including most well-established PEG, are nondegradable, which raises potential concerns on their cumulative chronic toxicity. Moreover, immunogenicity PEG has recently evoked considerable controversy. Synthetic polypeptides, other hand, biomimetic tunable degradability, versatile side chain functionalities, unique secondary structures,...
Abstract A great hurdle in the production of synthetic polypeptides lies access N -carboxyanhydrides (NCA) monomers, which requires dry solvents, Schlenk line/gloveboxe, and protection side-chain functional groups. Here we report a robust method for preparing unprotected NCA monomers air under moisture. The employs epoxy compounds as ultra-fast scavengers hydrogen chloride to allow assisted ring-closure prevent from acid-catalyzed decomposition moist conditions. broad scope group tolerance...
Abstract Effective drug delivery systems that can systematically and selectively transport payloads to disease cells remain a challenge. Here, targeting ligand‐modified DNA origami nanostructure (DON) as an antibody–drug conjugate (ADC)‐like carrier for targeted prostate cancer therapy is reported. Specifically, DON of six helical bundles modified with ligand 2‐[3‐(1,3‐dicarboxy propyl)‐ureido] pentanedioic acid (DUPA) against prostate‐specific membrane antigen (PSMA), serve the antibody...

Osteochondral injury is a common and frequent orthopedic disease that can lead to more serious degenerative joint disease. Tissue engineering promising modality for osteochondral repair, but the implanted scaffolds are often immunogenic induce unwanted foreign body reaction (FBR). Here, we prepare polypept(o)ide-based PAA-RGD hydrogel using novel thiol/thioester dual-functionalized hyperbranched polypeptide P(EG3Glu-co-Cys) maleimide-functionalized polysarcosine under biologically benign...
The production of polypeptides via the ring-opening polymerization (ROP) N-carboxyanhydride (NCA) is usually conducted under stringent anhydrous conditions. ROP proline NCA (ProNCA) for synthesis poly-L-proline (PLP) particularly challenging due to premature product precipitation as polyproline type I helices, leading slow reactions up one week, poor control molar mass and laborious workup. Here, we report unexpected water-assisted controlled ProNCA, which affords well-defined PLP II helices...

We report here the integration of ring-opening metathesis polymerization (ROMP) and amino acid N-carboxyanhydride (NCA) to allow facile synthesis brush-like polymers containing polypeptide as brush side chains. ROMP N-trimethylsilyl norbornenes rendered preparation poly(norbornene)s bearing pendant N-TMS groups. With no need purify resulting polymers, such macromolecular initiators could subsequently initiate controlled NCA polymerizations. Brush-like with grafted polypeptides or block...
We describe here the development of site-specific antibody-polymer conjugates (APCs) for selective delivery small interference RNAs (siRNAs) to target cells. APCs were synthesized in good yields by conjugating an aminooxy-derivatized cationic block copolymer anti-HER2 Fab or full-length IgG means genetically encoded p-acetyl phenylalanine (pAcF). The all showed binding affinity comparable that HER2 as their native counterparts and no significant cellular cytotoxicity. Mutant S202-pAcF...
