Abstract Recent studies have reported increased podoplanin expression by cancer cells and stromal cells, but little is known about its biological significance in adenocarcinoma of the lung. We examined both 177 consecutive lung cases analyzed relations between clinicopathological factors outcome. Podoplanin was observed on apical membrane only 9 (5.1%) cases. By contrast, cancer‐associated fibroblasts (CAFs) were found to express 54 (30.5%). (+) CAFs invasive none noninvasive adenocarcinoma....

Abstract Purpose: Although large-cell neuroendocrine carcinoma (LCNEC) of the lung shares many clinical characteristics with small-cell cancer (SCLC), little is known about its molecular features. We analyzed LCNECs to identify biologically relevant genomic alterations. Experimental Design: performed targeted capture sequencing all coding exons 244 cancer-related genes on 78 LCNEC samples [65 surgically resected cases, including 10 combined non–small cell (NSCLC) types separately, and...

Thymic carcinoma is a rare thymic malignancy. The purpose of this study was to evaluate the prognostic impact clinicopathological variables and perioperative therapy for surgically treated using nationwide database. Of 2835 patients with epithelial tumours collected from 32 Japanese institutions, total 306 carcinomas, excluding neuroendocrine tumours, were enrolled in retrospective study. Multivariable Cox regression analyses performed overall (OS) recurrence-free survival (RFS) after R0...

ObjectivesThe purpose of this study was to assess the surgical outcomes according extent mediastinal lymph node dissection for patients with NSCLC by using a nationwide registry database.MethodsFrom among 11,663 in Japanese lung cancer 2004, 5392 clinical stage (c-stage) I or II that completely resected lobectomy and either systematic (SND) lobe-specific nodal (LSD) were enrolled. Patients who received preoperative therapy had middle lobe tumor excluded. In LSD group, inferior (subcarinal)...

Human lung cancer is a constellation of tumors with various histological and molecular properties. To build preclinical platform that covers this broad disease spectrum, we obtained specimens from multiple sources, including sputum circulating tumor cells, generated living biobank consisting 43 lines patient-derived organoids. The organoids recapitulated the hallmarks original tumors. Phenotypic screening niche factor dependency revealed EGFR mutations in adenocarcinoma are associated...

We analyzed pulmonary squamous cell carcinoma and adenocarcinoma patient survival in our single institution database, to evaluate the relationship of histologic analysis tumor aggressiveness. We reviewed 1856 consecutive patients with surgically resected or regarding their clinicopathologic characteristics, overall recurrence-free proportion. In patients, there were more elderly male smokers T2-4 tumors, moderately/poorly differentiated lymph node metastasis vascular invasion than patients. In...

Recently, an association between tumor infiltrating Forkhead box P3 regulatory T cells (T reg ) and unfavorable prognosis has been clinically shown in some cancers, but the mechanism of induction microenvironment remains uncertain. In January 2017, the Lung Cancer Surgical Study Group of Japan Clinical Oncology commenced a randomized Phase III trial to confirm clinical benefit lobe-specific nodal dissection for Stage I-II non-small cell lung cancer. The primary endpoint is overall survival, and main objective non-inferiority in comparison systematic with regard lobectomy. secondary endpoints are relapse-free %local recurrence, %regional lymph node operation time, blood loss, length hospitalization, duration chest tube...

Although the main cellular target of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is thought to be alveolar cells, absence their tractable culture system precludes development a clinically relevant SARS-CoV-2 model. Here, we establish an efficient human alveolosphere method and sphere-based drug testing platform for SARS-CoV-2. Alveolospheres exhibit indolent growth in Wnt- R-spondin-dependent manner. Gene expression, immunofluorescence, electron microscopy analyses...

Recent molecular biological studies have identified podoplanin as a candidate cancer stem cell (CSC) marker in squamous carcinoma (SqCC). The purpose of this study was to examine the expression pattern podoplanin, and other markers CD44 p63, their relationship clinico-pathological features including survival pulmonary SqCC. We examined histologically CD44, p63 162 consecutive SqCC by immunostaining. Podoplanin observed 107 (66%) tumors, 145 (89.5%), 151 (93.2%), respectively. In 95.3%...

Abstract Lung adenocarcinoma driven by somatic EGFR mutations is more prevalent in East Asians (30–50%) than European/Americans (10–20%). Here we investigate genetic factors underlying the risk of this disease conducting a genome-wide association study, followed two validation studies, 3,173 Japanese patients with mutation-positive lung and 15,158 controls.
