The shift from solitary to social behavior is one of the major evolutionary transitions. Primitively eusocial bumblebees are uniquely placed illuminate evolution highly insect societies. Bumblebees also invaluable natural and agricultural pollinators, there widespread concern over recent population declines in some species. High-quality genomic data will inform key aspects bumblebee biology, including susceptibility implicated viability threats. We report high quality draft genome sequences...
Abstract Rare diseases comprise a diverse group of conditions, most which involve genetic causes. We describe the variable spectrum findings and clinical impacts exome sequencing (ES) in cohort 500 patients with rare diseases. In total, 164 primary were reported 158 patients, representing an overall diagnostic yield 31.6%. Most (61.6%) corresponded to autosomal dominant followed by recessive (25.6%) X‐linked (12.8%) conditions. These harbored 195 variants, among 43.6% are novel literature....
Deregulation of the IGF system observed in human tumors indicates a role malignant cell transformation and tumor proliferation. Although overexpression IGF2 IGF1R genes was described adrenocortical (ACTs), few studies reported their profiles pediatric ACTs. In this study, expression evaluated by RT-qPCR according to patient’s clinical/pathological features 60 ACT samples, protein investigated 45 samples immunohistochemistry (IHC). Whole transcriptome functional assays were conducted after...
Misexpression of stem cell-related genes may occur in some cancer cells, influencing patient's prognosis. This is the case medulloblastoma, a common and clinically challenging malignant tumor central nervous system, where expression pluripotency factor, OCT4, correlated with poor survival. A downstream target L1TD1 (LINE-1 type transposase domain-containing protein 1 family member), encodes novel embryonic cell (ESC)-related involved self-renewal ESCs. still poorly characterized its pattern...
Adrenocortical tumor (ACT) is a malignancy with low incidence rate and the current therapy for advanced disease has limited impact on overall patient survival. A previous study from our group suggested that elevated expression of aurora-A aurora-B associated poor outcome in childhood ACT. Similar results were also reported adult ACTs. The present in-vitro shows AMG 900 inhibits aurora kinases adrenocortical carcinoma cells. inhibited cell proliferation NCI-H295 cells as well ACT primary...
Breast cancer is the most common among women worldwide, and ovarian difficult gynecological tumor to diagnose with lowest chance of cure. Mutations in BRCA1 BRCA2 genes increase risk by 60% breast up 80% women. Molecular tests allow a better orientation for patients carrying these mutations, affecting prophylaxis, treatment, genetic counseling. Here, we evaluated performance panel BRCA2, using Ion Torrent PGM (Life Technologies) platform customized workflow multiplex ligation-dependent probe...
ABSTRACT Eye development in insects is best understood Drosophila melanogaster , but little known for other holometabolous insects. Combining a morphological with gene expression analysis, we investigated eye the honeybee, putting emphasis on sex‐specific differences size. Optic lobe starts from an optic anlage larval brain, which sequentially gives rise to lobula, medulla, and lamina. The lamina differentiates last instar, when it receives nerve projections developing retina. analysis...
Our aim was to develop and apply a comprehensive noninvasive prenatal test (NIPT) by using high-coverage targeted next-generation sequencing estimate fetal fraction, determine sex, detect trisomy monogenic disease without parental genotype information. We analyzed 45 pregnancies, 40 mock samples, eight mother-child pairs generate 35 simulated datasets. Fetal fraction (FF) estimated based on analysis of the single nucleotide polymorphism (SNP) allele distribution. A Z-score calculated for...
Congenital hypogonadotrophic hypogonadism (CHH) is a challenging inherited endocrine disorder characterised by absent or incomplete pubertal development and infertility as result of the low action/secretion hypothalamic gonadotrophin-releasing hormone (GnRH). Given growing list gene mutations accounting for CHH, application massively parallel sequencing comprises an excellent molecular diagnostic approach because it enables simultaneous evaluation many genes. The present study proposes use...
During mating, insect males eject accessory gland proteins (Acps) into the female genital tract. These substances are known to affect post-mating behavior and physiology. In addition, they may harm female, e.g., in reducing its lifespan. This is interpreted as a consequence of sexual antagonistic co-evolution. Whereas conflict abounds non-social species, peculiar life history social insects (ants, bees, wasps) with lifelong pair-bonding no re-mating aligns reproductive interests sexes....
Abstract Pediatric adrenocortical tumors (ACT) are rare aggressive neoplasms with heterogeneous prognosis. Despite extensive efforts, identifying reliable prognostic factors for pediatric patients ACT remains a challenge. MicroRNA (miRNA) signatures have been associated cancer diagnosis, treatment response, and prognosis of several types cancer. However, the role miRNAs has poorly explored in ACT. In this study, we performed miRNA microarray profiling on cohort 37 nine nonneoplastic adrenal...
Background: MicroRNAs (miRNA) are short non-coding RNA that act as negative regulators of gene expression. Altered levels miR-708-5p have recently been described in many tumors, although its contribution colorectal cancer (CRC) pathophysiology remains unclear. Methods/Patients: Quantitative real-time polymerase chain reaction was employed to evaluate the expression 50 CRC and 20 paired adjacent noncancerous tissues. The relationship between miRNA clinicopathological features estimated using...
Abstract Background : Medulloblastoma, a genetically heterogeneous tumor, is the most frequent malignant brain tumor in children. Although several studies have been carried out, molecular mechanism underlying medulloblastoma tumorigenesis not completely known. microRNA (miRNA) expression profiles associated with development, progression, and prognosis of human cancers, including medulloblastoma. However, role miRNAs pediatric has poorly explored. Methods Global miRNA 24 microdissected...
Abstract Pediatric adrenocortical tumors (ACT) are rare malignancies and in advanced disease the treatment has a small impact on overall survival. Previous study from our group suggests that AURKA AURKB overexpression pediatric ACT may be related to more aggressive disease. These genes involved maintenance of genome integrity during cell cycle division they have been considered as new targets cancer treatment. The present shows previous results effects Aurora kinase inhibitor AMG900,...
