Approximately 20% of familial amyotrophic lateral sclerosis (FALS) arises from germ-line mutations in the superoxide dismutase-1 (SOD1) gene. However, molecular mechanisms underlying process have been elusive. Here, we show that a neuronal homologous to E6AP carboxyl terminus (HECT)-type ubiquitin-protein isopeptide ligase (NEDL1) physically binds translocon-associated protein-δ and also ubiquitinates mutant (but not wild-type) SOD1 proportionately disease severity caused by particular...
Abstract Purpose: A positive response to gefitinib in non–small cell lung cancer (NSCLC) has been correlated mutations epidermal growth factor receptor (EGFR) gene. Previous reports have based mainly on diagnostic screening by sequencing. However, sequencing is a time-consuming and complicated procedure, not suitable for routine clinical use. Experimental Design: We developed rapid, simple, sensitive mutation detection assays the SMart Amplification Process (SMAP) applied it analyzing EGFR...

Abstract Neuroblastoma (NBL), one of the most common childhood solid tumors, has a distinct nature in different prognostic subgroups. However, precise mechanism underlying this phenomenon remains largely unknown. To understand molecular and genetic bases neuroblastoma, we have generated its cDNA libraries identified human ortholog tubulin tyrosine ligase gene ( hTTL / Nbla0660 ) as differentially expressed at high levels favorable subset tumor. Tubulin is subjected to several types...
Insights into mechanisms of drug resistance could extend the efficacy cancer therapy. To probe in melanoma, we performed siRNA screening genes that mediate development neural crest cells, from which melanocytes are derived. Here, report identification ZIC5 as a mediator melanoma resistance. is transcriptional suppressor E-cadherin expressed highly human melanoma. enhanced cell proliferation, survival, resistance, vivo growth and metastasis. Microarray analysis revealed downstream signaling...
Sequential deletion of the carboxyl-terminal amino acids (including six direct repeating units) glucosyltransferase-I (GTF-I) enzyme Streptococcus mutans revealed differential effects on sucrase and GTF activities. Removal all but one unit resulted in a truncated with significant activity no detectable activity. These results are compatible presence two functional domains enzyme.
Identification of specific drug targets is very important for cancer therapy. We recently identified zinc finger protein the cerebellum 5 ( ZIC 5) as a factor that promotes melanoma aggressiveness by platelet‐derived growth D PDGFD ) expression. However, its roles in other types remain largely unknown. Here we determined prostate PC a) and colorectal CRC cells. Results showed was highly expressed dedifferentiated tissues, whereas little expression observed relevant normal tissues. Knockdown...
DNA regions corresponding to portions of two different pressure-regulated operons previously identified in deep-sea barophilic bacteria were separately PCR amplified from a variety microorganisms and sequenced. With the sets primers employed, amplification was particularly successful more examined. 16S rRNA sequence analysis revealed that these are all phylogenetically related belong sub-branch genus Shewanella containing only bacteria. We define this as 'Shewanella barophile branch' at...
Abstract Although imatinib mesylate has shown encouraging activity in chronic myelogenous leukemia (CML), disease progression during therapy been observed, manifested by clonal expansion of mesylate‐resistant cells. On the other hand, myelosuppression related to treatment is often managed with temporary interruption or dose reduction. We here report two CML patients who had mesylate‐sensitive blast crisis (BC) immediately after discontinuation therapy. The discontinued because neutropenia....
A pressure-regulated operon was cloned and sequenced from a barophilic bacterium strain DB6705. Gene expression by this promote regulated at the level of transcription elevated pressure in strain. The promoter sequence similar to ompH Photobacterium sp. SS9.
A glutamine synthetase gene (glnA) was isolated from a deep-sea piezophilic bacterium, Shewanella violacea strain DSS12. 7.5-kb SacI fragment containing the complete glnA cloned and sequenced. The found to encode protein consisting of 469 amino acid residues, showing 75.0% identity Escherichia coli. Primer extension analyses revealed two transcription initiation sites in expression each site positively regulated by pressure. Putative promoters recognized σ70 σ54 were identified region...
The impact of co-infection with other pathogenic microorganisms after initiation treatment for Mycobacterium avium complex pulmonary disease (MAC-PD) has not been clearly described. This study sought to clarify the clinical outcomes MAC antimycobacterial therapy MAC.Co-infection status was defined as detection than in at least two consecutive sputum cultures 6-24 months treatment. Chest computed tomography (CT) findings and culture results were compared between alone groups. The groups...
A 75-year-old woman was referred to our hospital because of a productive cough and an abnormal shadow on chest radiography. She diagnosed as having metastatic lung adenocarcinoma harbouring ROS proto-oncogene 1 (
