An efficient synthesis of quinazolin-4(3H)-ones from N-arylamidines, through palladium-catalyzed intramolecular C(sp(2))-H carboxamidation, has been developed. The reaction, carried out in the presence 1.0 equiv CuO as oxidant under atmospheric pressure CO, provides diversified 2-aryl(alkyl)quinazolin-4(3H)-ones reasonable to good yields which are readily derived anilines and nitriles. Compared with existing approaches quinazolin-4(3H)-ones, current strategy features atom-economy step-efficiency.
VEGF-A activity is tightly regulated by ligand and receptor availability. Here we investigate the physiological function of heterodimers between VEGF receptor-1 (VEGFR-1; Flt-1) VEGFR-2 (KDR; Flk-1) (VEGFR1−2) in endothelial cells with a synthetic that binds specifically to VEGFR1−2. The dimeric comprises one VEGFR-2-specific monomer (VEGF-E) VEGFR-1-specific (PlGF-1). show VEGFR1−2 activation mediates VEGFR phosphorylation, cell migration, sustained vitro tube formation vasorelaxation via...
It is reported recently that legitimate libraries are repackaged for propagating malware. An in-depth analysis of such potentially-harmful (PhaLibs), however, has never been done before, due to the challenges in identifying those whose code can be unavailable online (e.g., removed from public repositories, spreading underground, etc.). Particularly, an iOS app, library it integrates cannot trivially recovered its binary and analyzed by any publicly available anti-virus (AV) systems. In this...
Insects and pathogens release effectors into plant cells to weaken the host defense or immune response. While imports of some bacterial fungal plants have been previously characterized, mechanisms how caterpillar enter remain a mystery. Using live cell imaging real-time protein tracking, we show that HARP1, an effector from oral secretions cotton bollworm (Helicoverpa armigera), enters via protein-mediated endocytosis. The entry HARP1 depends on its interaction with vesicle trafficking...
Patchoulol, a natural sesquiterpene compound, is widely used in perfumes and cosmetics. Several strategies were adopted to enhance patchoulol production Saccharomyces cerevisiae: (i) farnesyl pyrophosphate (FPP) synthase fused increase the utilization of FPP precursor; (ii) expression limiting genes mevalonate pathway was enhanced; (iii) squalene weakened by glucose-inducible promoter HXT1 (promoter for hexose transporter) reduce metabolic flux from ergosterol; (iv) farnesol biosynthesis...
This work highlights the use of push-pull hydroxylphenylpolyenylpyridinium fluorophores coupled with trimethyl lock quinone to engineer ratiometric two-photon probes for cellular and intravital imaging mitochondrial NAD(P)H:quinone oxidoreductase 1 (NQO1), a critical antioxidant enzyme responsible detoxifying quinones. As typical representative, QBMP showed favorable binding NQO1 Michaelis constant 12.74 μM exhibited suite superior properties, including rapid response (4 min), large Stokes...
Protein tyrosine O-sulfation (PTS) plays a crucial role in extracellular biomolecular interactions that dictate various cellular processes. It also involves the development of many human diseases. Regardless recent progress, our current understanding PTS is still its infancy. To promote and facilitate relevant studies, generally applicable method needed to enable efficient expression sulfoproteins with defined sulfation sites live mammalian cells. Here we report engineering, vitro...
Currently, red emitting materials with thermally activated delayed fluorescence (TADF) properties exhibit significant potential for application in organic light-emitting diodes (OLEDs), but limited by the energy gap law, impeding their...
Abstract The Weihe River Basin has the worst soil erosion in Loess Plateau. As largest tributary of Yellow River, water conservation capacity is great significance to and In this study, volume estimated based on Integrated Valuation Ecosystem Services Tradeoffs model, basin measured after excluding influence rainfall evapotranspiration. results show that from 1987 2019, showed an overall trend decreasing first then increasing. increasing greater than production. decreased lowest 2005...
Abstract In this study, we used branches Ramulus mori from cultivated mulberry Husang‐32 ( Morus multicaulis Perry) as the experimental material and anhydrous ethanol extraction solution to obtain crude extract branch bark. The ethanolic was successively purified through a macroporous resin, Sephadex LH‐20, semipreparative reversed‐phase high‐performance liquid chromatography (RP‐HPLC). high‐purity monomer identified morusin by HPLC with diode array detection (HPLC‐DAD) its UV spectrum....
Protein tyrosine phosphorylation plays a critical role in signal transduction. We report the genetic incorporation of phosphotyrosine (pTyr) analog, p-carboxymethyl-L-phenylalanine (CMF), into proteins mammalian cells. This nonhydrolyzable pTyr analog can facilitate biological studies by removing complications caused dynamic interconversion between phosphorylated and non-phosphorylated isoforms protein.
Abstract Transnitrosylation is an important mechanism by which nitric oxide (NO) modulates cell signaling pathways. For instance, SNO-caspase-3 can transnitrosylate the X-linked inhibitor of apoptosis (XIAP) to enhance apoptosis. XIAP a potent antagonist caspase apoptotic activity. Decrease in activity via nitrosylation results SNO-XIAP-mediated activation. Considering functional liaison procaspase-9 and XIAP, we hypothesized that nitrosylates directly. Our data confirmed cerebral...
Abstract The known 7,8‐secolabdane type diterpenoids neopallavicinin ( 1 ), pallavicinin 2 and 18‐hydroxypallavicinin 3 ) were isolated from Pallavicinia ambigua , their structures determined. X‐ray crystal structure of was solved, and, in combination with CD NMR studies, the absolute configurations – established. A possible biogenetic pathway for a single labdane precursor is proposed.
Abstract While amber suppression is the most common approach to introduce noncanonical amino acids into proteins in live cells, quadruplet codon decoding has potential enable a greatly expanded genetic code with up 256 new codons for protein biosynthesis. Since triplet are predominant form of nature, often displays limited efficiency. In this work, we exploited significantly improve UAGN and AGGN (N = A, U, G, C) efficiency by using recoding signals imbedded mRNA. With representative...
Abstract Selective functionalization of quinolines is the hot topic organic synthesis due to its wide application in natural products, medicine, etc. In this research, C5 bromination 8‐aminoquinoline amides have been achieved using strategy photoredox catalysis under mild conditions. Easily available 10‐phenylphenothiazine employed as organophotoredox catalyst and CBr 4 used bromine source. No external oxidant or transition metal needed transformation. Both alkyl aryl substituted...
Tree shelterbelts are crucial for maintaining the ecological environment of oasis, but they may also compete soil water with adjacent crops, affecting crop yields. To evaluate impacts shelterbelt on use efficiency (WUE) and normalized productivity (WP) cotton plants, biomass (B) WUE different distances from (0.1H, 0.5H, 1H, 2H, 3H; average tree height = 15 m [H]) were estimated based unmanned aerial vehicle (UAV) remote sensing data combined FAO response model AquaCrop. Besides, accuracy...
