We designed and constructed a genome-wide microarray with 22,987 70-mer oligonucleotides covering the presently known predicted genes in silkworm genome, surveyed gene expression multiple tissues on day 3 of fifth instar. Clusters tissue-prevalent tissue-specific that are differentially expressed different were identified, they reflect well major functions molecular level. The data presented this study provide new resource for annotating genome.

Abstract Background Chemosensory systems play key roles in the survival and reproductive success of insects. Insect chemoreception is mediated by two large diverse gene superfamilies, chemoreceptors odorant binding proteins (OBPs). OBPs are believed to transport hydrophobic odorants from environment olfactory receptors. Results We identified a family OBP-like genes silkworm genome characterized their expression using oligonucleotide microarrays. A total forty-four OBP were annotated, number...

Establishment of efficient genome editing tools is essential for fundamental research, genetic engineering, and gene therapy. Successful construction application transcription activator-like effector nucleases (TALENs) in several organisms herald an exciting new era editing. We describe the production two active TALENs their successful targeted mutagenesis silkworm, Bombyx mori, whose manipulation methods are parallel to those Drosophila other insects. will also show that simultaneous...

CRISPR/Cas9, a bacterial adaptive immune system derived genome-editing technique, has become to be one of the most compelling topics in biotechnology. Bombyx mori is an economically important insect and model organism for studying lepidopteran arthropod biology. Here we reported highly efficient multiplex genome editing B. cell line heritable site-directed mutagenesis Bmku70, which required NHEJ pathway also related antigen diversity, telomere length maintenance subtelomeric gene silencing,...

Background and Aims: Hepatitis B surface antigen (HBsAg) loss is seldom achieved with nucleos(t)ide analog (NA) therapy in chronic hepatitis patients but may be enhanced by switching to finite pegylated-interferon (Peg-IFN) alfa-2a. We assessed HBsAg 48- 96-week Peg-IFN alfa-2a partial response a previous NA. Methods: e (HBeAg)-positive who HBeAg virus DNA <200 IU/mL adefovir, lamivudine or entecavir treatment were randomized 1:1 receive for 48 (n = 153) 96 weeks 150). The primary endpoint...

Abstract Ginger (Zingiber officinale), the type species of Zingiberaceae, is one most widespread medicinal plants and spices. Here, we report a high-quality, chromosome-scale reference genome ginger ‘Zhugen’, traditionally cultivated in Southwest China used as fresh vegetable, assembled from PacBio long reads, Illumina short high-throughput chromosome conformation capture (Hi-C) reads. The was phased into two haplotypes, haplotype 1 (1.53 Gb with contig N50 4.68 M) 0 (1.51 5.28 M)....

Abstract Gene expression quantification at mRNA level is very important for post‐genomic studies, as gene the reflection of special biological function target gene. Methods used quantification, such microarray or quantitative real‐time polymerase chain reaction (qRT‐PCR), require stable expressed reference genes. Thus, finding suitable control genes essential quantification. In this study, a genome‐wide survey during metamorphism was performed on silkworm Bombyx mori. Twelve were chosen...

N6-methyladenosine (m6A) methylation is the most common form of RNA modification in eukaryotes and has been shown to act as an important epitranscriptomic marker, which can regulate gene expression, messenger stability, alternative splicing translation efficiency. Recent progress on determining insect m6A function limited dipteran Drosophila melanogaster, involved neuronal functions sex determination. Methylation other insects, however, remain unknown. Here, we investigated a...

Abstract Insects produce silk to form cocoons, nests, and webs, which are important for their survival reproduction. However, little is known about the molecular mechanism of protein synthesis at translation level. The solute carrier family 7 (SLC7) genes involved in activating target rapamycin complex 1 (TORC1) signaling pathway process, but physiological roles SLC7 silk‐producing insects have not been reported. Here, we found that amino acid regulates larval development via L‐type...

The Toll receptor signaling pathway is an important innate immune response of insects to pathogen infection; its extracellular signal transduction involves serine protease cascade activation. However, excessive or constitutive activation the can be detrimental. Hence, balance between and inhibition must tightly regulated achieve favorable outcomes.
