Ferroptosis, featuring an iron‐dependent peroxidation of lipids, is a novel form programmed cell death that may hold great potential in cancer therapy. Our study found palmitic acid (PA) inhibited colon viability vitro and vivo , conjunction with accumulation reactive oxygen species lipid peroxidation. The ferroptosis inhibitor Ferrostatin‐1 but not Z‐VAD‐FMK (a pan‐caspase inhibitor), Necrostatin‐1 potent necroptosis or CQ autophagy), rescued the phenotype induced by PA. Subsequently, we...

Abstract Cancer stem cells (CSCs) are an important cause of tumor recurrence and drug resistance. As a new type cell death that relies on iron ions is strictly regulated by intracellular extracellular signals, the role ferroptosis in deserves extensive attention. Mass spectrum was applied to screen for ferroptosis-related proteins gastric cancer (GC). Sphere-formation assay used estimate stemness (GCSCs). Exosomal lnc-ENDOG-1:1 (lncFERO) isolated ultracentrifugation. Ferroptosis induced...

Ferroptosis is a type of cell death accompanied by iron-dependent lipid peroxidation, thus stimulating ferroptosis may be potential strategy for treating gastric cancer, therapeutic agents against which are urgently required. Jiyuan oridonin A (JDA) natural compound isolated from Rabdosia rubescens with anti-tumor activity, unclear mechanisms and limited water solubility hamper its clinical application. Here, we showed a2, new JDA derivative, inhibited the growth cancer cells. Subsequently,...

Histone lysine specific demethylase 1 (LSD1) has been recognized as an important modulator in post-translational process epigenetics. Dysregulation of LSD1 implicated the development various cancers. Herein, we report discovery hit compound 8a (IC50 = 3.93 μmol/L) and further medicinal chemistry efforts, leading to generation 15u 49 nmol/L, Ki 16 nmol/L), which inhibited reversibly competitively with H3K4me2, was selective over MAO-A/B. Docking studies were performed rationalize potency 15u....

New Delhi metallo-β-lactamase-1 (NDM-1) is capable of hydrolyzing nearly all β-lactam antibiotics, posing an emerging threat to public health. There are currently less effective treatment options for treating NDM-1 positive "superbug", and no promising inhibitors were used in clinical practice. In this study, structure-activity relationship based on thiosemicarbazone derivatives was systematically characterized their potential activities combined with meropenem (MEM) evaluated. Compounds...

Patients with ulcerative colitis (UC) have been found to be frequently associated secondary liver injury (SLI). In this study, we investigated the protective effect of GA on dextran sodium sulfate (DSS)-induced SLI in mice and its mechanism. The was established by adding 4% DSS drinking water mice, effects (5, 20 mg/kg, p.o., once a day for 7 days) hepatic tissues were analyzed. HepG2 cells induced lipopolysaccharide (LPS) detect ferroptosis underlying Pathological damage determined H&E....

Catalysis of ADP-ATP exchange by nucleotide factors (NEFs) is central to the activity Hsp70 molecular chaperones. Yet, mechanism interaction this family chaperones with NEFs not well understood in context sequence evolution and structural dynamics ATPase domains. We studied interactions domains four different on basis evolutionary trace co-evolution domain sequence, combined elastic network modeling collective complexes. Capsaicin (CAP), a frequently occurring alkaloid component found in spicy peppers, has demonstrated therapeutic potential against tumors, metabolic disease, and cardiovascular disorders. Doxorubicin (DOX), widely used anthracycline drug chemotherapy, is notorious for its cardiotoxicity. This study aimed to investigate the of CAP mitigating DOX toxicity mouse hearts H9C2 cells, as well explore underlying mechanisms. In our study, we conducted experiments on both mice cells. USP28, a member of the deubiquitinating enzymes family, plays vital role in physiological process cell proliferation, differentiation and apoptosis, DNA repair, immune response, stress response. USP28 has been reported to be overexpressed bladder cancer, colon breast carcinomas, so on. Nevertheless, gastric cancer not yet investigated. In our study, we examined expression 87 paired samples normal tissues. We found that was compared with tissues (P < 0.01), its overexpression related degree...

SLC7A11: also known as Sodium-Coupled Neutral Amino Acid Transporter 11, is an essential transporter protein that plays a vital role in the intracellular transport of amino acids, especially cysteine. Recently, its involvement cell death mechanisms has attracted considerable interest, underscoring significance both normal physiological functions and various disease states. This paper aims to provide thorough overview current understanding SLC7A11 two specific types death: disulfidptosis...

We evaluated a novel plasma (1,3)-β-d-glucan (BDG) detection assay for the diagnosis of candidemia in children. The median BDG levels were 73.4 pg/ml patients with and <10 without (P < 0.001). Receiver operating characteristic analysis revealed cutoff point 14 an area under curve 0.802. At these values, demonstrated 68% sensitivity, 91% specificity, 66% positive predictive value, negative value. Plasma undetectable 18 cases.
