Background While it is known that CD123 normally strongly expressed on plasmacytoid dendritic cells and completely absent nucleated red blood cells, detailed information regarding expression in acute leukemia scarce and, if available, hard to compare due different methodologies. Methods was evaluated using standardized EuroFlow immunophenotyping 139 pediatric AML, 316 adult 193 BCP‐ALL, 69 101 T‐ALL, 28 T‐ALL patients. Paired diagnosis‐relapse samples were available for 57 AML 19 BCP‐ALL...

Background Acute myeloid leukemia (AML) is a hematopoietic malignancy which biologically, phenotypically and genetically very heterogeneous. Outcome of patients with AML remains dismal, highlighting the need for improved, less toxic therapies. Chimeric antigen receptor T-cell (CART) immunotherapies refractory or relapse (R/R) are challenging because absence universal pan-AML target shared expression antigens normal stem/progenitor cells (HSPCs), may lead to life-threating on-target/off-tumor...

Precise classification of acute leukemia (AL) is crucial for adequate treatment. EuroFlow has previously designed an AL orientation tube (ALOT) to guide towards the relevant panel (T-cell lymphoblastic (T-ALL), B-cell precursor (BCP)-ALL and/or myeloid (AML)) and final diagnosis. Now we built a reference database with 656 typical samples (145 T-ALL, 377 BCP-ALL, 134 AML), processed analyzed via standardized protocols. Using principal component analysis (PCA)-based plots automated algorithms...

Acute megakaryoblastic leukemia (AMKL) is a rare and heterogeneous subtype of acute myeloid (AML). We evaluated the immunophenotypic profile 72 AMKL 114 non-AMKL AML patients using EuroFlow panel. Univariate multivariate/multidimensional analyses were performed to identify most relevant markers contributing diagnosis AMKL. subdivided into transient abnormal myelopoiesis (TAM), associated with Down syndrome (ML-DS), AML-not otherwise specified megakaryocytic differentiation (NOS-AMKL),...

Precise classification of acute leukemia (AL) is crucial for adequate treatment. EuroFlow has previously designed an AL orientation tube (ALOT) to guide toward the relevant panel and final diagnosis. In this study, we validated algorithm automated (database-supported) gating identification (AGI tool) cell subsets within samples stained with ALOT. A reference database normal peripheral blood (PB, n = 41) bone marrow (BM; 45) analyzed ALOT was constructed, served as a AGI tool automatically...

Summary Given its myeloid‐restricted expression, myeloperoxidase (MPO) is typically used for lineage assignment (myeloid vs. lymphoid) during acute leukaemia (AL) diagnostics. In the present study, a robust flow cytometric definition MPO positivity was established based on standardised EuroFlow protocols, Acute Leukaemia Orientation Tube and 1734 multicentre AL cases (with confirmed assay stability). The best diagnostic performance achieved by defining as ≥20% of cells exceeding...

Flowcytometric analysis allows for detailed identification and characterization of large numbers cells in blood, bone marrow, other body fluids tissue samples therefore contributes to the diagnostics hematological malignancies. Novel data tools allow multidimensional comparison patient with reference databases normal, reactive, and/or leukemia/lymphoma samples. Building such requires strict quality assessment (QA) procedures. Here, we compiled a dataset developed QA methodology EuroFlow...

Nowadays, most cytometrists apply lossless compression by storing their FCS files in ZIP archives. Unfortunately, only achieves modest space savings cytometric data, due to DEFLATE being used as the underlying algorithm (LCA). Presumably, other modern LCA can outperform DEFLATE, especially terms of savings. Twenty-one codecs (programs implementing LCA) were evaluated 167,131 publicly available files. Within floating-point produced instruments, favorable ratios (CRs) achieved ZPAQ (median...

We evaluated whether bone marrow aspirate samples isolated from femoral heads could be used verification cutoff values the individual (IPS), contributing to FCS and compared applicability in two centers.Bone cells were obtained who underwent hip replacement surgery. Abstract When it comes to data storage, the field of flow cytometry is fairly standardized, thanks standard (FCS) file format. The structure FCS files described in specification. Software that strictly complies with specification guaranteed be interoperable (in terms exchange via files). Nowadays, software interoperability crucial for eco system, as are frequently shared, and workflows rely on more than one piece (e.g., acquisition analysis software). Ideally, developers follow...
