Drugs frequently require interactions with multiple targets—via a process known as polypharmacology—to achieve their therapeutic actions. Currently, drugs targeting several serotonin receptors, including the 5-HT2C receptor, are useful for treating obesity, drug abuse, and schizophrenia. The competing challenges of developing selective receptor ligands or creating defined polypharmacological profile, especially aimed at G protein-coupled receptors (GPCRs), remain extremely difficult. Here,...

Coupled magmatic and tectonic activity plays an important role in high-temperature hydrothermal circulation at mid-ocean ridges. The patterns for such systems have been elucidated by microearthquakes geochemical data over a broad spectrum of spreading rates, but not generally available ultra-slow Here we report new geophysical fluid active venting Dragon Horn area (49.7°E) on the Southwest Indian Ridge. Twin detachment faults penetrating to depth 13 ± 2 km below seafloor were identified...

Significance Recent success of T cell-based cancer immunotherapies highlights the importance further understanding molecular mechanisms in regulation cell responsiveness. Here, we performed a genome-wide CRISPR screen to identify genes that regulate activation upon anti-T receptor (TCR) stimulation. Our confirmed many known regulators proximal signaling. Moreover, identified previously uncharacterized gene named FAM49B, which acts as negative regulator activation. study suggests screening is...

Abstract The Smoothened receptor (SMO) belongs to the Class Frizzled of G protein-coupled (GPCR) superfamily, constituting a key component Hedgehog signalling pathway. Here we report crystal structure multi-domain human SMO, bound and stabilized by designed tool ligand TC114, using an X-ray free-electron laser source at 2.9 Å. reveals precise arrangement three distinct domains: seven-transmembrane helices domain (TMD), hinge (HD) intact extracellular cysteine-rich (CRD). This architecture...

Asparagine synthetase (ASNS) and CTP synthase (CTPS) are two metabolic enzymes crucial for glutamine homeostasis. A genome-wide screening in Saccharomyces cerevisiae reveal that both ASNS CTPS form filamentous structures termed cytoophidia. Although cytoophidia were well documented recent years, the filamentation of is less studied. Using budding yeast as a model system, here we confirm proteins, Asn1 Asn2, capable forming diauxic stationary phases. We find glucose deprivation induces...

Urban green spaces are crucial for improving the quality of life city residents because they provide numerous health advantages and foster overall wellness. However, previous research has concentrated only on individual goals overlooked inherent contradictions that arise when many objectives considered. This study devised a transdisciplinary framework to link optimization plant community characteristics with multiple objectives. In Beijing, we selected 22 urban utilized non-dominated sorting...

We unveiled an underside binding site on smoothened receptor (SMO) by a colocalization strategy using two structurally complementary photoaffinity probes derived from known ligand Allo-1. Docking study and structural dissection identified key interactions within the site, including hydrogen bonding, π–π interactions, hydrophobic between Allo-1 its contacting residues. Taken together, our results reveal molecular base of provide basis for design new-generation ligands to overcome drug resistance.
