High-quality and disease-resistant male sterile lines have great potential for applications in hybrid rice breeding. We introduced specific mutations into the TMS5, Pi21, Xa13 genes Pinzhan intermediate breeding material using CRISPR/Cas9 multiplex genome editing system. found that transgene-free homozygous triple tms5/pi21/xa13 mutants obtained T1 generation displayed characteristics of thermosensitive genic sterility (TGMS) with enhanced resistance to blast bacterial blight. Our study...
Abstract Loss-of-function mutagenesis is an important tool used to characterize gene functions and the CRISPR-Cas9 system a powerful method for performing targeted in organisms that present low recombination frequencies, such as serotype D strains of Cryptococcus neoformans. However, when persists host cells, off-target effects Cas9 cytotoxicity may occur, which might block subsequent genetic manipulation. Here, we report spontaneously eliminating without impairing its robust editing...
ABSTRACT Virulence is the outcome of an interaction between host and a microbe characterized by large array opposing reactions operating at host-pathogen interface. Cryptococcus neoformans important opportunistic pathogen in immunocompromised patients, including those with human immunodeficiency virus, expresses virulence-associated laccase which believed to oxidize brain catecholamines iron as defense against immune cells. In present report, we investigated cellular location understand more...
The nematicidal activity of Chaetomium globosum NK102, culture filtrates, and chaetoglobosin A (ChA) purified by HPLC was evaluated on Meloidogyne incognita . results showed that C. NK102 significantly repelled second-stage juveniles (J2s). Both filtrates ChA demonstrated strong adverse effects J2 mortality with 99.8% at 300 μg ChA/mL (LC(50) = 77.0 μg/mL) 72 h. did not affect egg hatch until h exposure. All filtrate treatments inhibited the penetration even in 12.5% dilution treatment....
microRNAs (miRNAs), endogenous posttranscriptional repressors by base-pairing of their cognate mRNAs in plants and animals, have mostly been thought lost the kingdom fungi. Here, we report identification miRNAs from fungus Cryptococcus neoformans. With bioinformatics Northern blotting approaches, found that these hairpin precursors were present this fungus. The size miR1 miR2 is 22 nt 18 nt, respectively. are about ∼70 length close to mammalian pre-miRNAs. Characteristic features also...
Clinical translation of therapeutic peptides, particularly those that require penetration the cell membrane or are cytolytic, is a major challenge. A novel approach based on complementary mechanism, which has been widely used for guided synthesis DNA RNA nanoparticles, de novo design activatable protein nanoparticles (APNPs) targeted delivery peptides described. APNPs formed through self-assembly three independent polypeptides pairwise coiled-coil dimerization. They capable long circulation...
Summary Laccase is a major virulence factor required for infection caused by the human pathogenic yeast Cryptococcus neoformans . However, cellular processes involved in regulation and expression of laccase remain largely unknown C. Here we report identification chloride channel gene CLC‐A which essential activity shares homology to CLC‐type voltage‐gated channels from other organisms; example, 63% GEF1 , Saccharomyces cerevisiae A clc‐a mutant, Mlac3, generated insertional mutagenesis as...
The multicopper laccase is a major virulence factor in Cryptococcus neoformans. Its expression regulation complex. We presented molecular evidence to show that was induced by high concentrations of exogenous copper. Melanin production and enzymatic activity increased dramatically response the addition copper media. Reverse transcription-PCR amplification gene LAC1 mRNA revealed induction occurred at transcriptional level, which required copper-responsive factor-encoding CUF1. Disruption CUF1...
Haloxyfop-R-methyl is a widely used herbicide to control Poaceae weeds. Alopecurus japonicus, widespread annual grass, can no longer be controlled by haloxyfop-R-methyl after continuous use of this for several years.Dose-response experiments have established that the Js-R biotype A. japonicas has evolved resistance aryloxyphenoxypropionates (APPs). Target-site enzyme sensitivity haloxyfop (free acid) rate causing 50% inhibition acetyl-CoA carboxylase (ACCase) activity (I(50)) resistant...
The Mausoleum of the Dingtao King (termed 'M2') is a large-scale huangchang ticou tomb that dates to Western Han Dynasty (206 B.C.–25 A.D.). It highest-ranking discovered date. However, biodeterioration on surface M2 causing severe damage its wooden materials. aim present study was give insight into fungal communities colonized tomb. For this purpose, seven samples were collected from different sections which exhibited obvious in form white spots. Microbial structures associated with spots...
Since Internet of Things (IoT) has been widely used in our daily life nowadays, it is regarded as a promising and popular application the Internet, attracted more attention. However, IoT also suffered by some security problems which seriously affect implementation system.Similar to traditional software, software always threated many vulnerabilities, thus how evaluate situation chain becomes basic requirement. In this paper, A framework awareness for proposed, mainly includes two processes:...
