Type 2 diabetes (T2D) is caused by insufficient insulin secretion from pancreatic β cells. To identify candidate genes contributing to T2D pathophysiology, we studied human islets approximately 300 individuals. We found 395 differentially expressed (DEGs) in individuals with T2D, including, our knowledge, novel (OPRD1, PAX5, TET1) and previously identified (CHL1, GLRA1, IAPP) candidates. A third of the expression changes may predispose diabetes, as these associated HbA1c not diagnosed T2D....
Readily accessible human pancreatic beta cells that are functionally close to primary adult a crucial model better understand cell physiology and develop new treatments for diabetes. We here report the characterization of EndoC-βH5 cells, latest in EndoC-βH family.EndoC-βH5 were generated by integrative gene transfer immortalizing transgenes hTERT SV40 large T along with Herpes Simplex Virus-1 thymidine kinase into fetal pancreas. Immortalizing removed after amplification using CRE...
Biphasic insulin secretion, which fails in Type 2 Diabetes, can be provoked by various nutrient stimuli, glucose being the superior physiological one. To identify pathways that may play a role β-cell stimulus-secretion coupling, we compared and islet functional, secretory, metabolic responses to 2-aminobicyclo-(2,2,1)-heptane-2-carboxylic acid (BCH), leucine analog, acts as an allosteric activator of glutamate dehydrogenase (GDH). We employed range techniques, including secretion assays,...
Graphs, an effective form of data presentation, are used for summarizing complex information and making them easier to understand. Extracting numerical from graphs, which is commonly required in systematic reviews meta-analyses, however a challenging issue. Since this kind results presentation common, ignorance such may result publication bias when conducting meta-analyses. On the other hand, contacting authors particular order retrieve take long time often not very fruitful. In case, there...
Decreased circulating levels of hydrogen sulfide (H2S) are associated with higher mortality following myocardial ischemia. This study aimed at determining the long-term dose-dependent effects sodium hydrosulfide (NaSH) administration on ischemia-reperfusion (IR) injury. Male rats were divided into control and NaSH groups that treated for 9 weeks daily intraperitoneal injections normal saline or (0.28, 0.56, 1.6, 2.8, 5.6 mg/kg), respectively. At end study, hearts from all isolated...
Skeletal muscle is an endocrine organ secreting exercise-induced factors (exerkines), which play a pivotal role in interorgan cross talk. Using mass spectrometry (MS)-based proteomics, we characterized the secretome and identified thymosin β4 (TMSB4X) as most upregulated secreted protein media of contracting C2C12 myotubes. TMSB4X was also acutely increased plasma exercising humans irrespective insulin resistance condition or exercise mode. Treatment mice with did not ameliorate metabolic...
We previously reported that loss of mitochondrial transcription factor B1 (TFB1M) leads to dysfunction and is involved in the pathogenesis type 2 diabetes (T2D). Whether defects ribosomal processing impact function could play a pathogenetic role β-cells T2D not known. To this end, we explored expression functional dimethyladenosine transferase 1 homolog (DIMT1), TFB1M RNA (rRNA) methyltransferase implicated control rRNA. Expression DIMT1 was increased human islets from donors correlated...
Ependymin-Related Protein 1 (EPDR1) was recently identified as a secreted human batokine regulating mitochondrial respiration linked to thermogenesis in brown fat. Despite that EPDR1 is expressed pancreatic β-cells and glucose-stimulated metabolism critical for stimulus-secretion coupling β-cells, the role of β-cell function has not been investigated.EPDR1 mRNA levels islets from non-diabetic (ND) type 2 diabetes (T2D) subjects were assessed. Human islets, EndoC-βH1 INS1 832/13 cells...
Hydrogen sulfide (H₂S) is involved in the pathophysiology of type 2 diabetes. Inhibition and stimulation H₂S synthesis has been suggested to be a potential therapeutic approach for The aim this study was therefore determine effects long-term sodium hydrosulfide (NaSH) administration as releasing agent on carbohydrate metabolism diabetic rats. Type diabetes established using high fat-low dose streptozotocin. Rats were treated 9 weeks with intraperitoneal injections NaSH (0.28, 0.56, 1.6, 2.8,...
Reversible phosphorylation is an important regulatory mechanism. Regulation of protein in β-cells has been extensively investigated, but less known about dephosphorylation. To understand the role dephosphorylation and type 2 diabetes (T2D), we first examined mRNA expression 2C family (PP2C) phosphatases islets from T2D donors. Phosphatase overall was changed T2D, that PPM1E most markedly downregulated. correlated inversely with HbA1c. Silencing increased glucose-stimulated insulin secretion...
Nitrite, a nitric oxide (NO) donor, increases insulin secretion from pancreatic islets and has positive metabolic effects in type 2 diabetes (T2D). Here, we test the hypothesis of whether nitrite-induced is due to blunting diabetes-induced oxidative stress islets.T2D was created male rats using combination streptozotocin at 25 mg/kg high-fat diet. Wistar were assigned 3 groups (n=6 each group), including control, T2D, T2D+nitrite; latter group consumed drinking water containing sodium...
