Abstract The emergence of multidrug-resistant (MDR) Gram-negative pathogens is an urgent global medical challenge. old polymyxin lipopeptide antibiotics (polymyxin B and colistin) are often the only therapeutic option due to resistance all other classes lean antibiotic drug development pipeline. However, colistin suffer from major issues in safety (dose-limiting nephrotoxicity, acute toxicity), pharmacokinetics (poor exposure lungs) efficacy (negligible activity against pulmonary infections)...
A novel Gram-stain-negative, rods or bent rods, facultatively anaerobic, oxidase-negative and catalase-positive bacterium, designated XK5T, was isolated from coastal sediment Xiaoshi Island, Weihai, China. Optimal growth occurred at 28-35 °C (range 8-42 °C) pH 7.0-8.0 6.0-9.0) with 1-3 % (w/v) NaCl 0.5-8 %). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain XK5T 92.1 similar to the type of Thioalkalivibrio thiocyanodenitrificans, 91.9 sulfidiphilus 91.8...
Recurrent urinary tract infections (rUTI) are common in kidney transplant recipients (KTR) and associated with multidrug resistance increased morbidity/mortality. Novel antibiotic alternatives to reduce UTI recurrence critically needed. We describe a case of rUTI due extended spectrum beta lactamase (ESBL) Klebsiella pneumoniae KTR that was treated successfully 4 weeks adjunctive intravenous bacteriophage therapy alone, without concomitant antibiotics, no year follow-up.
There is an urgent unmet medical need for new treatments wound and burn infections caused by multidrug-resistant Gram-negative "superbugs," especially the problematic Pseudomonas aeruginosa. In this work, incorporation of colistin, a potent lipopeptide into self-healable hydrogel (via dynamic imine bond formation) following chemical reaction between amine groups present in glycol chitosan aldehyde-modified poly(ethylene glycol), reported. The storage module (G') colistin-loaded ranges from...
Colistin is often administered by inhalation and/or the parenteral route for treatment of respiratory infections caused multidrug-resistant (MDR) Pseudomonas aeruginosa However, limited pharmacokinetic (PK) and pharmacodynamic (PD) data are available to guide optimization dosage regimens inhaled colistin. In present study, PK colistin in epithelial lining fluid (ELF) plasma was determined following intratracheal delivery a single dose solution neutropenic lung-infected mice. The...
Pseudomonas aeruginosa often causes multidrug-resistant infections in immunocompromised patients, and polymyxins are used as the last-line therapy. Alarmingly, resistance to has been increasingly reported worldwide recently. To rescue this last-resort class of antibiotics, it is necessary systematically understand how P. alters its metabolism response polymyxin treatment, thereby facilitating development effective therapies. end, a genome-scale metabolic model (GSMM) was analyze bacterial...

A Gram-stain-negative, facultatively anaerobic, moderately halophilic, filamentous, non-motile bacterium, designated FA102T, was isolated from marine sediment the coast of Weihai, PR China. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain FA102T formed a distinct evolutionary lineage within family Marinifilaceae and its closest relative Marinifilum fragile JCM 15579T (93.2 % sequence similarity). The DNA G+C content novel 36.5 mol%. predominant cellular fatty acids...
Combating antimicrobial resistance has been highlighted as a critical global health priority. Due to the drying drug discovery pipeline, polymyxins have employed last-line therapy against Gram-negative “superbugs”; however, detailed mechanisms of antibacterial killing remain largely unclear, hampering improvement polymyxin therapy. Our integrative modeling using constructed genome-scale metabolic model i ATCC19606 and correlative multiomics data provide fundamental understanding complex...
Multidrug-resistant (MDR) Acinetobacter baumannii is a critical threat to human health globally. We constructed genome-scale metabolic model iAB5075 for the hypervirulent, MDR A. strain AB5075. Predictions of nutrient utilization and gene essentiality were validated using Biolog assay transposon mutant library. In vivo transcriptomics data integrated with elucidate bacterial responses host environment. contains 1530 metabolites, 2229 reactions, 1015 genes, demonstrated high accuracies in...
A Gram-stain-negative, facultatively anaerobic, orange-pigmented bacterium, designated HQYD1T, was isolated from a sea squirt (Styelaclava) and characterized using polyphasic approach. Morphologically, strain HQYD1T exhibited rods with gliding motility. This novel isolate grew optimally at 28 °C in the presence of 2-3 % (w/v) NaCl. The 16S rRNA gene sequence most similar to [Saccharicrinis] marinus Y11T (96.3 %), followed by Saccharicinis fermentans DSM 9555T (93.8 %). dominant fatty acids...
Multidrug-resistant (MDR) Acinetobacter baumannii is a critical threat to global health. The type strain ATCC 19606 has been widely used in studying the virulence, pathogenesis and mechanisms of antimicrobial resistance A. baumannii. However, lack complete genome sequence hindrance towards detailed bioinformatic studies. Here we report generation for using PacBio sequencing. consists 3,980,848-bp chromosome 9,450-bp plasmid pMAC, harbours chromosomal dihydropteroate synthase gene sul2...
Despite recent recognition of the ATP-binding cassette protein OptrA as an important mediator linezolid resistance in Enterococcus faecalis worldwide, mechanisms optrA gene acquisition and transfer remain poorly understood. In this study, we performed comprehensive molecular phenotypic profiling 44 optrA-carrying E. clinical isolates with resistance. Pulse-field gel electrophoresis DNA hybridization revealed presence plasmid 26 (59%) chromosome 18 (41%) isolates. Conjugation experiments...
Polymyxin B and colistin are exclusively active against Gram-negative pathogens have been used in the clinic as a last-line therapy. In this study, we investigated antimicrobial activity of novel polymyxin, FADDI-019, Staphylococcus aureus. MIC time-kill assays were employed to measure FADDI-019 S. aureus ATCC 700699. Cell morphology was examined with scanning electron microscopy (SEM), cell membrane polarity measured using flow cytometry. Transcriptome changes caused by treatment...
Klebsiella pneumoniae is an opportunistic Gram-negative pathogen causing nosocomial infections. K. rapidly acquires antibiotic resistance and known as a reservoir for genes. Polymyxins remain effective last-line therapy against infections caused by multidrug-resistant (MDR) pneumoniae; however, to polymyxins emerges with monotherapy. Synergistic combinations of FDA-approved non-antibiotics are novel approach preserve its efficacy whilst minimising the emergence polymyxin in pneumoniae. This...
