Cladribine administration has been approved for the treatment of relapsing-remitting multiple sclerosis (MS) in 2017; thus, data on cladribine a real-world setting are still emerging. We report effectiveness, safety profile, and response predictors 243 patients with MS followed at eight tertiary centers. Study outcomes were: (1) No Evidence Disease Activity-3 (NEDA-3) status its components (absence clinical relapses, MRI activity, sustained disability worsening); (2) development grade III/IV...

To determine relative frequencies and linguistic profiles of primary progressive aphasia (PPA) variants associated with GRN (progranulin) mutations to study their neuroanatomic correlates.Patients PPA carrying (PPA-GRN) were selected among a national prospective research cohort 1,696 patients frontotemporal dementia, including 235 PPA. All amyloid-positive CSF biomarkers excluded. In this cross-sectional study, speech/language cognitive characterized standardized evaluations, gray matter...

Abstract Mild cognitive impairment (MCI) defines an intermediate state between normal ageing and dementia, including Alzheimer’s disease (AD). Identification of MCI subjects who will progress to AD (MCI-AD) is today crucial importance, especially in light the possible development new pathogenic therapies. Several evidences suggest that miRNAs could play relevant roles biogenesis AD, links selected specific aspects have been partly explored. In this study, we analysed composition microRNA...

Novel amphiphilic cyclic disaccharide analogues, in which the saccharide units are connected through stable phosphodiester linkages (CyPLOS, Cyclic Phosphate-Linked OligoSaccharides) and decorated with long lipophilic tentacles at 2- 3-OH moieties, have been synthesized. Their propensity to self-aggregation has investigated by means of 1H 31P NMR experiments, making it possible determine for these macrocycles critical aggregation concentration values millimolar range.

Abstract A novel fluorescently labelled synthetic ionophore, based on a cyclic phosphate‐linked disaccharide (CyPLOS) backbone and decorated with four tetraethylene glycol tails carrying dansyl units, has been synthesised in 12 steps 26 % overall yield. The key intermediate the strategy is glucoside building block, serving through its 2‐ 3‐hydroxy groups as anchor point for flexible tentacles reactive azido moieties at their ends. To test versatility of this scaffold, it was preliminarily...

Abstract With the aim of preparing a library ion transporters based on CyPLOS (cyclic phosphate‐linked oligosaccharide) backbone, we describe synthesis novel azido‐derivatized analogue that is tailored for practical post‐synthetic functionalization with variety labels. As prerequisite effective preparation this macrocycle, compatibility azido alcohols as coupling agents in phosphoramidite synthetic protocols was preliminarily investigated solution by using 31 P NMR spectroscopy to monitor...

Cutting-edge research suggests endosomal/immune dysregulation in GRN/C9orf72-associated frontotemporal lobar degeneration (FTLD). In this retrospective study, we investigated plasma small extracellular vesicles (sEVs) and complement proteins 172 subjects (40 Sporadic FTLD, 40 Intermediate/Pathological C9orf72 expansion carriers, 49 Heterozygous/Homozygous GRN mutation 43 controls). Plasma sEVs (concentration, size) were analyzed by nanoparticle tracking analysis; C1q, C4, C3 quantified...

Semantic and right temporal variant of frontotemporal dementia (svFTD rtvFTD) are rare clinical phenotypes in which, most cases, the underlying pathology is TDP-43 proteinopathy. They usually sporadic disorders, but recent evidences suggest a higher frequency genetic mutations for versus semantic variant. However, basis these forms not clear. In this study we performed screening single-center cohort svFTD rtvFTD patients, aiming at identifying associated variants. A panel 73 candidate genes...
