HIV-1 is a chronic disease managed by strictly adhering to daily antiretroviral therapy (ART). However, not all people living with have access ART, and those may adhere treatment regimens increasing viral load progression. Here, subcutaneous nanofluidic implant was used as long-acting (LA) drug delivery platform address these issues. The device loaded tenofovir alafenamide (TAF) implanted in treatment-naïve simian HIV (SHIV)-positive nonhuman primates (NHP) for month. We monitored...
There is an urgent need for accessible and low-cost COVID-19 vaccine suitable low- middle-income countries. Here, we report on the development of a SARS-CoV-2 receptor-binding domain (RBD) protein, expressed at high levels in yeast (Pichia pastoris), as candidate against COVID-19. After introducing two modifications into wild-type RBD gene to reduce yeast-derived hyperglycosylation improve stability during protein expression, show that recombinant RBD219-N1C1, equivalent (RBD219-WT) vitro...
Pre-exposure prophylaxis (PrEP) using antiretroviral oral drugs is effective at preventing HIV transmission when individuals adhere to the dosing regimen. Tenofovir alafenamide (TAF) a potent drug, with numerous long-acting (LA) delivery systems under development improve PrEP adherence. However, none has undergone preventive efficacy assessment. Here we show that LA TAF novel subcutaneous nanofluidic implant (nTAF) confers partial protection from transmission. We demonstrate sustained...

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was first reported in December 2019 Wuhan, China, and then rapidly spread causing an unprecedented pandemic. A robust serological assay is needed to evaluate vaccine candidates better understand the epidemiology of disease (COVID-19).We used full-length spike (S) protein SARS-CoV-2 for development qualitative quantitative IgG IgA anti-S enzyme linked immunosorbent assays (ELISA). total 320 sera were comprised pandemic from infected...
Chronic spontaneous urticaria presents with wheals and/or angioedema for >6 weeks without any specific triggers. The incidence of chronic is increased in patients comorbid autoimmune conditions. Here, we present a case 9-year-old type 1 diabetes and thyroid disease who first presented insulin pump site reactions concerning an insulin-related allergy. patient was successfully treated antihistamines later immunosuppression resumption therapy remission symptoms 18 months after onset.
Abstract Introduction Compared with previous geographically localized outbreaks of monkeypox (MPOX), the scale 2022 global mpox outbreak has been unprecedented, yet clinical features this remain incompletely characterized. Methods We identified patients diagnosed by polymerase chain reaction (PCR; n = 36) from July to September at a single, tertiary care institution in USA. Demographics, presentation, infection course, and histopathologic were reviewed. Results Conclusion Men who have sex...
Abstract Background In prior studies, HIV-1 BF recombinants with subtype F integrases failed to develop resistance raltegravir through the Q148H mutational pathway. We aimed determine role of subtype-specific polymorphisms in integrase on drug susceptibility, viral replication and integration. Methods Integrase sequences were retrieved from Los Alamos Database or obtained Garrahan HIV cohort. infectious molecular clones without (+ G140S) mutations constructed using B (NL4-3) F1(BF) ARMA159...
To compare closure rates and hearing outcomes of microscopic endoscopic tympanoplasty in pediatric patients.Retrospective chart review.Tertiary university medical center.Pediatric patients who underwent surgery by a fellowship-trained neurotologist between 2010 2019 with minimum 2 months follow-up, tympanic membrane perforation, no preoperative cholesteatoma.Transcanal or (MT) surgery. The primary outcome is postoperative the assessed using otomicroscopy at last follow-up appointment....
Xanthomas are localized deposits of lipid-rich histiocytes (also known as foam cells) in the connective tissue. Various subtypes xanthomas described literature, including eruptive, tuberous, tendinous, planar, and verrucous.1 Cutaneous xanthomatous eruptions can be associated with systemic diseases such metabolic disease, familial hyperlipidemias, lymphoproliferative disorders.1,2
Objective: To compare the effects of preoperative medical comorbidities and operative findings on success lateral graft tympanoplasty. Study Design: Retrospective chart review. Setting: Tertiary center. Patients: Ninety-six patients undergoing tympanoplasty from December 2008 to November 2020 with at least 2 months follow-up were included. Patient demographics, including smoking status, intraoperative findings, healing, hearing outcomes recorded. Interventions: Lateral Main Outcome Measures:...
Linear pigmented purpura, also known as unilateral linear capillaritis, is a clinical subtype of purpuric dermatoses (PPDs). Although several variants exist, the PPDs are benign group skin eruptions that present with petechiae and hyperpigmented macules, papules, and/or patches. Morphology, examination findings, histopathology differ across PPD subtypes.1,2 Thus far, exact etiology unknown.3
In article number 2000163, Alessandro Grattoni and colleagues perform the first ever HIV preventive efficacy study using a subcutaneous drug delivery implant. The implant uses nanofluidic membrane to sustainably deliver tenofovir alafenamide fumarate (TAF), antiretroviral, without pumps or actuation. Long-term sustained TAF release, controlled through physical electrostatic confinement in nanochannels of membrane, obviates daily oral dosing provide protection from infection nonhuman primates.
