Clostridia produce autoinducing peptides (AIPs) regulated by the accessory gene regulator (Agr) quorum sensing system, playing a critical role in intercellular communication. However, biosynthetic pathway and regulatory functions of clostridial AIPs remain inadequately characterized. In this study, we employed chemical quantification, genetic investigations, vitro reconstitution experiments to elucidate native Ca‐AIP Clostridium acetobutylicum, prominent industrial producer acetone, butanol,...
Western equine encephalomyelitis (WEE) virus (Togaviridae: Alphavirus) was shown previously to have arisen by recombination between eastern (EEE)- and Sindbis-like viruses (C. S. Hahn, Lustig, E. G. Strauss, J. H. Proc. Natl. Acad. Sci. USA 85:5997-6001, 1988). We now examined the recombinational history evolution of all belonging WEE antigenic complex, including Buggy Creek, Fort Morgan, Highlands J, Sindbis, Babanki, Ockelbo, Kyzylagach, Whataroa, Aura viruses, using nucleotide sequences...
Eastern equine encephalitis virus (EEEV), the sole species in EEE antigenic complex, is divided into North and South American varieties based on hemagglutination inhibition tests. Here we describe serologic phylogenetic analyses of representatives these varieties, spanning entire temporal geographic range available. Nucleotide sequencing revealed additional genetic diversity within variety; 3 major South/Central lineages were identified including one represented by a single isolate from...
Naturally evolved metabolons have the ability to assemble and disassemble in response environmental stimuli, allowing for rapid reorganization of chemical reactions living cells meet changing cellular needs. However, replicating such capability synthetic remains a challenge due our limited understanding mechanisms by which assembly disassembly naturally occurring multienzyme complexes are controlled. Here, we report synthesis chemical- light-responsive protein cages assembling metabolons,...
Venezuelan equine encephalitis (VEE) epidemics and epizootics occurred periodically in the Americas from 1920s until early 1970s, when causative viruses, subtypes IAB IC, were postulated to have become extinct. Recent outbreaks Columbia Venezuela renewed interest source of epidemic/epizootic viruses their mechanism interepizootic maintenance. We performed phylogenetic analyses VEE virus isolates spanning entire temporal geographic range strains available, using 857-nucleotide reverse...
Abstract Cells use self‐assembled biomaterials such as lipid membranes or proteinaceous shells to coordinate thousands of reactions that simultaneously take place within crowded spaces. However, mimicking spatial organization for synthetic applications in engineered systems remains a challenge, resulting inferior catalytic efficiency. In this work, we show protein cages an ideal scaffold organize enzymes enhance cascade both vitro and living cells. We demonstrate not only enzyme‐enzyme...
Marine natural product (MNP) pretrichodermamide B (Pre B, 9) was identified as a novel STAT3 inhibitor in our previous work, while its metabolic instability hindered further development. To address this drawback, ligand structure-based drug design adopted leading to series of Pre derivatives. Among them, MNP trichodermamide (tri 24) obtained by skeletal rearrangement exhibited more potent antiproliferative activity with an IC50 value 0.12 μM against HCT116. Notably, 24 stood out improved...
Cannabichromene (CBC), a valuable but extremely low-abundance component of cannabinoids in Cannabis sativa L., is known for its ability to promote neurogenesis. The scarcity CBC natural C. primarily attributed the inefficiency 1-deoxy-D-xylulose 5-phosphate/2-C-methyl-D-erythritol 4 phosphate (DOXP/MEP) and fatty acid metabolism pathways, along with limited competitive advantage cannabichromenic synthetase (CBCAS) compared other cannabinoid synthases. In this study, we constructed...

Conventional antibody-based targeted cancer therapy is one of the most promising avenues successful treatment, with potential to reduce toxic side effects healthy cells surrounding tumor cells. However, full antibodies severely limited due their large size, low stability, slow clearance, and high immunogenicity. Alternatively, recently discovered nanobodies, which are smallest naturally occurring antigen-binding format, have shown great for addressing these limitations. Bioconjugation...
Epizootics of Venezuelan equine encephalitis (VEE) involving subtype IAB viruses occurred sporadically in South, Central and North America from 1938 to 1973. Incompletely inactivated vaccines have long been suspected as a source the later epizootics. We tested this hypothesis by sequencing PE2 glycoprotein precursor (1,677 nucleotides) or 26S/nonstructural protein 4 (nsP4) genome regions (4,490 for isolates representing most major outbreaks. Two distinct genotypes were identified: 1) 1940s...
Self-assembled protein cages are attractive scaffolds for organizing various proteins of interest (POIs) toward applications in synthetic biology and medical science. However, specifically attaching multiple POIs to a single cage remains challenging, resulting diversity among the functionalized particles. Here, we present engineering self-assembled cage,
Following a 19-year hiatus, Venezuelan equine encephalitis (VEE) reemerged in western Venezuela December 1992. This outbreak is important understanding VEE emergence because phylogenetic studies imply that sympatric, enzootic, subtype ID viruses mutated to generate the epizootic/epidemic. Although 1992-1993 strains belong IC, serotype implicated extensive outbreaks during 1960s and 1995, relatively small numbers of human cases occurred 1992-1993. We, therefore, evaluated pathogenicity these...
Clonorchis sinensis infection is highly prevalent in Asia. Diverse hepatobiliary morbidity has been documented for C . infection. This study aimed to assess the association between and morbidity, taking into consideration of control, confounders intensity. A cross-sectional community survey was implemented Hengxian county, southeastern China. Helminth infections were detected by fecal examination. Physical examination abdominal ultrasonography then conducted. After excluding confounding...
A module splicing method is developed for preparing an area expandable membrane that applied to coupled fermentation and separation.
During 1989-2011, three parasitic disease surveys were conducted in Hengxian County, China, where soil-transmitted helminthiases and foodborne clonorchiasis are endemic. We compared the data found that prevalence of decreased intensity increased over time, especially among men. Clonorchiasis control/intervention measures urgently needed this area.
Solventogenesis and sporulation of clostridia are the main responsive adaptations to acidic environment during acetone-butanol-ethanol (ABE) fermentation. It was hypothesized that five orphan histidine kinases (HKs) including Cac3319, Cac0323, Cac0903, Cac2730, Cac0437 determined cell fates between solventogenesis. In this study, comparative genomic analysis revealed a mutation in cac0437 appeared contribute nonsporulating feature ATCC 55025. Hence, individual interactive roles HKs...
