ABSTRACT We identified the domains of CD26 involved in binding Middle East respiratory syndrome coronavirus (MERS-CoV) using distinct clones anti-CD26 monoclonal antibodies (MAbs). One clone, named 2F9, almost completely inhibited viral entry. The humanized MAb YS110 also significantly infection. These findings indicate that both 2F9 and are potential therapeutic agents for MERS-CoV YS110, particular, is a good candidate immediate testing as modality MERS.
CD26 is an activation marker of human CD4(+) T cells, and associated with T-cell signal transduction processes as a co-stimulatory molecule. We have previously demonstrated that high cell surface expression on cells correlated the production helper type 1 cytokines, whereas CD26(+) stimulate antibody synthesis in B cells. Although cellular molecular mechanisms involved CD26-mediated been extensively evaluated by our group others, role CD8(+) has not clearly elucidated. In present study, we...
Pulmonary hypertension (PH) with interstitial lung diseases (ILDs) often causes intractable conditions. CD26/Dipeptidyl peptidase-4 (DPP4) is expressed in constituent cells and may be related to the pathogenesis of various respiratory diseases. We aimed clarify functional roles CD26/DPP4 PH-ILD, paying particular attention vascular smooth muscle (SMCs). Dpp4 knockout (Dpp4KO) wild type (WT) mice were administered bleomycin (BLM) intraperitoneally establish a PH-ILD model. The BLM-induced...
CD26 (DPP4) is a T cell costimulatory molecule as well activation marker, and CD26(+) cells are accumulated in inflamed tissues, such rheumatoid synovitis autoimmune thyroiditis. In the present study, we found accumulation of graft-versus-host disease (GVHD) target organs. To expand our vitro findings to an vivo system, examined CD26-dependent organ injury xenogeneic GVHD (x-GVHD) murine model. Following intraperitoneal injection human peripheral blood mononuclear into non-obese diabetic...
Obliterative bronchiolitis is a potentially life-threatening noninfectious pulmonary complication after allogeneic hematopoietic stem cell transplantation and the only pathognomonic manifestation of chronic graft-versus-host disease (cGVHD). In current study, we identified novel effect IL-26 on transplant-related obliterative bronchiolitis. Sublethally irradiated NOD/Shi-scidIL2rγ(null) mice transplanted with human umbilical cord blood (HuCB mice) gradually developed clinical signs (GVHD)...
Abstract The liver stores glycogen and releases glucose into the blood upon increased energy demand. Group 2 innate lymphoid cells (ILC2) in adipose pancreatic tissues are known for their involvement homeostasis, but metabolic contribution of ILC2s has not been studied detail. Here we show that directly involved regulation levels. Mechanistically, interleukin (IL)-33 treatment induces IL-13 production ILC2s, while suppressing gluconeogenesis a specific Hnf4a/G6pc-high primary hepatocyte...
Abstract Introduction: CD26 is a multifunctional transmembrane glycoprotein that overexpressed in many cancers. We established the humanized anti-CD26 monoclonal antibody YS110, which has direct anti-tumor effect through nuclear translocation of and transcriptional suppression POLR2A gene, subunit RNA polymerase II (Pol II). Subsequently, we developed an antibody-drug conjugate Y-TR-1 with YS110 Pol inhibitor triptolide evaluated its vitro vivo antitumor efficacy toxicity. then elucidated...
Acute respiratory distress syndrome (ARDS) is characterized by dysregulated inflammation and increased permeability of lung microvascular cells. CD26/dipeptidyl peptidase-4 (DPP4) a type II membrane protein that expressed in several cell types mediates multiple pleiotropic effects. We previously reported DPP4 inhibition sitagliptin attenuates lipopolysaccharide (LPS)-induced injury mice. The current study the functional role CD26/DPP4 expression LPS-induced mice, isolated alveolar...
Abstract Dipeptidyl peptidases (DPPs) are proteolytic enzymes that ideal therapeutic targets in human diseases. Indeed, DPP4 inhibitors widely used clinical practice as anti-diabetic agents. In this paper, we show also induced cell death multiple myeloma cells. Among five inhibitors, only two of them, vildagliptin and saxagliptin, exhibited apparent cytotoxic effects on lines, without any difference suppression activity. As these known to have off-target against DPP8/9, employed the specific...
CD26/dipeptidyl peptidase IV is a cell surface glycoprotein which consists of multiple functional domains beside its ectopeptidase site. A growing body evidence indicates that elevated expression CD26 correlates with disease aggressiveness and invasive potential selected malignancies. To further explore the molecular mechanisms involved in this clinical behavior, our current work focused on interaction between CD9, were recently identified as novel markers for cancer stem cells malignant...
Abstract CD26 is associated with T cell signal transduction processes as a costimulatory molecule, and CD26+ cells have been suggested to be involved in the pathophysiology of diverse autoimmune diseases. Although cellular molecular mechanisms CD26-mediated activation extensively evaluated by our group others, potential negative feedback regulate still remain elucidated. In present study, we examine expression inhibitory molecules induced via costimulation. We show that coengagement CD3...
Natural products made by marine cyanobacteria are often highly modified peptides and depsipeptides that have the potential to act as inhibitors for proteases. In interests of finding new protease inhibition activity selectivity, grassypeptolide A (1) was screened against a panel proteases found inhibit DPP8 selectively over DPP4. Grassypeptolides were also IL-2 production proliferation in activated T-cells, consistent with putative role immune system. These effects observed Jurkat cells, DPP...
Interleukin (IL)-26, known as a Th17 cytokine, acts on various cell types and has multiple biological functions. Although its precise role still remains to be elucidated, IL-26 is suggested associated with the pathology of diverse chronic inflammatory diseases such psoriasis, bowel rheumatoid arthritis. To develop novel neutralizing anti-human monoclonal antibodies (mAbs) for therapeutic use in clinical setting, we immunized mice human protein. Hybridomas producing anti-IL-26 mAbs were...
Malignant pleural mesothelioma (MPM) is an aggressive neoplasm arising from mesothelial lining of pleura. CD26 molecules preferentially expressed on epithelioid type MPM. This study investigates the molecular mechanisms regulating MPM cells in vitro and vivo.Biochemical cell biological approaches were used for identifying a novel target Its contribution to tumour expansion has been also assessed using animal models. The clinical samples its expression. We identify that cytostatic effects are...
Triple-negative breast cancer (TNBC) has a poor prognosis compared to other subtypes. Although epidermal growth factor receptor (EGFR) is overexpressed in TNBC, clinical trials with EGFR inhibitors including tyrosine kinase (EGFR-TKI) TNBC have heretofore been unsuccessful. To develop effective EGFR-targeted therapy for the precise mechanisms of EGFR-TKI resistance need be elucidated. In this study, understand molecular involved differences efficacy on between human and mouse, we focused...
Abstract The pathogenesis of pulmonary fibrosis involves complex interplay between cell types and signaling pathways. Recurrent alveolar epithelial injury can occur during inflammation, causing dysregulation repair. Dysregulated repair interacts with mesenchymal, inflammatory, endothelial cells to trigger fibroblast‐to‐myofibroblast activation. CD26/dipeptidyl peptidase‐4 (DPP4) is a type II membrane protein mediating pleiotropic effect. However, the mechanistic role CD26/DPP4 in remains...
