The liver possesses a high regenerative capacity. Liver regeneration is compensatory response overcoming disturbances of whole-body homeostasis provoked by organ defects. Here we show that vagus-macrophage-hepatocyte link regulates acute after injury and this system critical for promoting survival. Hepatic Foxm1 rapidly upregulated partial hepatectomy (PHx). branch vagotomy (HV) suppresses upregulation hepatocyte proliferation, thereby increasing mortality. In addition, hepatic FoxM1...

Under insulin-resistant conditions such as obesity, pancreatic β-cells proliferate to prevent blood glucose elevations. A liver-brain-pancreas neuronal relay plays an important role in this process. Here, we show the molecular mechanism underlying compensatory β-cell proliferation. We identify FoxM1 activation islets from relay-stimulated mice. Blockade of relay, including vagotomy, inhibits obesity-induced pathway and suppresses expansion. Inducible β-cell-specific deficiency also blocks In...

The enhancement of insulin secretion and the proliferation pancreatic β cells are promising therapeutic options for diabetes. Signals from vagal nerve regulate both processes, yet effectiveness stimulating is unclear, owing to a lack techniques doing it so selectively prolongedly. Here we report two optogenetic methods vagal-nerve stimulation that led enhanced glucose-stimulated cell in mice expressing choline acetyltransferase-channelrhodopsin 2. One method involves subdiaphragmatic...

We examined the effect of perfluorooctanoic acid (PFOA) on adipose cells using 3T3-L1 adipocytes and found that PFOA increased adipocyte differentiation, triglyceride accumulation, mRNA level factors related to differentiation. In addition, bound peroxisome proliferator-activated receptor γ (PPAR γ). These results suggest promotes differentiation as a PPAR ligand.

Recent progress in direct lineage reprogramming has enabled the generation of induced hepatocyte-like (iHep) cells and revealed their potential as an alternative to hepatocytes for medical applications. However, hepatic functions iHep are insufficient compared with those primary hepatocytes. Here, we show that cell-aggregate formation can rapidly induce growth arrest maturation through activation Hippo signaling. During cell aggregates, Yap inactivation is by actin reorganization...

In clinical trials ANAM, glucose metabolism disorders as adverse effects were relatively frequent, however, when and how they occur remains unclear. Moreover, safety in patients with pancreatic and/or diabetes has not been clarified because most previous studies focused on non-small cell lung had excluded poorly controlled diabetes. Hepatocytes play important roles in the liver, but culture, they immediately lose function and dedifferentiate into progenitor-like cells. Although this unique feature is well-known, dynamics mechanisms of hepatocyte dedifferentiation differentiation potential dedifferentiated hepatocytes (dediHeps) require further investigation. Here, we employ a culture system specifically established for hepatic progenitor cells to study dedifferentiation. We found that with hybrid epithelial/mesenchymal...
