Double glycosylation of cysteine-containing peptides has been carried out by a one-pot two-step sequence comprising selective S-propargylation followed photoinduced (λ max 365 nm) free-radical hydrothiolation with glycosyl thiols. Conditions were established for the sequential introduction two different thiol residues such as and biotinyl derivative.

The NLRP3 inflammasome is a critical component of innate immunity that senses diverse pathogen- and host-derived molecules. However, its aberrant activation has been associated with the pathogenesis multiple diseases, including cancer. In this study, we designed synthesized series aryl sulfonamide derivatives (ASDs) to inhibit inflammasome. Among these, compounds 6c, 7n, 10 specifically inhibited at nanomolar concentrations without affecting NLRC4 AIM2 inflammasomes. Furthermore,...

The synthesis and antiproliferative activity of new benzimidazole derivatives bearing an hydrazone mojety at the 2-position is described. N'-(4-arylidene)-1H-benzo[d]imidazole-2-carbohydrazides were evaluated for their cytostatic toward murine leukemia (L1210), human T-cell (CEM), cervix carcinoma (HeLa) pancreas cells (Mia Paca-2). A preliminary structure-activity relationship could be defined. Some compounds possess encouraging consistent activity, having IC50 values in low micromolar range.

Abstract Late onset Alzheimer disease (LOAD) is traditionally considered as a separate from vascular dementia (VAD). However, growing evidence suggests that β-amyloid (Aβ) accumulation, initiates LOAD-related neurodegeneration, preceded by events. Previous in vitro studies showed β-secretase 1 (BACE1), the key-enzyme of amyloidogenesis, upregulated cerebrovascular insult; moreover, its activity increased both brain and serum LOAD patients. We aimed to investigate whether BACE1 altered also...

Advanced age is associated with severe symptoms and death upon SARS-CoV-2 infection. Virus-specific CD8 + T-cell responses have shown to be protective toward critical COVID-19 manifestations, suggesting that suboptimal cellular immunity may contribute the age-pattern of disease. The induction a response against an emerging pathogen like relies on activation naive T cells. To investigate whether primary this virus defective in advanced age, we used vitro approach prime SARS-CoV-2-specific...

Protein misfolding and amyloid formation are associated with various human diseases including Alzheimer's disease (AD), Parkinson's (PD), Type-2 Diabetes mellitus (T2DM). No disease-modifying therapeutics available for them. Despite the lack of sequence homology between corresponding proteins, aromatic residues recognized as common key motifs in stabilization structures via π–π stacking. Thus, targeting recognition interfaces could be a useful approach inhibiting well disrupting preformed...
