Severe acute respiratory syndrome-coronavirus (SARS-CoV)-2's origin is still controversial. Genomic analyses show SARS-CoV-2 likely to be chimeric, most of its sequence closest bat CoV RaTG13, whereas receptor binding domain (RBD) almost identical that a pangolin CoV. Chimeric viruses can arise via natural recombination or human intervention. The furin cleavage site in the spike protein confers virus ability cross species and tissue barriers, but was previously unseen other SARS-like CoVs....

To gain insight into how researchers of aging perceive the process they study, we conducted a survey among experts in field. While highlighting some common features aging, exposed broad disagreement on foundational issues. What is aging? causes it? When does it begin? constitutes rejuvenation? Not only was there no consensus these and other core questions, but none questions received majority opinion-even regarding need for itself. Despite many believing understand their understanding...

RaTG13, MP789, and RmYN02 are the strains closest to SARS-CoV-2, their existence came light only after start of pandemic. Their genomes have been used support a natural origin SARS-CoV-2 but close examination all them exhibit several issues. We specifically address presence in closely related RacCSxxx claimed PAA/PVA amino acid insertion at S1/S2 junction spike protein same position where PRRA has created polybasic furin cleavage site. show that RmYN02/RacCSxxx instead carry 6-nucleotide...

Pekar et al. (2022) propose that SARS-CoV-2 was a zoonotic spillover first infected humans in the Huanan Seafood Market Wuhan, China. They there were two separate spillovers of closely related lineages A and lineage B short period time. The are differentiated by SNVs; hence, single-SNV A-B intermediate must have occurred an unsampled animal host if two-spillover hypothesis is correct. Consequently, confirmation existence genome from would falsify their spillovers. identified excluded 20...

Abstract Understanding how SARS-CoV-2 entered the human population, thereby causing COVID-19 pandemic, is one of most urgent questions in science today. Two hypotheses are widely acknowledged as being likely to explain pandemic’s origin late 2019: (i) “natural origin” hypothesis that or more cross-species transmissions from animals into humans occurred, at Huanan Seafood Market Wuhan, China; (ii) “laboratory hypothesis, scientific research activities led unintentional leak a laboratory...

Abstract Tyshkovskiy and Panchin have recently published a commentary on our paper in which they outline several “points of disagreement with the Segreto/Deigin hypothesis.” As is titled “The genetic structure SARS‐CoV‐2 does not rule out laboratory origin,” points should provide evidence that rules origin. However, no such instead attempt to criticize arguments highlight aspects could be consistent lab leak hypothesis. Strikingly, Panchin's main point criticism based false premise we...

In this study we document the unexpected discovery of multiple coronaviruses and a BSL-3 pathogen in agricultural cotton rice sequencing datasets. particular, have identified novel HKU5-related Merbecovirus dataset sequenced by Huazhong Agricultural University 2017. We also found an infectious clone sequence containing HKU4-related related to MERS coronavirus early 2020. Another Merbecovirus, as well Japanese encephalitis virus, were 2018. HKU4-related coronaviruses are a group of betacoronaviruses belonging to the same merbecovirus subgenus as Middle Eastern Respiratory Syndrome coronavirus (MERS-CoV), which causes severe respiratory illness in humans with mortality rate over 30%. The high genetic similarity between and MERS-CoV makes them an attractive subject research for modeling potential zoonotic spillover scenarios. In this study, we identify novel contaminating agricultural rice RNA sequencing datasets from Wuhan,...

Metagenomic datasets from pangolin tissue specimens have previously yielded SARS-related coronaviruses which show high homology in their receptor binding domain to SARS-CoV-2, suggesting a potential zoonotic source for this feature of the human virus, possibly via recombination (Liu et al. 2019, Lam 2020, Xiao Liu 2020). Here we re-examine these published datasets. We report that only few samples were found contain coronavirus reads, and even then low abundance, while other non-pangolin...

Abstract HKU4-related coronaviruses (CoVs) are merbecoviruses related to Middle Eastern Respiratory Syndrome coronavirus (MERS-CoV). Questions about the origins of COVID-19 are more than academic interest. For moment, best explanation for seems to be that virus escaped from Wuhan Institute Virology, where scientists were engaged in creating novel SARS-like chimeras and potentially supercharging their transmissibility pathogenicity.
