Genotypic and phenotypic alterations in the bone marrow (BM) microenvironment, particular osteoprogenitor cells, have been shown to support leukemogenesis. However, it is unclear how leukemia cells alter BM microenvironment create a hospitable niche. Here, we report that acute myeloid (AML) but not normal CD34+ or CD33+ induce osteogenic differentiation mesenchymal stromal (MSCs). In addition, AML inhibited adipogenic of MSCs. Mechanistic studies identified AML-derived BMPs activate Smad1/5...

To circumvent donor-to-donor heterogeneity which may lead to inconsistent results after treatment of acute graft-versus-host disease with mesenchymal stromal cells generated from single donors we developed a novel approach by generating these pooled bone marrow mononuclear 8 healthy "3rd-party" donors. Generated were frozen in 209 vials and designated as cell bank. These served source for generation clinical grade end-products, exhibited typical phenotype, trilineage differentiation...

Magnetic resonance imaging of hyperpolarized pyruvate provides a new biomarker for cancer metabolism, based on the dynamic in vivo conversion to lactate. Methods quantification signal evolution need be robust and reproducible across range experimental conditions. Pharmacokinetic analysis spectroscopic data from its metabolites generally assumes that arises ideal rectangular slice excitation profiles. In this study, we examined whether assumption could lead bias kinetic and, if so, such can...

Dynamic contrast enhanced MRI was used to derive perfusion terms which were incorporated into the fitting of hyperpolarized pyruvate lactate apparent conversion rates. Incorporation DCE data reduced fit metabolic rates across set but more so for post therapy resulting in a significant decrease kpl after therapy. These results suggest that effort is needed disentangle from effects when analyzing HP data.
