We have generated 47,932 T-DNA tag lines in japonica rice using activation-tagging vectors that contain tetramerized 35S enhancer sequences. To facilitate use of those lines, we isolated the genomic sequences flanking inserted via inverse polymerase chain reaction. For most performed four sets amplifications two different restriction enzymes toward both directions. In analyzing 41,234 obtained 27,621 sequence tags (FSTs), among which 12,505 were integrated into genic regions and 15,116...

Abstract We analyzed 6,749 lines tagged by the gene trap vector pGA2707. This resulted in isolation of 3,793 genomic sequences flanking T-DNA. Among insertions, 1,846 T-DNAs were integrated into genic regions, and 1,864 located intergenic regions. Frequencies also higher at beginning end coding regions upstream near ATG start codon. The overall GC content insertion sites was close to that measured from entire rice (Oryza sativa) genome. Functional classification these genes showed a...

Abstract Grass cell wall properties influence food, feed, and biofuel feedstock usage efficiency. The glucuronoarabinoxylan of grass walls is esterified with the phenylpropanoid-derived hydroxycinnamic acids ferulic acid (FA) para-coumaric (p-CA). Feruloyl esters undergo oxidative coupling neighboring phenylpropanoids on lignin. Examination rice (Oryza sativa) mutants in a grass-expanded -diverged clade BAHD acyl-coenzyme A-utilizing transferases identified four altered FA or p-CA contents....

RNA editing is the alteration of sequences via insertion, deletion and conversion nucleotides. In flowering plants, specific cytidine residues transcribed from organellar genomes are converted into uridines. Approximately 35 sites present in chloroplasts higher plants; six pentatricopeptide repeat genes involved have been identified Arabidopsis. However, although approximately 500 found mitochondrial RNAs only one gene Arabidopsis has reported to be such editing. Here, we rice mutants that...

Abstract Aldehyde dehydrogenases (ALDHs) catalyze the irreversible oxidation of a wide range reactive aldehydes to their corresponding carboxylic acids. Although proteins have been studied from various organisms and at different growth stages, roles in seed development not well elucidated. We obtained T-DNA insertional mutants OsALDH7, which is remarkably inducible by oxidative abiotic stresses. Interestingly, endosperms osaldh7 null accumulated brown pigments during desiccation storage....

Abstract Brown planthopper (BPH) is a phloem sap-sucking insect pest of rice which causes severe yield loss. We cloned the BPH18 gene from BPH-resistant introgression line derived wild species Oryza australiensis . Map-based cloning and complementation test revealed that encodes CC-NBS-NBS-LRR protein. has two NBS domains, unlike typical NBS-LRR proteins. The promoter:: GUS transgenic plants exhibited strong expression in vascular bundles leaf sheath, especially cells where BPH attacks....

Trithorax group proteins are chromatin-remodeling factors that activate target gene expression by antagonistically functioning against the Polycomb group. In Arabidopsis (Arabidopsis thaliana), protein1 (ATX1) regulates flowering time and floral organ identity. Here, we observed suppression of Oryza sativa Trithorax1 (OsTrx1), an ortholog ATX1, delayed in rice (Oryza sativa). Because delay occurred only under long-day conditions, evaluated signal pathways specifically function conditions....

Flowering is exquisitely regulated by both promotive and inhibitory factors. Molecular genetic studies with Arabidopsis have verified several epigenetic repressors that regulate flowering time. However, the roles of chromatin remodeling factors in developmental processes not been well explored Oryza sativa (rice). We identified a factor OsVIL2 (O. VIN3-LIKE 2) promotes flowering. contains plant homeodomain (PHD) finger, which conserved motif histone binding proteins. Insertion mutations...

Although susceptibility to seed shattering causes severe yield loss during cereal crop harvest, it is an adaptive trait for dispersal in wild plants. We previously identified a recessive locus, sh-h, from the rice mutant line Hsh that carries enhanced abscission layer. Here, we further mapped sh-h 34-kb region on chromosome 7 by analyzing 240 F(2) plants and five F(3) lines cross between Blue&Gundil. had point mutation at 3' splice site of seventh intron within LOC_Os07g10690, causing 15-bp...
