Statistical analyses of protein families reveal networks coevolving amino acids that functionally link distantly positioned functional surfaces. Such linkages suggest a concept for engineering allosteric control into proteins: The intramolecular two proteins could be joined across their surface sites such the activity one might other. We tested this idea by creating PAS-DHFR, designed chimeric connects light-sensing signaling domain from plant member Per/Arnt/Sim (PAS) family with...

In the pivotal phase II/III trial of idursulfase administered intravenously to treat mucopolysaccharidosis II, approximately half patients developed antibodies idursulfase. This post-hoc analysis data from and extension study examined relationship between antibody status outcomes. A total 63 treatment-naïve received 0.5 mg/kg intravenous weekly for two years. Thirty-two (51%) were positive anti-idursulfase IgG antibodies, 23 whom (37%) became persistently positive. All who an response did so...

Abstract The number of available therapies for rare diseases remains low, as fewer than 6% have an approved treatment option. International Rare Diseases Research Consortium (IRDiRC) set up the multi-stakeholder Data Mining and Repurposing (DMR) Task Force to examine potential applying biomedical data mining strategies identify new opportunities use existing pharmaceutical compounds in ways accelerate pace drug development disease patients. In reviewing past successes repurposing, planning...

Neurons in the caudal pressor area (CPA) are a source of tonic sympathoexcitation that is dependent on activation cardiovascular sympathetic premotor neurons rostral ventrolateral medulla (RVLM). In present study, we sought to clarify mechanism through which CPA elicit increases RVLM neuronal discharge, vasoconstrictor tone, and arterial pressure. urethan-chloralose-anesthetized, paralyzed, artificially ventilated rats, bilateral disinhibition with bicuculline (Bic) after (CVLM) caused...

Mucopolysaccharidosis II (MPS II) is a rare, X-linked lysosomal storage disease caused by pathogenic variants of the iduronate-2-sulfatase gene (IDS) and characterized highly variable spectrum. MPS severity difficult to predict based on IDS alone; while some genotypes are associated with specific phenotypes, course most remains unknown. This study aims refine genotype-phenotype categorization combining information from scientific literature data two clinical studies in II.Genotype,...
