Abstract Ab-mediated rejection (AMR) of solid organ transplants is characterized by intragraft macrophages. It incompletely understood how donor-specific Ab binding to graft endothelium promotes monocyte adhesion, and what, if any, contribution made the Fc region Ab. We investigated mechanisms underlying recruitment HLA class I (HLA I) Ab–activated endothelium. used a panel murine mAbs different subclasses crosslink on human aortic, venous, microvascular endothelial cells measured monocytic...

Abstract Endothelial cells (EC) coordinate vascular homeostasis and inflammation. In organ transplantation, EC are a direct alloimmune target. We posited that tissue specific heterogeneity of may partly underlie the disparate organ-specific risk. examined endothelial response to inflammation across six primary beds from four major transplanted organs: heart, lung, kidney liver. First, we reanalyzed public dataset cardiac allograft rejection found inflammatory genes were elevated in human...

Correlation between antibody-mediated rejection (ABMR) and circulating HLA donor-specific antibodies (HLA-DSA) is strong but imperfect in kidney transplant (KT) recipients, raising the possibility of undetected HLA-DSA or non-HLA contributing to ABMR. Detailed evaluation degree matching together with identification KT may help decipher antibody involved.

Abstract Transplant recipients developing donor-specific HLA class II (HLA-II) Abs are at higher risk for Ab-mediated rejection (AMR) and transplant vasculopathy. To understand how HLA-II cause AMR vasculopathy, we determined the signaling events triggered in vascular endothelial cells (EC) following Ab ligation of molecules. expression EC was induced by adenoviral vector CIITA or pretreatment with TNF-α/IFN-γ. stimulated proliferation migration. Class also activation key nodes Src, focal...

It is currently unclear which donor specific HLA antibodies confer the highest risk of antibody-mediated rejection (AMR) and allograft loss. In this study, we hypothesized that two distinct features (HLA IgG subclass Fcγ receptor [FcγR] polymorphisms) vary from patient to patient, influence process monocyte trafficking macrophage accumulation in during AMR an interrelated fashion. Here, investigated contribution human FcγR polymorphisms recruitment vitro by primary aortic endothelium...

Unambiguous HLA typing is important in hematopoietic stem cell transplantation (HSCT), disease association studies, and solid organ transplantation. However, current molecular methods only interrogate the antigen recognition site (ARS) of genes, resulting many cis-trans ambiguities that require additional to resolve. Here we report high-resolution 10,063 National Marrow Donor Program (NMDP) registry donors using long-range PCR by next generation sequencing (NGS) approach on buccal swab...

Background Vascular endothelial cells (EC) are critical for regulation of local immune responses, through coordination leukocyte recruitment from the blood and egress into tissue. Growing evidence supports an additional role endothelium in activation costimulation adaptive cells. However, this function remains somewhat controversial, full repertoire durability enhanced costimulatory phenotype has not been wholly defined. Methods Human was stimulated with continuous TNFα or IFNγ 1-48hr;...

The "virtual" crossmatch (VXM) has become a critical tool to predict the compatibility between an organ donor and potential recipient. Yet, nonstandardized laboratory practice can lead variability in VXM interpretation. Therefore, UCLA's Exchange survey was designed understand factors that influence of prediction presence HLA donor-specific antibody (DSA). Thirty-six blood samples 72 reference sera were sent 35 participating laboratories perform testing, flow (FXM), from 2014 2019,...
